Abstract:To investigate the distribution and histochemical characteristics of mast cells in major lymphatic organs of rabbits. Rabbits' spleen and sacculus rotundus tissues were fixed with Carnoy's fluid or neutral buffered formalin (NBF) stained by routine toluidine blue or long time toluidine blue (LTB) and then observed under microscopy. Mast cells of sacculus rotundus were mainly distributed in mucosa epithelium or submucosa connective tissues, and those of spleen were located in lymphocytes of red pulp. No mast cell was found in the lymph follicle of thymus independent region in sacculus rotundus or spleen. Staining result of MCs fixed with Carnoy's fluid was excellent, and staining was enhanced by LTB staining. Mast cells of sacculus rotundus and spleen are thymus dependent or T cell dependent, and have close ties with the location of T lymphocytes. The amount of mast cells is varioius with different fixtations and staining times.
