Mechanism Study on Liver Mitochondrial Damage in APAP- Intoxicited Mice

Abstract:OBJECTIVE To study the mechanism of liver mitochondrial damage in APAP-intoxicited mice.METHODS Mice were randomly divided into the normal group,40 mg·kg-1,80 mg·kg-1,160 mg·kg-1,320 mg·kg-1 APAP groups.Alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities and level of liver glutathione(GSH) in mice serum were detected after 12 h intraperitoneal injection with APAP.Meanwhile,the mitochondrial membrane potential,the sensitivity of mitochondrial swelling and mitochondrial calcium content were measured.RESULTS Compared with the normal group,the serum ALT and AST activities and level of liver GSH in APAP-toxicitied mice obviously raised in APAP groups.The mitochondrial membrane potential and sensitivity of mitochondrial swelling were reduced when treated with APAP.Intramitochondrial Ca2+ overload occurred in the liver in 80 mg·kg-1,160 mg·kg-1 and 320 mg·kg-1 groups.The degree of the above changes in APAP damaged mice was gradually increased with the increasing of concentration of APAP.CONCLUSION The damage to liver mitochondria indicated that the suppression on mitochondrial membrane potential and sensitivity of mitochondrial swelling occurred in all APAP exposed groups and increased gradually with the dose increasing of APAP.The mechanisms underlying the liver mitochondrial damage may be related to the excessive opening of mitochondrial permeability transition pore induced by intramitochondrial Ca2+ overload.
