ROLES OF BRADYKININ RECEPTORS IN NEURONAL DAMAGE CAUSED BY HYPOXIA/REOXYGENATION

Abstract:To explore the roles of bradykinin B1 and B2 receptor(B1R and B2R) in neuronal damage caused by hypoxia/reoxygenation(H/R),the expressions of B1R and B2R in primary cultured rat cortical neurons were detected by using immunofluoresence cytochemical method.The model of H/R neurons for mimicking ischemia/reperfusion in vivo was established,and the neuronal survival and apoptosis were determined by methods of MTT and TUNEL,respectively.The neuronal survival was also examined after the B1R or B2R was activated or inhibited under the condition of H/R.The results showed that there was an obvious B2R expression in the neurons cultured under normal condition.H/R not only induced B1R and B2R expressions,but also caused an increase in neuronal apoptosis.Under the condition of H/R,the apoptosis of neurons could be aggravated by activating B1R,but was decreased by activating B2R.Moreover,the effects of B1R and B2R on neuronal damage could be inhibited by their specific antagonists.These results indicated that B1R and B2R should exert different functions to neuronal damage caused by H/R,and it may be benefit to treat some diseases such as cerebral ischemia by using inhibiting B1R and enhancing B2R expression.
