Abstract:Two types of dendritically functionalized iron(II) porphyrins were prepared (Scheme) and investigated in the presence of 1,2-dimethylimidazole (1,2-DiMeIm) as the axial ligand as model systems for T(tense)-state hemoglobin (Hb) and myoglobin (Mb). Equilibrium O2- and CO-binding studies were performed in toluene and aqueous phosphate buffer (pH 7). UV/VIS Titrations (Fig. 4) revealed that the two dendritic receptors 1 Fe-II-1,2-DiMeIm and 2 . Fe-II-1,2-DiMeIm (Fig. 2) with secondary amide moieties in the dendritic branching undergo reversible complexation (Fig. 5) with O-2 and CO in dry toluene. Whereas the CO affinity is similar to that measured for the natural receptors, the 02 affinity is greatly enhanced and exceeds that of T-state Hb by a factor of ca. 1500 (Table). The oxygenated complexes possess half-lives of several h (Fig. 6). This remarkable stability originates from both dendritic encapsulation of the iron(II) porphyrin and formation of a H-bond between bound O-2 and a dendritic amide NH moiety (Fig. 11). Whereas reversible CO binding was also observed in aqueous solution (Fig. 10), the oxygenated iron(II) complexes are destabilized by the presence of H2O with respect to oxidative decay (Fig. 9), possibly as a result of the weakening of the O-2...H-N H-bond by the competitive solvent. The comparison between the two dendrimers with amide branchings and ester derivative 3 . Fe-II-1,2-DiMeIm (Fig. 2), which lacks H-bond donor centers in the periphery of the porphyrin, further supports the role of H-bonding in stabilizing the O-2 complex against irreversible oxidation. All three derivatives bind CO reversibly and with similar affinity (Fig. 8) in dry toluene, but the oxygenated complex of 3 . Fe-II-1,2-DiMeIm undergoes much more rapid oxidative decomposition (Fig. 7).
The Strong Cooperativity of Dioxygen Binding by Dicobalt (II) Diporphyrin Compounds in Nonaqueous Solution at Room Temperature

Abstract:Thermodynamic data were determined for the reversible binding of O2 to two compounds of dicobalt(II) di(meso-tetra-phenyl)porphyrin derivatives with different lengths of diamidoaliphatic bridge (abbreviated to Co2PP8 and Co2PP4) in N,N-dimethylformamide at room temperature. The partial pressure of dioxygen necessary for half-oxygenation (P 1/2) and hill coefficient (n) at 298 K were determined as follows:P 1/2=54.2 kPa,n=2.0 for Co2PP8 andP 1/2=6.8 kPa,n=1.8 for Co2PP4, respectively. The rate equations of reversible oxygen binding by Co2PP8 were determined and the reaction path was proposed. The results of ther-modynamic and kinetic studies indicate that there exists strong cooperative effect during oxygenation of the compounds. The ESR observation reveals that the dioxygen complexes formed in the solutions are of superoxo (Co-O 2 − ) type.

Abstract:Why does binding of dioxygen (O2) to metal centers, the initial step of O2 storage, transportation, and activation, almost inevitably induce metal-to-O2 single-electron transfer and generate superoxo (O_2^(-∙)) species, instead of genuine O20 adducts? To address this question, this study describes highly correlated wavefunction based ab initio calculations using CASSCF/NEVPT2 (CASSCF = complete active space self-consistent field, and NEVPT2 = N-electron valence state second-order perturbation theory) approaches to explore the electronic-structure evolution of O2 binding on Fe(II)(BDPP) (H2BDPP = 2,6-bis((2-(S)-diphenylhydroxylmethyl-1-pyrrolidinyl)methyl)pyridine) and Co(II)(BDPP) to produce S = 3 Fe(III)(BDPP)(O_2^(-∙)) (1) and S = 1/2 Co(III)(BDPP)(O_2^(-∙)) (2). CASSCF/NEVPT2 calculations suggest that the O2 binding processes furnishing 1 and 2 feature an avoided crossing resulting from interactions of two diabatic curves, of which one is characterized as Co(II) and Fe(II) centers interacting with a triplet O2 ligand and the other as Co(III) and Fe(III) centers bound to a superoxo ligand. In both cases, the avoided crossing induces a one-electron transfer from the divalent metal center to the incoming O2 ligand and leads to formation of trivalent metal-O_2^(-∙) complexes. To facilitate interpretation of complicated multireference wavefunctions, we formulated two-fragment spin eigenfunctions utilizing Clebsch-Gordan coefficients (CGCs) to rationalize computed spin populations on metal centers and O2 ligand and compared it with usual valence bonding (VB) analyses. It turns out that both methods give the same results and are complementary to each other. Finally, the limitation of DFT approaches in describing complex electronic structures involving metal-ligand magnetic coupling is delineated.

Metalloporphyrin-dioxygen interactions and the effects of neutral axial ligands

Abstract:We present here results of a series of density functional theory studies on models of metalloporphyrins and address the influence of nitrogen-containing neutral axial ligands on the binding process of dioxygen (O-2) to metalloporphyrins. There are two closely lying spin states for metalloporphyrins, namely, triplet and quintet. However, the ground state for oxy-metalloporphyrins is the open-shell singlet. DFT calculations indicate that back-donation to O-2 is more accessible because of the axial ligand, facilitating the binding of O-2. Moreover, the axial ligands lengthen the O-O bond of oxy-metalloporphyrins and make the bound O-2 negatively charged with a spin population between that Of O-3(2) and O-2(-). Therefore, the coordination of the axial ligand makes O-2 more active than free O-2, triggering the catalytic oxidation processes. Finally, the larger the electronic density of the bound nitrogen atom (N-L) of the axial ligand is, the more active the binding O-2 becomes.

Abstract:The reaction of dioxygen with a dicobalt di( meso - tetraphenylporphyrin) linked by a diamido - aliphatic chain in N, N - dimethylformamide solution was studied. Thermodynamic values under standard state (298K, 100kPa O-2) are: P-1/2 = 6.8kPa, n = 1.84, DeltaG(0) = - 12.3kJ . mol-(1), DeltaH(0) = - 172.09kJ . mol(-1), DeltaS(0) = - 536.21 J . mol(-1).K-1. Thermodynamic data show that each of the two cobalt( II) porphyrin rings has strong cooperative effect of oxygen binding with another. The reaction of dioxygen with the complex was also studied by ESR spectroscopy. The results show that bonding way of dioxygen complex is of the superoxo Co - O-2(-) type.
Toward Understanding Macrocycle Specificity of Iron on the Dioxygen-Binding Ability: a Theoretical Study

Abstract:In an effort to examine the interaction between dioxygen and iron-macrocyclic complexes, and to understand how this interaction was affected by those different macrocyclic ligands, dioxygen binding with iron-porphyrin, iron-phthalocyanine, iron-dibenzotetraaza[14]annulene, and iron-salen complexes is investigated by means of quantum chemical calculations utilizing Density Functional Theory (DFT). Based on the analysis of factors influencing the corresponding dioxygen binding process, it showed that different macrocyclic ligands possess different O-O bond distances, and different electronic configurations for the bound O(2) and non-aromatic macrocyclic ligands favor dioxygen activation. Furthermore, the smaller the energy gap between the HOMO of iron-macrocyclic complexes and the LUMO of dioxygen, the more active the bound O(2) becomes, with a longer O-O bond distance and a shorter Fe-O bond length.

Abstract:A disulfide-bridged dimeric cobalt porphyrin 2Co has been successfully synthesized. The cyclic voltam-mogram (CV) curves of 2Co exhibited three oxidation and two reduction potentials in CH2Cl2, indicating that the 2Co enable stabilizing multi-negative and positive charges. The ORR catalytic property of 2Co in acid media were investigated. The reduction of O-2 using the dimer 2Co as catalyst showed high stability and activity to give the higher the value of transferred electrons number (n) between 3.5 and 3.6 under dioxygen. The catalytic performance of 2Co indicates that dimerization of cobalt porphyrins have a slightly positive influence on the catalytic properties for oxygen reduction reactions (ORRs) in the acidic medium.

Abstract:Dioxygen adducts of [Fe2L(O2CC6H5)]X2, where L represents the dinucleating ligands HPTB (anion of N,N,N',N'-tetrakis(2-benzimidazolylmethyl)-2-hydroxy-1,3-diaminopropane), its N-ethyl analogue, and its tetrakis(pyridine) analogue, HPTP, can form and serve as models for the putative oxygenated intermediates of methane monooxygenase and ribonucleotide reductase. [Fe2(N-Et-HPTB)(O2CC6H5)](BF4)2 (1) crystallizes in the triclinic space group P1BAR with cell constants a = 13.04 (1) angstrom, b = 14.248 (7) angstrom, c = 18.09 (1) angstrom, alpha = 73.56 (6)degrees, beta = 78.22 (7)degrees, gamma = 67.71 (6)degrees, V = 2963 (9) angstrom3, Z = 2; R = 0.069, and R(w) = 0.085. The Fe(II) sites are bridged by the alkoxide of the dinucleating ligand and a benzoate, affording a diiron core with an Fe-mu-O-Fe angle of 124.0 (3)degrees and an Fe-Fe distance of 3.473 (7) angstrom. Both Fe(II) centers have trigonal bypyramidal geometry, and NMR studies show that the remaining coordination sites are accessible to ligands such as DMSO and Ph3PO. The iron centers are antiferromagnetically coupled with J is similar to 20-26 cm-1 (H = JS1.S2). Irreversible dioxygen adducts form upon exposure of the diferrous complexes to O2 at low temperatures. The 1/O2 adduct and its HPTB analogue, 2/O2, are stable indefinitely in CH2Cl2 at -60-degrees-C but decompose upon warming; the addition of DMSO or other polar aprotic solvents further stabilizes the adducts, allowing them to persist for short periods even at ambient temperature. The adduct of the pyridine analogue, 3/O2, on the other hand, is not observed at -80-degrees-C unless a polar aprotic solvent is added to the CH2Cl2 solution. The adducts exhibit visible absorption maxima near 600 nm and resonance Raman features at approximately 470 cm-1 (nu(Fe-O)) and approximately 890-900 cm-1 (nu(O-O)). The latter is characteristic of a mu-1,2-peroxo species; in support, the NMR properties of the HPTB adducts indicate the presence of a moderately strong antiferromagnetic coupling interaction (J is similar to 140 cm-1). Carboxylate substitution on 1 effects a shift of the absorption maximum of the adduct, indicating that the carboxylate remains coordinated in the adduct. Thus, the adducts are proposed to have tribridged (mu-1,2-peroxo)(mu-carboxylato)(mu-alkoxo)diferric cores. The differing stabilities of the dioxygen adducts are also reflected in differences in reactivity. The addition of 2,4-di-tert-butylphenol or Ph3P does not affect the 1/O2 adduct at -50-degrees-C but does accelerate the decomposition of the 3/O2 adduct, affording 0.5-0.6 equiv of the corresponding biphenol or OPPh3, respectively. The one-electron oxidation of a phenol by 3/O2 suggests that such an oxygenated species may be involved in the mechanism of the tyrosyl radical formation in ribonucleotide reductase; however, some further activation step is likely to be required for such a species to participate in the alkane hydroxylation mechanism of methane monooxygenase.

Abstract:This paper reports the thermodynamics of reversible oxygen binding to a dicobalt di(meso-tetraphenylporphyrin) linked by a diamido- aliphatic chain in N,N-dimethylformamide solution. Thermodynamic values(standard state 101325 PaO2): Delta G(e)(298 K)=-3.1 kJ center dot mol(-1), Delta H-e (298 K)=-37.1 kJ center dot mol(-1), Delta S-e (298 K)=-114 J center dot mol(-1 center dot)K(-1), Co:O-2=1:1. The ESR parameters of the dioxygen complex in DMF at 133 K are as follows: g(parallel to)=2.000, g(parallel to)=2.046, A(_)(C0)=15G, A(0 parallel to)(C)=22G, The pressure of dioxygen necessary for half-oxygenation of the system (p(1/2)=42.7 kPa at 288 K) is comparable with that for cobalt substituted human hemoglobin at T state.

A Study of Conformation Equilibrium and π-πInteraction of Porphyrin Dimers

Abstract:A series of porphyrin dimers and their zinc complex covalently linked with -O(CH2)nO-(n: 2-10) chain were synthesized and characterized. Their steady-state fluorescence spectra show the existence of intramolecular energy transfer processes in these free-base porphyrin dimers. C4 free dimer has the greatest efficiency of energy transfer. By using IR and 1H NMR methods, the relationship between the conformation equilibrium and the length of the alkoxyl chain, the metal complexation was studied. With the increase of the length of the chain, the conformation equilibrium of porphyrin dimers moves to the closed conformation, the chain length of C4 dimer was suitable to form the closed conformation. The metallation with zinc ion increases the magnitude of π-π interaction of two porphyrin rings while it does not affect the conformation equilibrium of porphyrin dimers.
The Secondary Coordination Sphere and Axial Ligand Effects on Oxygen Reduction Reaction by Iron Porphyrins: A Dft Computational Study

Abstract:Oxygen reduction reaction (ORR) catalyzed by a bio-inspired iron porphyrin bearing a hanging carboxylic acid group over the porphyrin ring, and a tethered axial imidazole ligand was studied by DFT calculations. BP86 free energy calculations of the redox potentials and pK a’s of reaction components involved in the proton coupled electron transfer (PCET) reactions of the ferric-hydroxo and -superoxo complexes were performed based on Born–Haber thermodynamic cycle in conjunction with a continuum solvation model. The comparison was made with iron porphyrins that lack either in the hanging acid group or axial ligand, suggesting that H-bond interaction between the carboxylic acid and iron-bound hydroxo, aquo, superoxo, and peroxo ligands (de)stabilizes the Fe–O bonding, resulting in the increase in the reduction potential of the ferric complexes. The axial ligand interaction with the imidazole raises the affinity of the iron-bound superoxo and peroxo ligands for proton. In addition, a low-spin end-on ferric-hydroperoxo intermediate, a key precursor for O–O cleavage, can be stabilized in the presence of axial ligation. Thus, selective and efficient ORR of iron porphyrin can be achieved with the aid of the secondary coordination sphere and axial ligand interactions.

Abstract:Substituent effects of iron porphyrin complexes on the structures and kinetic processes have been examined for the first time. Basing on the premise that iron porphyrin is functional analogous to heme, a series of iron porphyrin derivatives bearing different substituents at the meso positions of the corrole ring are investigated as to their electrochemistry, the relationships among the electron transfer (ET) processes, their structures, and orbital energies. The good coherence between the experiment and theory indicates that the ET rate can be accelerated when electron-donating substituents are introduced to the iron porphyrin ring. Finally, the implications of the results are discussed in the influence of stability of iron porphyrin complexes on the ability to carry molecular oxygen, which may suggest it possible to dominate the biological activity of heme by selecting the appropriate substituents to iron porphyrin ring.

Synthesis, Crystal Structure of Diferrous Complex and Its Reactivity with Dioxygen

Abstract:The dinuclear complex of [Fe 2 L{O 2 P(OPh) 2 }](ClO 4 ) 2 · Et 2 O(1) (where L represents the dinucleating ligand N-Et-HPTB, anion of N, N, N’, N’-tetrakis (N-ethyl-2-benzimidazolyemethyl)2-hydroxy-1, 3-diamino-propane) has been synthesized and crystallizes in the triclinic space group P1 with cell constants a = 1.526(3) nm, b = 1.259 8(3) nm, c = 1.563 0(3)° nm, α = 94.41(3)°, β = 115.31(3)°, γ = 99.90(3)°, V = 3.267(1) nm 3 , z = 2, R = 0.084 7 and R w = 0.177 8. The Fe(II) sites are bridged by an alkoxide of the dinucleating ligand and a phosphate, affording a diiron core with an Fe-μ-O-Fe angle of 131.20(3)° and an Fe-Fe distance of 0.364 9 nm. Both Fe(II) centers have trigonal bipyramidal geometry. Dioxygen adduct (1/O 2 ) forms upon exposure of the diferrous complex to O 2 at low temperature (?60°C). The 1/O 2 adduct is stable at ? 60°C but decomposes upon warming. The adduct exhibits visible absorption maximum near 606 nm and resonance Raman features at 478 cm -1 (γ Fe-O ) and 897 cm -1 (γ O-O ), and the latter is the characteristic of a μ-1,2-peroxo species, indicating that dioxygen adducts can serve as models for the putative oxygenated intermediate of some non-heme diiron-oxo proteins.
Density Functional Theory Study of the Oxygen Reduction Reaction on Metalloporphyrins and Metallophthalocyanines

Abstract:In this paper, density functional theory is applied to study the electrochemical reduction of oxygen on iron phthalocyanine (FePc), iron porphyrin (FeP), cobalt phthalocyanine (CoPc), and cobalt porphyrin (CoP). According to the calculation results, for the four metal macrocyclic complexes, O-2 will not directly be cleaved without the cooperation of hydrogen. In the reduction process, on FePc or FeP, H2O2 does not form as an intermediate, and O-2 is reduced to H2O, while on CoPc or CoP, O-2 is just reduced to H2O2. The reason why the oxygen reduction ability of FePc or FeP is higher than that of CoPc or CoP, respectively, is that the energy level of the highest-occupied 3d orbital of the former is higher than that of the later. The high energy level of the metal 3d orbital leads to the strong ability of oxygen reduction.
Protoporphyrin IX iron(II) revisited. An overview of the Mössbauer spectroscopic parameters of low-spin porphyrin iron(II) complexes

Abstract:Mössbauer parameters of low-spin six-coordinate [Fe(II)(Por)L2] complexes (where Por is a synthetic porphyrin; L is a nitrogenous aliphatic, an aromatic base or a heterocyclic ligand, a P-bonding ligand, CO or CN) and low-spin [Fe(Por)LX] complexes (where L and X are different ligands) are reported. A known point charge calculation approach was extended to investigate how the axial ligands and the four porphyrinato-N atoms generate the observed quadrupole splittings (ΔEQ) for the complexes. Partial quadrupole splitting (p.q.s.) and partial chemical shifts (p.c.s.) values were derived for all the axial ligands, and porphyrins reported in the literature. The values for each porphyrin are different emphasising the importance/uniqueness of the [Fe(PPIX)] moiety, (which is ubiquitous in nature). This new analysis enabled the construction of figures relating p.c.s and p.q.s values. The relationships presented in the figures indicates that strong field ligands such as CO can, and do change the sign of the electric field gradient in the [Fe(II)(Por)L2] complexes. The limiting p.q.s. value a ligand can have and still form a six-coordinate low-spin [Fe(II)(Por)L2] complex is established. It is shown that the control the porphyrin ligands exert on the low-spin Fe(II) atom limits its bonding to a defined range of axial ligands; outside this range the spin state of the iron is unstable and five-coordinate high-spin complexes are favoured. Amongst many conclusions, it was found that oxygen cannot form a stable low-spin [Fe(II)(Por)L(O2)] complex and that oxy-haemoglobin is best described as an [Fe(III)(Por)L(O2-)] complex, the iron is ferric bound to the superoxide molecule.

Abstract:In this paper the oxygenation of HDTHCo homo and heterodinuclear complexes with Cu(II), Ni(II) and Pb(II) in aqueous solution by control of the stoichiometry of metal ions and HDTH as well as p[H] of solution was investigated (HDTH is a dinucleating 28-membered hexaazadiphenol macrocyclic ligand, 3,7,11,19,23,27-hexaaza-33,34-dihydroxy-15,31-dimethyl-tricyclo-tetratriaconta-1(32),13,15,17(34),29(33),30-hexaene). The pH potentiometric method was utilized successfully to determine oxygenation constants and to determine the distribution of species present in the solution as a function of p[H]. Spectrophotometry was used to investigate the oxygenation process of the homo and heterodinuclear complexes. The X-ray crystal structure of homodinuclear complexes of Ni(II) is also reported. These studies suggested autooxidation takes place during the oxygenation of homo and heterodinuclear Co(II) complexes of the macrocyclic ligand. The neighboring effect increases in the order Ni(II)<Cu(II)<Pb(II)<Co(II). Pb(II) stimulates the neighboring Co(II) to accept dioxygen in its sixth vacant position. Ni(II) is not helpful to Co(II) in its oxygenation.
Hydroxylation of cyclohexane catalyzed by iron(III)–iron(III) porphyrin dimers and DABCO with molecular oxygen: evidence for the conformation effect of porphyrin dimers on the catalytic activity

Abstract:A series of p/p type iron(III)–iron(III) porphyrin dimers and corresponding zinc(II)–zinc(II) porphyrin dimers covalently linked by a flexible alkoxy chain (–O(CH2)nO–, n=2–10) have been synthesized and characterized. The hydroxylation of cyclohexane catalyzed by iron(III)–iron(III) porphyrin dimers with molecular oxygen under mild conditions are reported, 1 eq. of bidentate ligand DABCO (1,4-diazobicyclo[2,2,2] octane) is added exactly as axial ligand to the catalytic system. The catalytic activities of these dimers are supposed to be related to the special conformations formed by porphyrin dimers and DABCO. By using 1H-NMR, the conformation equilibrium of corresponding zinc(II)–zinc(II) porphyrin dimers and their binding behavior with 1 eq. DABCO has been discussed in detail. It has been proven that the higher catalytic activities of the catalytic system mainly result from the formation of the ternary sandwich complexes of iron(III)–iron(III) porphyrin dimers with DABCO.
