Optimization on Medium for Tissue Culture and Rapid Propagation of Polygonum Multiflorum

Abstract:To screen the optimum culture condition for the tissue culture of Polygonum multiflorum,the stem with axillary bud of P.multiflorum.was used as explants in the experiment with the completely random block design and the media inducing multiple shoots and adventitious roots contained different concentrations of 6-BA,NAA,IBA,6-BA and NAA,6-BA,IBA and paclobutrazol).The results showed that 6-BA could promote to induce multiple shoots,and NAA,IBA inhibit the induction of buds in a certain concentration range.Moreover,the optimum medium for multiplication and differentiation was MS+6-BA 1.0 mg/L and its multiplication coefficient reached 11.43.Paclobutrazol had a promotion on the adventitious root induction,the optimum rooting medium for 100% rooting rate was 1/2 MS+ paclobutrazol 0.5 mg/L,and average rooting number was 10.42 per plant.
