Effect of Lycium Chinensis on Hippocampal Neurons of CA3 Region Induced by Chronic Psychological Stress in Rats

Abstract:Objective To study the effect of Lycium chinensis on the quantity and morphology of hippocampal neurons of CA3 region in rats induced by chronic psychological stress.Methods96 SD rats were divided into eight groups.Chronic compound psychological stress was adopted.The morphological changes of hippocampal neurons of CA3 region were observed by HE staining.ResultsThe serum cortisol levels of stress group,low dose Lycium chinensis + stress group and high dose Lycium chinensis+stress group were significantly higher than those of the control group(P<0.01).The serum cortisol levels of low,medium,and high doses Lycium chinensis groups,medium dose Lycium chinensis+stress and high dose Lycium chinensis + stress group were significantly lower than those of the stress group(P<0.01).Comparing with the control group,the quantity of hippocampal of neurons CA3 region of the stress group significantly decreased(P<0.01).Comparing with the stress group,the quantity of hippocampal neurons of CA3 region of low,medium and high doses Lycium chinensis+stress groups increased significantly(P<0.05).Cells in stress group irregularly lined,widened gap,some of nuclei condensed into triangular or irregular shapes were observed under light microscope.The morphology of low,medium,and high doses groups was basically the same with the control group.In low,medium and high doses+stress groups,the hippocampal cells patterns were in-between the control group and the stress group.ConclusionsChronic psychological stress may induce the serum cortisol levels to rise causing the damage of hippocampal CA3 pyramidal cell.Lycium chinensis may reduce the levels of serum cortisol.By reducing the injury of stressed hippocampus,there would be protective effect on injured nerve element.
