Abstract:A simple, air-stable, inexpensive and easily prepared molecule, N-methyliminodiacetic acid (MIDA), is reported as a ligand for palladium-catalyzed Suzuki-Miyaura cross-coupling reaction of phenylboronic acid with aryl chlorides. The yield of the corresponding Suzuki coupling reaction is up to around 90% at both high temperature of 80 degrees C and room temperature under ambient atmosphere. Abstract:Palladium-catalyzed C-H functionalization of acyldiazomethanes with aryl iodides has been developed. This reaction is featured by the retention of the diazo functionality in the transformation, thus constituting a novel method for the introduction of diazo functionality to organic molecules. Consistent with the experimental results, the density functional theory (DFT) calculation indicates that the formation of Pd-carbene species in the catalytic cycle through dinitrogen extrusion from the palladium ethyl diazoacetate (Pd-EDA) complex is less favorable. The reaction instead proceeds through Ag2CO3 assisted deprotonation and subsequently reductive elimination to afford the products with diazo functionality remained. This C-H functionalization transformation can be further combined with the recently evolved palladium-catalyzed cross-coupling reaction of diazo compounds with aryl iodides to develop a tandem coupling process for the synthesis of α,α-diaryl esters. DFT calculation supports the involvement of Pd-carbene as reactive intermediate in the catalytic cycle, which goes through facile carbene migratory insertion with a low energy barrier (3.8 kcal/mol).

Abstract:The Suzuki-Miyaura coupling is among the most important C-C bond-forming reactions available due to its reliability, chemoselectivity, and diversity. Aryl halides and pseudohalides such as iodides, bromides, and triflates are traditionally used as the electrophilic coupling partner. The expansion of the reaction scope to nontraditional electrophiles is an ongoing challenge to enable an even greater number of useful products to be made from simple starting materials. Herein, we present how an NHC-based Pd catalyst can enable Suzuki-Miyaura coupling where the C(acyl)-O bond of aryl esters takes on the role of electrophile, allowing the synthesis of various ketone-containing products. This contrasts known reactions of similar esters that provide biaryls via nickel catalysis. The underlying cause of this mechanistic divergence is investigated by DFT calculations, and the robustness of esters compared to more electrophilic acylative coupling partners is analyzed.

Abstract:In the presence of Na2CO3 the combination of PdCl2(dppf), dppp and CuI catalyzes the decarbonylative coupling of arylcarboxylic acid 2-pyridyl esters with terminal alkynes to afford 1,2-disubstituted acetylenes. (Hetero)aryl, alkyl, and silylacetylenes and various electron-donor and -withdrawing group-substituted arylcarboxylic acid 2-pyridyl esters can be used in this transformation. A range of functional groups can be compatible.

Abstract:A palladium-catalyzed Negishi cross-coupling reaction of ethyl bromodifluoroacetate with aryl bromides or aryl triflates to construct C(sp2)-CF2 bonds is described. The reaction was conducted under mild reaction conditions, and no preparation of organozinc reagents is required. This is the first report encompassing the conversion of aryl triflates into products containing C-CF2 bonds. In addition, the construction of C(sp2)-CHF bonds was achieved under mild conditions via a cross-coupling of aryl iodides with ethyl bromofluoroacetate.

Abstract:Palladium-catalyzed Suzuki-Miyaura cross-coupling or aryl halides is widely employed in the synthesis of many important molecules in synthetic chemistry, including pharmaceuticals, polymers and functional materials. Herein, we disclose the first palladium-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of amides for the synthesis of biaryls through the selective activation of the N-C(O) bond of amides. This new method relies on the precise sequence engineering of the catalytic cycle, wherein decarbonylation occurs prior to the transmetallation step. The reaction is compatible with a wide range of boronic acids and amides, providing valuable biaryls in high yields (>60 examples). DFT studies support a mechanism involving oxidative addition, decarbonylation and transmetallation and provide insight into high N-C(O) bond activation selectivity. Most crucially, the reaction establishes the use of palladium catalysis in the biaryl Suzuki-Miyaura cross-coupling of the amide bond and should enable the design of a wide variety of cross-coupling methods in which palladium rivals the traditional biaryl synthesis from aryl halides and pseudohalides.

Abstract:2-Arylphenols were conveniently synthesized from aryl iodides and 6-diazo-2-cyclohexenones, in moderate to excellent yields, via tandem Pd-catalyzed cross-coupling/aromatization. The preliminary results for the corresponding enantioselective version showed that the coupling products could be generated in up to 72% ee.

Abstract:Pd-catalyzed decarboxylative cross-couplings of 2-(2-azaaryl)acetates with aryl halides and triflates have been discovered. This reaction is potentially useful for the synthesis of some functionalized pyridines, quinolines, pyrazines, benzoxazoles, and benzothiazoles. Theoretical analysis shows that the nitrogen atom at the 2-position of the heteroaromatics directly coordinates to Pd(II) in the decarboxylation transition state.

Abstract:Efficient palladium-catalyzed cross-coupling reactions of alpha-diazocarbonyl compounds and arylboronic acids or aryl halides have been developed. The reaction proceeds smoothly for a range of diazo compounds, boronic acids, and halides. The coupling reaction conditions tolerate various substituents on the aromatic rings of the substrates, such as chloro, fluoro, acyl, oxo, ester, and nitro groups. This coupling reaction constitutes a novel access to alpha-aryl-substituted alpha,beta-unsaturated carbonyl compounds. Mechanistically, palladium-carbene is supposed to be the key intermediate; its formation is followed by migratory insertion of an aryl group to the carbenic carbon of the palladium-carbene complex and subsequent beta-hydride elimination. Kinetic isotope effect (KIE) data measured for intra- and intermolecular competition experiments suggest that beta-hydride elimination is not involved in the rate-determining step.

Abstract:The first efficient reductive cross-couplings of aryl halides mediated by an abundant feedstock, sodium formate, are de-scribed. These processes, which exploit air-stable Pd(I) iodide dimers, are especially effective for challenging 2-pyridyl systems due to intervention of chelated intermediates. Furthermore, orthogonality with respect to Suzuki and Buchwald-Hartwig coupling processes is displayed, as pinacol boronates and anilines are tolerated. Although palladium-catalyzed transfer hydrogenolyses of aryl halides mediated by formate are longstanding, it is not a major competing pathway under the present conditions. Experimental and computational studies corroborate a novel catalytic cycle for cross-coupling where the Pd(I) precatalyst, [Pd(I)(PtBu3)]2, is converted to the active dianionic catalyst, [Pd2I4][NBu4]2, from which aryl halide oxi-dative addition is more facile. Rapid, reversible Pd-to-Pd transmetalation delivers iodide-bridged diarylpalladium dimers. The hetero-diarylpalladium dimers are more stable than the homodimers and have lower barriers to reductive elimination, resulting in high cross-selectivity.
