Abstract:A versatile synthetic route to prepare all four stereoisomeric 4-fluoro-glutamines was developed by exploiting a Passerini three-component reaction. The skeleton of 4-substituted glutamine derivatives was efficiently constructed. Subsequent four-step reactions, highlighted by a "neutralized" TASF fluorination, provided the desired products with high yields and excellent optical purity. The optically pure fluorine-18 labeled 4-fluoroglutamines were also successfully prepared using either a 18-crown-6/KHCO(3) or K[222]/K(2)CO(3) catalysis system. Preliminary cell uptake and inhibition studies using the 9L tumor cells and SF188(Bcl-xL) tumor cells (a glutamine addicted tumor derived from glioblastoma) provided strong evidence for their potential application in conjunction with positron emission tomography (PET) for in vivo imaging of tumors, which use glutamine as an alternative energy source.

Abstract:We report the preparation of a novel glutamine derivative, (2S,4S)-2,5-diamino-4-(4-(2-fluoroethoxy)benzyl)-5-oxopentanoic acid, (2S, 4S)4-[18F]FEBGln ([18F]4), through efficient organic and radiosyntheses. In vitro assays of [18F]4 using MCF-7 cells showed that it entered cells via multiple amino acid transporter systems including system L and ASC2 transporters but not through the system A transporter. [18F]4 showed promising properties for tumor imaging and may serve as a lead compound for further optimizing and targeting the system L transporter associated with enhanced glutamine metabolism in cancer cells.

Abstract:A novel synthesis method of dipeptides containing glutamine is reported. Protected L-amino acids were prepared by using inexpensive phthaloyl as the protecting group. Then the phthaloyl-L-amino acids were condensed with glutamine salts by the mixed anhydride method to afford phthaloyl dipeptides. Subsequently, the phthaloyl was removed from the dipeptides with hydrazine hydrate. As a result, optically pure glutamine-containing dipeptides were obtained in good yields.

Abstract:Aim To design and synthesize new L-iso-glutamine derivatives and to evaluate their aminopeptidase N inhibitory activities in vitro.Methods The target compounds were synthesized starting from the acylation of 3,4-dichlorobenzoic acid with L-glutamic acid to form the basic scaffold,followed by the cyclization in acetic anhydride and ammonolysis,and their aminopeptidase N inhibitory activities are assayed in vitro.Results and conclusion Fifteen novel L-iso-glutamine derivatives were synthesized,and their structures were confirmed by1H-NMR spectra,MS and IR.Compound Ⅱ4 and Ⅱ6 exhibit potent aminopeptidase N inhibitory activities(IC50=20-40 μmol·L-1) and are worthy of further investigation.

Abstract:A new practical approach to prepare diethyl N-{4-[N-methyl-N-(3-(2,4-dioxo-1,2,3,4-tetrahydropyrimidin-5-yl)amino-2-hydroxypropyl)amino]benzoyl}-L-glutamate and its derivatives has been realized from ethyl 4-aminobenzoate (4a) and diethyl N-(4-aminobenzoyl)-L-glutamate (4b). Six novel 5-(substituted)amino pyrimidines 9 similar to 11 were synthesized via methylation, allylation, bromohydroxylation, epoxidation, ring-opening, deprotection reaction and so on, and were characterized by (1)H NMR, (13)C NMR and MS techniques. The inhibitory activities against human dihydrofolate reductase of the compounds have been determined. The results showed that the bioactivities of the compounds were related with L-glutamate side chain.

Abstract:Using cheap L-glutamic acid as raw material, phthalic anhydride was employed to protect the α-amino group of L-glutamic acid, and acetic anhydride was used to form the glutamic anhydride. Then, the glutamic anhydride was attacked by isopropylamine to open the ring. Finally, hydrazine hydrate was used to deprotect the phthaloyl, and isopropyl-L-glutamine was obtained. The structure of product in each step was confirmed by 1H-NMR, FT-IR and LC-MS. The synthesis was optimized under different reaction temperature, time and reactant ratios.

Abstract:A series of Mannich base type derivatives of Glaucocalyxin A (GLA) were designed and prepared. The cytotoxicity of these compounds was evaluated against six tumor cell lines (SMMC-7721, B16, SGC-7901, A549, KB, HL-60). Most compounds exhibited potent antiproliferative effects with low micromolar IC50 values. Compound 1 with para methyl benzyl amine moiety and compound 16 with cyclohexylamine moiety displayed the highest inhibition efficacy. Significantly, the cytotoxicity of compound 1 was much lower than GLA against the normal human liver cell (HL-7702). The in vitro stability assay revealed that transformation of GLA to Mannich base type derivatives improved the compound stability in rat plasma. Finally, decomposition product analysis supported that compound 1 could act as a prodrug and release GLA in the intracellular environment.

Abstract:OBJECTIVETo establish a new approach to synthesis of diethyl N-[4-[(2,4-diaminopyrido[3,2-d]pyrimidin-6-yl)methylamino]benzoyl]-L-glutamate.METHODSTarget compound (5) was synthesized by the use of (2,4-dioxo-tetrahydropyridopyrimidin-6-yl)methyl acetate (1) as starting material via hydrolysis, chlorination, condensation with diethyl (p-aminobenzoyl)glutamate and aminolysis.RESULTSA new approach to synthesis of diethyl N-[4-[(2,4-diaminopyrido[3,2-d]pyrimidin-6-yl)methylamino]benzoyl]-L-glutamate was established. This synthetic route has hydrolysis reaction, chlorination, diethyl N-(p-aminobenzoyl)-L-glutamate condensation reaction and ammonolysis reaction. The total yield is 36.7%.The structures of those compounds have identified by 1H nuclear magnetic resonance, 13C nuclear magnetic resonance and mass spectrometry. This synthetic route avoid the unstable brominated reaction product and improves the harsh condition of ammonolysis reaction.CONCLUSIONThe new synthetic route has improved the reaction condition and the stability of the intermediate, and increased the extent of the derivative compounds, which has great significance to anti-folic acid of anti-tumor inhibitor synthesis.

Abstract:Tumor cells often exhibit metabolic reprogramming to maintain their rapid growth and proliferation. Glutaminase 1 (GLS1) has been viewed as a promising target in the glutamine metabolism pathway for the treatment of malignant tumors. Using structure-based drug design approaches, a novel series of GLS1 allosteric inhibitors were designed and synthesized. Compound 41a (LWG-301) with an alkane chain "tail" group had potent biochemical and cellular GLS1 activity, and improved metabolic stability. LWG-301 exhibited moderate antitumor effects in HCT116 xenograft model, with TGI of 38.9% in vivo. Mechanistically, LWG-301 could significantly block glutamine metabolism, resulting in changes in the corresponding amino acid levels in cells, induce a concentration-dependent increase in intracellular ROS levels, and induce apoptosis. Taken together, this paper provides more structural references and new design strategy for the development of GLS1 allosteric inhibitors.

Abstract:OBJECTIVE To synthesize a novel series of 3- amidebenzamide derivatives and to study their anti- tumor activity in vitro. METHODS The target compounds were synthesized from 5- amino- 2- methylbenzoic acid and 5- amino- 2- chlorobenzoic acid by protection of 5- amino using Boc,formation of amide,deprotection and acylation,etc. The anti- tumor activity in vitro of these compounds was studied by MTT assay on prostate carcinoma cells PC- 3,ovarian carcinoma cells SKOV- 3,human colorectal cancer cells HCT116,human hepatocellular carcinoma cells Hep- G2,malignant melanoma cells A375,human lung carcinoma cells A549 and human erythroleukemia cells K562. RESULTS and CONCLUSION Nine new compounds were prepared,and the chemical structures were confirmed by1HNMR and MS. Preliminary anti- tumor activity in vitro displays that compound Ⅳh and Ⅳi exhibited good activity against chronic myelogenous leukemia.

Abstract:The invention discloses a method for preparing an N-(2)-L-alanyl-L-glutamine compound. The N-(2)-L-alanyl-L-glutamine compound is prepared by adopting a particle process crystal product molecule assembly and form optimization technique. The compound has the characteristics of high purity, high yield, low hygroscopicity and good stability. Meanwhile, the invention also discloses a pharmaceutical composition prepared from the alanyl glutamine prepared by the method. The pharmaceutical composition is prepared through the steps of preparing the alanyl glutamine into a 10-30% aqueous solution, adsorbing the aqueous solution with medicinal activated charcoal according to the ratio (w/v) of 0.1-0.5%, then, carrying out big-dish freeze-drying, and carrying out aseptic packaging. The whole preparation process is simple in operation and does not need any excipient, and the product has better stability compared with the products in the past.

Abstract:In order to look for novel antitumor drugs and virucides with high activity and low toxicity, a series of amino acid linked phosphoryl nitrogen mustard derivatives have been synthesized. The structures of all compounds prepared were confirmed by H-1 NMR, P-31 NMR, IR MS spectroscopy, and elemental analyses. IN the case of dl-alpha-amino acid derivatives, the presence of multiple chiral centers in the target products leads to the generation of diastereoisomers, which can be detected by spectroscopic and analytical (TLC) methods. Preliminary bioassays indicate that some of compounds 5 have high inhibitory activities against some tumor cells and tobacco mosaic virus (TMV). In addition, some of them display certain herbicidal activities and significant fungicidal activities against wheat leaf rust.
Antitumor Activities of O-Sulfonated Derivatives of (1 -> 3)-Alpha-d-glucan from Different Lentinus Edodes

Abstract:Four water-insoluble (1 -> 3)-alpha-D-glucans, coded L-II1, L-II2, L-II3 and L-II4, with different molecular weights were isolated from four kinds of fruiting bodies of Lentinus Edodes. The four alpha-D-glucans were O-sulfonated to obtain derivatives (SL-II) having degrees of substitution (DS) from 0.9 to 2.1 respectively. The structure of the samples was analyzed by infrared spectra, elemental analysis, and C-13 NMR. The weight-average molecular weight (M-w), radii of gyration (s(2)>(1/2)(z)) and intrinsic viscosity ([eta]) of the native alpha-D-glucans and O-sulfonated derivatives were measured by size-exclusion chromatography combined with laser light scattering (SEC-LLS), LLS, and viscometry in 0.2M aqueous NaCl and in dimethyl sulfoxide (DMSO) containing 0.25M LiCl at 25 degrees C respectively. The M-w values of the O-sulfonated derivatives were much lower than those of the native alpha-D-glucans. The experimental results indicate that the O-sulfonated derivatives are water-soluble and exist as an expanded flexible chain in aqueous solution owing to intramolecular hydrogen bonding or interaction between charge groups. The in vivo and in vitro antitumor activities of the native alpha-D-glucans and their O-sulfonated derivatives against solid tumor Sarcoma 180 cells were evaluated and compared. Interestingly, all of the O-sulfonated derivatives exhibited higher antitumor activities than those of the native glucans. The results reveal that the effect of O-sulfonation of the alpha-D-glucan on the improvement of their antitumor activities was considerable.

Abstract:A series of isoquinolonic acid derivatives(4a–4o) was synthesized via one-pot synthesis for their anti-tumor activity. The structures of all the targeted compounds were confirmed by 1H nuclear magnetic resonance (1H NMR) spectrometry and mass spectrometry(MS). The anti-tumor activities of compounds 4a–4o against MG63(human osteosarcoma cells) and B16-F10(mouse melanoma cells) were examined. To evaluate the antitumor effect of the as-synthe sized compounds, we compared the half maximal inhibitory concentration(IC50) of compounds 4a–4o to that of camptothecin(CPT) which appeared to be active against a broad range of human cancers. Among all the compounds, compound 4l shows the most potent biological activity against MG63 cells[IC50=(2.16±0.26) μmol/L]_and B16-F10 cells[IC50=(6.95±0.24) μmol/L], thus providing useful information for the antitumor activity and potential practical use of isoquinolonic acid compounds. In addition, we screened out an efficient compound(4l) that shows potential inhibit activity against Topoisomerase I(Topo I) by docking simulation.

Abstract:In order to discover novel high efficiency and low toxic anticancer drugs, a series of novel 2-amino-5-ethylpyrimidine derivatives (XIIa–x) were designed and synthesized. Their antiproliferative activities against four human cancer cells were evaluated by MTT assay. Among the synthetic target compounds, the compound 5-ethyl-6-((4-methoxybenzyl)thio)-N4-(3,4,5-trimethoxyphenyl)pyrimidine-2,4-diamine (XIIw) showed the most potent antiproliferative activity against MGC-803 (human gastric cancer cells), with the IC50 values of 1.02 ± 0.16 μM. Meanwhile, this compound inhibited colony formation of MGC-803 cells. Furthermore, this compound blocked the cell cycle in S phase and induced cell apoptosis. Collectively, our findings indicated that this compound was a valuable lead compound to design new anticancer drugs.

Abstract:Glutaminase 1 (GLS1) is overexpressed in multiple types of malignant tumors and is viewed as a promising target in cancer therapy. Thus, the discovery for small-molecule GLS1 inhibitors is being urgent. Based on our previous study of C147, a potent GLS1 allosteric inhibitor yet with a limited metabolic stability, a structure-based optimization was carried out, with a series of novel GLS1 allosteric inhibitors rationally designed, synthesized and biologically evaluated. Such endeavor has culminated in the identification of 41e, a promising GLS1 allosteric inhibitor with 4-piperidinamine linker and aromatic heterocycles. 41e displayed robust GLS1 binding affinity, superior liver microsome metabolic stability, and moderate anti-tumor activity (TGI: 47.5%) in HCT116 xenograft model with no considerable toxicity in vivo. The mechanism underlying the anti-proliferative effect of 41e on HCT116 cells was studied, revealing that the compound blocked the glutamine metabolism, induced the production of ROS, and triggered apoptosis. These findings merit further investigation of 41e as a targeted cancer therapeutic.

Abstract:In order to search for novel high activity and low toxicity antitumor drugs, thirteen novel 5-fluorouracil derivatives containing amino acid Schiff base were synthesized via the condensation reactions of N-1-(2-furanidyl)-N-3-(4-bromobutyl)-5-fluorouracil and amino acid Schiff bases. The structures of the products were confirmed by IR, H-1 NMR, NIS spectra and elemental analyses. The result of preliminary bioassay shows that the new compounds possess antitumor activities to some extent.
