Abstract:BACKGROUND: PhiC31 is an important enzyme in gene therapy, and its interaction with intracellular proteins may effect the cellular pathway and therefore influence the safety of gene therapy strategies. OBJECTIVE: To construct a zinc finger protein 403 (Znf403) prokaryotic vector and observe its expression in eucaryotic cells. DESIGN, TIME AND SETTING: A single sample experiment was performed at the State Key Laboratory of Genetic Engineering in Fudan University from October 2007 to December 2008. MATERIALS: Both colibacillus DH5αstrain and colibacillus Rosetta (DE3) strain were the preserved ones in the laboratory. METHOD: Human fetal brain cDNA library was screened using pLexA-phiC31 as "bait", after which positive clones were isolated. Znf 403 prokaryotic expression vector was constructed and then purified. Cellular transfection and localization observation were performed to eucaryotic cells under mediation with liposome. MAIN OUTCOME MEASURES: Construction and purification of Znf430 prokaryotic expression vector as well as its location in the cytoplasm of eucaryotic cells. RESULTS: Among the 61 positive clones isolated from screened clones with yeast two-hybrid, one consisted of gene sequence on 17 chromosome 17q12. The protein coded by the gene possessing a unique zinc finger structure and thus was named Znf 430. Znf430 prokaryotic expression vector was constructed successfully and the his-tagged Znf430 was purified. In eucaryotic cells, green fluorescence fusion vector was constructed which showed the localization of this protein in cytoplasm. CONCLUSION: Our study reveals a new phiC31 integrase interacting protein, and constructs Znf430 prokaryotic expression vector successfully. Znf 430 mainly accumulates and distributes in the cytoplasm.
