Observation of SOD Activity in Liver of Acute Nickel CarbonylPoisoned Rats and Its Gene Expression

Abstract:Objective To investigate the SOD activity and the mRNA expression of Cu-Zn SOD in liver tissue of acute nickel carbonyl poisoned rats in order to probe into the acute toxicity and mechanism of carbonyl nickel.Methods Healthy SD rats were exposed by statically inhaling to nickel carbonyl of 20,135,and 250 mg/m3 respectively for 30 min.In same way,SD rats were exposed to 250 mg/m3 chlorine for 30 min.as the exposed control group.Healthy SD rats were taken as the normal control group.Liver tissues of animals were taken at the 1st day,2nd day,3rd day,and 7th day respectively after exposure.The SOD activities of each group were studied using xanthine oxidase assay.Meanwhile,the mRNA expressions of Cu-Zn SOD gene of liver were studied by RT-PCR.Results Comparing with the normal control group,SOD activities of liver tissues significantly declined at the 3rd day after nickel carbonyl exposure(P<0.05)and decreased in all nickel carbonyl exposed groups(P<0.05).In exposed control group,the SOD activities significantly declined(P<0.01).Comparing with the normal control group,the mRNA expression of Cu-Zn SOD gene in rat liver tissues acutely exposed to three doses of nickel carbonyl significantly declined at the 3rd day(P<0.05).Conclusions The carbonyl nickel and chlorine could cause liver tissue oxidative damage and reduce SOD activity.The most obvious reduction takes place in the 3rd and 7th day showing noticeable time-effect relationship in liver tissue(P<0.05).
