Abstract:Anterior forebrain pathway (AFP), a basal ganglia-dorsal forebrain circuit, significantly impacts birdsong, specifically in juvenile or deaf birds. Despite many physiological experiments supporting AFP's role in song production, the mechanism underlying it remains poorly understood. Using a computational model of the anterior forebrain pathway and song premotor pathway, we examined the dynamic process and exact role of AFP during song learning and distorted auditory feedback (DAF). Our simulation suggests that AFP can adjust the premotor pathway structure and syllables based on its delayed input to the robust nucleus of the archistriatum (RA). It is also indicated that the adjustment to the synaptic conductance in the song premotor pathway has two phases: normal phases where the adjustment decreases with an increasing number of trials and abnormal phases where the adjustment remains stable or even increases. These two phases alternate and impel a specific effect on birdsong based on AFP's specific structures, which may be associated with auditory feedback. Furthermore, our model captured some characteristics shown in birdsong experiments, such as similarities in pitch, intensity, and duration to real birds and the highly abnormal features of syllables during DAF.

Abstract:The anterior forebrain pathway (AFP) of the song system in songbirds plays an important role in vocal learning in songbirds. The lateral magnocellular nucleus of the anterior neostriatum (LMAN) is the last output nucleus of AFP. AFP transmits the signals to the robust nucleus of the arcopallium (RA) by LMAN. LMAN and the high vocal centre (HVC) regulate the activity of RA and they make an impact on the vocal behavior. LMAN is likely to affect vocal plasticity via its monosynaptic connections with RA, a vocal motor area critical to song production throughout life. In this paper, the progress of LMAN and the plasticity of vocal learning in songbirds are reviewed in recent years.

Abstract:Like human speech, singing of songbirds is sensorimotor learning by tutor and auditory feedback. The development and crystallization of singing is depended on the integrity of the song system composed of vocal pathway and anterior forebrain pathway. X area in forebrain pathway plays an important role during learning and memory of singing. In this paper, we reviewed studies on the organization and the function in singing development and crystallization, the synaptic plasticity of X area. We also compared learning and memory function of X area to that of mammalian basal ganglia.

Abstract:Abstract Mistakes in performance feel disappointing, suggesting that brain pathways for aversive feedback may play a role in motor learning. Here we tested if the lateral habenula (LHb), an evolutionarily conserved part of the limbic system known in mammals to relay aversive feedback from ventral pallidum (VP) to ventral tegmental area (VTA) dopamine neurons, is involved in birdsong learning and production. By combining viral tract tracing and functional circuit mapping, we discovered that songbird LHb links VP and an auditory cortical area to singing-related DA neurons that signal song errors. As in mammals, VP stimulation activated LHb activity and LHb stimulation suppressed DA firing. To test this pathway’s role in learning we lesioned the LHb in juvenile zebra finches and recorded their songs in adulthood. Birds with the LHb lesioned as juveniles produced highly unusual vocalizations as adults, including prolonged high-pitch notes and species-atypical trills. These findings identify a songbird VP-LHb-VTA pathway with similar functional connectivity as mammals, expand the known territories of vocal learning circuits, and demonstrate that limbic circuits associated with disappointing outcomes are important for motor performance learning.

Abstract:Our previous work has shown that AFP could adjust synaptic conductance of RA network with its firing pattern changed from bursting to tonic firing, eventually leading to song degradation. However, it is difficult to explore the dynamic of RA network due to its high dimensionality and complexity. To address this issue, we use the mean field theory proposed by Wilten Nicola to reduce RA network to a system of switching ordinary differential equations. Numerical simulations prove that the mean field model could qualitatively and quantitatively describe the behavior of nucleus RA. As a result, we perform the non-smooth bifurcation analysis over the mean field model to understand the bifurcations that RA network displays. Then we determine that there is a subcritical-Andronov-Hopf bifurcation at a particular applied current, which causes the emergence of the bistability with current increased. The bistability consists of a stable node and a stable limit cycle, corresponding to tonic firing and bursting of the full network, respectively. With these properties, we can explain the dynamical principles of AFP’s adjustment on RA during song degradation. Furthermore, our results would effectively promote the combination of dynamical analysis and neural network modeling based on the mean field theory.

Abstract:Learning vocal behaviors, like speech and birdsong, is thought to rely on continued performance evaluation. Whether candidate performance evaluation circuits in the brain are sufficient to guide vocal learning is not known. Here, we test the sufficiency of VTA projections to the vocal basal ganglia in singing zebra finches, a songbird species that learns to produce a complex and stereotyped multi-syllabic courtship song during development. We optogenetically manipulate VTA axon terminals in singing birds contingent on how the pitch of an individual song syllable is naturally performed. We find that optical inhibition and excitation of VTA terminals are each sufficient to reliably guide learned changes in song. Inhibition and excitation have opponent effects on future performances of targeted song syllables, consistent with positive and negative reinforcement of performance outcomes. These findings define a central role for reinforcement mechanisms in learning vocalizations and demonstrate minimal circuit elements for learning vocal behaviors.

Abstract:Birdsong and human languages are both acquired by learning .During sensory period , templates are formed in the brain after juveniles heard tutor songs and stored in the related areas and in the motor-learning phase , juveniles compare their own songs to the templates and modify them to match the templates in their memories . When the songs are crystallized in the adult stage , auditory feedback is required to maintain the features and stabili-ty of their own songs.Hence, the sensory-motor integration of auditory and motor systems is vitally important to the formation of templates and the maintenance of crystallized songs .In the recent years , it is found that NIf-HVC syn-apses are related with the formation of templates memory and the maintenance of spatial features of plastic songs in the juveniles while this projection is crucial for the grammar structure of crystallized songs in the Bengalese finches . In songbirds , HVC if homologous to human Broca's Area and NIf has many similarities with Area Spt in the human speech processing circuit .Therefore the research on the function of NIf-HVC synapses in the speech circuit is not only helpful for the study of Area Broca and Spt of human but also advantageous for exploring the mechanism con -cerning sense and motion information integration involved in vocal learning .

Abstract:Learned vocalizations (long call and song) of adult male songbirds start from the high vocal center (HVC), and are integrated and output by the robust nucleus of the arcopallium (RA), which connects synaptic relationships with the dorsomedial nucleus of the intercollicular complex (DM). To determine the effect on learned vocalization of the unilateral forebrain and midbrain in adult male zebra finch, electrolytic lesions and acoustic analysis technology were used. The results indicated that RA and DM nuclei are involved in the control of learned vocalization, and the right side is dominant in the forebrain and midbrain.

Abstract:Abstract High dimensionality and complexity are the main difficulties of the study over network dynamics. Recently, Wilten Nicola proposed the mean field theory to research the bifurcations that the full networks display. Here, we use his approach on the birdsong neural network. Our previous work has shown that AFP could adjust the synapse conductance of nucleus RA and change RA’s firing patterns, eventually leading to song degradation. To understand the dynamical principle behind this, we use a technique to reduce the RA network to a mean field model, in the form of a system of switching ordinary differential equations. Numerical results have shown that the mean field equations can qualitatively and quantitatively describe the behavior of nucleus RA. Based on the non-smooth bifurcation analysis of the mean field model, we determine that there is a subcritical-Andronov-Hopf bifurcation at a particular stimulation, which can explain the role of AFP during song degradation. The results indicate that we can see AFP’s adjustment in RA synapse conductance as the adjustment of initial value in the bistable system. More precisely, during song degradation, the mean field system would transform to a stable node (corresponding to distorted songs) rather than a stable limit cycle (corresponding to normal songs).

Abstract:Songbirds'singing is similar to human language,which is a complex vocal learning behavior and isregulated by a group of interconnected nerve nuclei in the brain.These song control nuclei have a certain degreeof structural homology with the brain regions related to human vocal control,and may share some basicregulation mechanisms of vocal learning.Therefore,songbirds have become an important model animal to studythe neural mechanism of vocal learning,which can not only shed light on avian language learning,but alsoprovide an important reference for understanding the neural process of human language learning and the treatmentof language disorders.Willams Thorpe from University of Cambridge took the lead in bird singing research in1948.Subsequently,many researchers in the United States used a variety of experimental techniques to carry outthe study of birdsong neurobiology.In the1960s,Chinese scholar Professor LAN Shu-Cheng first carried out theresearch on bird singing neurobiology in the Northeast Normal University,and then the researchers in BeijingNormal University,South China Normal University,Hainan Normal University and Jiangxi Science andTechnology Normal University carried out the researches on the neural mechanism of bird singing usingmolecular biology and electrophysiological technology.In recent years,the application of songbird model ininterdisciplinary research of vocal behavior and neurobiology mainly focuses on the mechanism of vocalplasticity;the neurotransmitters,sex hormones and other bioactive substances regulated the song behavior;theeffect of auditory feedback on song behavior and application of photogenetic technology and genomics in thisfield.In the future,frontier technologies such as single cell sequencing and CRISPR/Cas9gene editing havepushed these research fields to a deeper level.In addition,the effects of neurotransmitters on the neural networkregulation of songbirds'song control nuclei and song behavior need to be further studied.At the same time,theapplication of some new fluorescent probes will help to further reveal the regulatory role of neurotransmitters inthe singing process of songbirds.The neurobiological study of songbirds'song behavior will bring importantenlightenment and theoretical support to the exploration of the pathogenesis and treatment of human languagedisorder related diseases.In conclusion,this paper systematically summarizes the research history,importantfindings and research progress of songbird song behavior,as well as the important enlightenment for the treatmentof related central nervous system diseases

Abstract:Songbird is one of the few animals with complex phonological learning ability except human. The song of a songbird is similar to the production of human language. Using songbirds as models, the comparative physiology study provides important insights into the neural mechanisms that regulate language sequences and language learning and production in humans. The singing of songbirds is regulated by the interrelated singing control nuclei. Dopamine, an important neurotransmitter in the brain, is involved in the regulation of movement, cognition, motivation, reward, addiction and learning in mammals, it has been linked to Parkinson's disease, schizophrenia and Huntington's syndrome in humans. DA is a key substance in the brain that regulates learning and motivation. In songbirds, dopamine and its receptors are found in a large number of singing-related nuclei. In addition, recent studies have shown that dopamine promotes songbird song learning in juvenile stage, song retention in adulthood, and song production in courtship by regulating the singing-related nuclei. Based on the research of our work , this paper reviews the research progress in the regulation of the dopamine system on the singing-related nuclei and singing behavior of songbirds, the potential mechanism of dopamine signal regulating singing behavior of songbirds was proposed.
A songbird forebrain area potentially involved in auditory discrimination and memory formation

Abstract:Songbirds rely on auditory processing of natural communication signals for a number of social behaviors, including mate selection, individual recognition and the rare behavior of vocal learning — the ability to learn vocalizations through imitation of an adult model, rather than by instinct. Like mammals, songbirds possess a set of interconnected ascending and descending auditory brain pathways that process acoustic information and that are presumably involved in the perceptual processing of vocal communication signals. Most auditory areas studied to date are located in the caudomedial forebrain of the songbird and include the thalamo-recipient field L (subfields L1, L2 and L3), the caudomedial and caudolateral mesopallium (CMM and CLM, respectively) and the caudomedial nidopallium (NCM). This review focuses on NCM, an auditory area previously proposed to be analogous to parts of the primary auditory cortex in mammals. Stimulation of songbirds with auditory stimuli drives vigorous electrophysiological responses and the expression of several activity-regulated genes in NCM. Interestingly, NCM neurons are tuned to species-specific songs and undergo some forms of experience-dependent plasticity in-vivo. These activity-dependent changes may underlie long-term modifications in the functional performance of NCM and constitute a potential neural substrate for auditory discrimination. We end this review by discussing evidence that suggests that NCM may be a site of auditory memory formation and/or storage.

Abstract:Disruption of the transcription factor FoxP2, which is enriched in the basal ganglia, impairs vocal development in humans and songbirds. The basal ganglia are important for the selection and sequencing of motor actions, but the circuit mechanisms governing accurate sequencing of learned vocalizations are unknown. Here, we show that expression of FoxP2 in the basal ganglia is vital for the fluent initiation and termination of birdsong, as well as the maintenance of song syllable sequencing in adulthood. Knockdown of FoxP2 imbalances dopamine receptor expression across striatal direct-like and indirect-like pathways, suggesting a role of dopaminergic signaling in regulating vocal motor sequencing. Confirming this prediction, we show that phasic dopamine activation, and not inhibition, during singing drives repetition of song syllables, thus also impairing fluent initiation and termination of birdsong. These findings demonstrate discrete circuit origins for the dysfluent repetition of vocal elements in songbirds, with implications for speech disorders. Disruption of FOXP2 cause Childhood Apraxia of Speech, a speech disorder marked by difficulties in accurately sequencing vocal motor actions. The authors show that disruption of FoxP2 in the adult songbird similarly disrupts birdsong and link dopaminergic signalling to disruptions in song production.
What is the Role of Thalamostriatal Circuits in Learning Vocal Sequences?

Abstract:Basal ganglia (BG) circuits integrate sensory and motor-related information from the cortex, thalamus, and midbrain to guide learning and production of motor sequences. Birdsong, like speech, is comprised of precisely sequenced vocal elements. Learning song sequences during development relies on Area X, a vocalization related region in the medial striatum of the songbird BG. Area X receives inputs from cortical-like pallial song circuits and midbrain dopaminergic circuits and sends projections to the thalamus. It has recently been shown that thalamic circuits also send substantial projections back to Area X. Here, we outline a gated-reinforcement learning model for how Area X may use signals conveyed by thalamostriatal inputs to direct song learning. Integrating conceptual advances from recent mammalian and songbird literature, we hypothesize that thalamostriatal pathways convey signals linked to song syllable onsets and offsets and influence striatal circuit plasticity via regulation of cholinergic interneurons (ChIs). We suggest that syllable sequence associated vocal-motor information from the thalamus drive precisely timed pauses in ChIs activity in Area X. When integrated with concurrent corticostriatal and dopaminergic input, this circuit helps regulate plasticity on medium spiny neurons (MSNs) and the learning of syllable sequences. We discuss new approaches that can be applied to test core ideas of this model and how associated insights may provide a framework for understanding the function of BG circuits in learning motor sequences.

Abstract:Speech is a complex sensorimotor skill, and vocal learning involves both the basal ganglia and the cerebellum. These subcortical structures interact indirectly through their respective loops with thalamo-cortical and brainstem networks, and directly via subcortical pathways, but the role of their interaction during sensorimotor learning remains undetermined. While songbirds and their song-dedicated basal ganglia-thalamo-cortical circuitry offer a unique opportunity to study subcortical circuits involved in vocal learning, the cerebellar contribution to avian song learning remains unknown. We demonstrate that the cerebellum provides a strong input to the song-related basal ganglia nucleus in zebra finches. Cerebellar signals are transmitted to the basal ganglia via a disynaptic connection through the thalamus and then conveyed to their cortical target and to the premotor nucleus controlling song production. Finally, cerebellar lesions impair juvenile song learning, opening new opportunities to investigate how subcortical interactions between the cerebellum and basal ganglia contribute to sensorimotor learning.
