Abstract:The active intermediates of hexacoordinated 4-nitrotrispirophosphoric amino acid (P(6)-AA), which were formed by the reaction of 4-nitro-O,O-phenylene phosphochloridate (NOPPC) with silanizing serine, threonine and cysteine, were observed by 31P NMR spectra. The reaction mechanism of these processes was discussed. In addition, some interesting results were observed by mixing 4-nitro-2,2,2-tricloro-1,3,2-benzodioxaphosph(v)ole (NTB) with silanizing serine and histidine.

Abstract:RATIONALE: alpha-Amino phosphonates with intrinsic biological activities have been used in a wide variety of applications. Because of the widespread existence of natural organophosphorus compounds containing P-C bonds such as the alpha-amino phosphonates, it is important to investigate the gas-phase chemistry of P-C bonds in order to determine their basic properties, which might provide some insights into their biosynthesis and catalytic cleavage.METHODS: Twenty alpha-amino phosphonates were successfully synthesized and their fragmentation behavior was systematically investigated using in-solution deuterium labeling in combination with high-resolution Fourier transform ion cyclotron resonance (FTICR) electrospray ionization tandem mass spectrometry.RESULTS: The fragmentation pathways of twenty alpha-amino phosphonates with different chemical structures were systematically studied. In general, P-C bonds could be easily cleaved via a novel intramolecular hydrogen atom migration from the amino group to the phosphoryl group through a five-membered-ring intermediate in the gas phase. A possible mechanism of the rearrangement of alpha-amino phosphonates is proposed.CONCLUSIONS: An interesting intramolecular hydrogen atom migration between the amino and phosphoryl groups was observed with cleavage of the P-C bond in the molecule through a five-membered-ring intermediate. This characteristic fragmentation pathway not only provides some insights into the basic chemistry of compounds with P-C bonds, but could also have some applications in the structural determination of the alpha-amino phosphonate analogues. Abstract:The β-OH group is more active than γ-OH in many processes, such as the catalysis of the active site in enzymes and the N→O rearrangement of N-phosphoryl amino acids in negative-ion electrospray ionization mass spectrometry. The pentacoordinate phosphoric intermediates are proposed and the rearrangement mechanisms are studied by ab initio and density functional calculations. The formation of the intermediates comprises a concerted route and two different stepwise pathways. The concerted route is more favored. The cleavage of the P-N bond is the rate-determining step and is favored by the proton transfer. For the rearrangement of dimethoxyl phosphoryl serine anion and homoserine anion, the activation energies are 91.2 and 156.9 kJ/mol at the B3LYP/6-31+G(d,p) level, respectively. Therefore, the β-OH group is predicted to be more active than γ-OH and the result is in good agreement with the experiments.

Abstract:The cationized N-diisopropyloxyphosphoryl amino acids/alcohols were analyzed by electrospray ionization tandem mass spectrometry. A rearrangement reaction leading to the P-N to P-O bond migration was observed, and cationized phosphoric acid isopropyl ester (PAIE) was formed. The rearrangement did not depend on the chain length of the amino acids/alcohols. A possible rearrangement mechanism was proposed. Abstract:P-31 NMR has become a widely applied spectroscopic probe of the structure of phosphorus-containing compounds. Meanwhile: the application of P-31 NMR has been rapidly expanded to biochemistry and medicinal chemistry of phosphorus-containing compounds because the growing importance of the phosphorus compounds is now widely realized. We report here the results of P-31 NMR study on some phosphorus-containing compounds, namely, O-alkyl O-4-nitrophenyl methyl phosphonates with different alkyl chain-length (MePO-n), 4-nitrophenyl alkylphenylphosphinates with different alkyl chain-length (PhP-n), diethyl phosphono- acetonitrile anion and diethyl phosphite anion Our results indicate that P-31 NMR can not only be applied to not only the study of the hydrolytic reactions of MePO-8 and PhP-8 bur also be applied to the study of thr presence of the: anions of diethylphosphonoaceronitrile and diethyl phosphite in nucleophilic reactions.

Abstract:A series of N-(N-alkyl)- and N-(O-alkyl)phosphopeptide analogs were determined by positive and negative ion fast atom bombardment mass spectrometry and their fragmentation pathways were investigated. Their mass spectra gave abundant [MH - NH2R'](+), [MH - HOR "](+), [MH - H2O](+) or [M - H - NH2R'](-), [M - H - HOR "](-) ions, which contained pentavalent tricoordinate phosphorus ions through the loss of amine or water. In the positive ion mass spectra, +NH3R' and +NH3CHRCONHR' were found, while [MH - NH2CHRCONHR'](+) was suppressed. In the negative ion mass spectra, the pseudomolecular ion [M - H](-) was the base peak, and almost all the fragment ions contained a phosphoryl group. (C) 1997 by John Wiley & Sons, Ltd.

Abstract:The ester exchange reaction on phosphorus of N-phosphorylhistidine with primary alcohol has been studied by MNDO method. Due to the participation of imidazole group, a hexa-coordinate phosphorus intermediate was formed, in which the iso-propoxyl group opposite to the imidazole was more labile than the others and much easier to leave because the activation energy was lowered by 64kJ·mol -1. When this iso-propoxyl left, to a new form penta-coordinate phosphorus intermediate which then was to be attacked by an alcohol from the direction opposite to the imidazole group. The mechanism of hexa-coordinate phosphorus intermediate explains the catalytic effect of imidazole.

Abstract:In this paper, based on known crystal structures of square pyramid (SP) and trigonal bipyramid (TBP) penta coordinated phosphorus compounds containing amino acid side chains, such as amino, carboxyl, hydroxyl or thiol, a software for survey the P(5)-structure of phosphorylated proteins was derived. By this software, it was found that 382 of 398 phosphorus related kinases (96%) from current PDB could go through the penta-coordinated phosphorus transition state or intermediate.

Abstract:The phosphoryl N --> O migration reaction and ester exchange reaction on phosphorus of phosphoserine with methyl alcohol involving hexa-coordinate phosphorus intermediate 3 had been studied by MNDO method, The three isomers structures of 3 were discussed in detail based on the calculation results. Among the six bonds in 3, the P-O bond between the phosphorus and carboxyl oxygen O3 was the weakest one, and could be easily broken to form a new penta-coordinate intermediate 4 which could yield the phosphoryl N-->O migration product 5 by breaking the weaker P-N bond, For these two isopropoxyl groups that bonded to phosphorus in 3, the one situated opposite to O6 of serine side chain, was easier to leave than the other one by about 37 kJ/mol to produce the intermediate 6. Then a methanol could attack phosphorus of 6 from this new formed vacancy position to yield the ester exchanged product, Thus, the hexa-coordinate phosphorus intermediate mechanism could be used to explain various bio-mimic reactions for phosphoserine.

Activities of Alpha-Cooh Vs Gamma-Cooh in N-Phosphoryl Amino Acids: A Theoretical Study

Abstract:The biomimic reactions of N-phosphoryl amino acids are very important in the study of many biochemical processes, such as origin of proteins and phosphorylation of amino acid residues in the proteins. The differences in reactivities between (alpha-COOH and gamma-COOH in phosphoryl amino acids were studied by ab initio and density functional methods. The pentacoordinate phosphoric intermediates 2 containing five-membered rings were predicted to be more stable than 3 with seven-membered rings. For intermediates 3, the isomers with apical-equatorial ring spanning arrangement were predicted to be more stable than those with diequatorial ring spanning arrangment. At the B3LYP/6-31G** level, intermediates 2 were 66.56 kJ/mol lower in energy than 3. It was shown that the transition states 4 and 5 involving an alpha-COOH or gamma-COOH group had energy barriers of DeltaE = 57.59 kJ mol(-1) and 120.93 kJ mol(-1), respectively. The theoretical calculations suggested that the (alpha-COOH group could be differentiated from the gamma-COOH intramolecularly in amino acids by N-phosphorylation. These might be helpful to understand why only alpha-COOH, not gamma-COOH, was involved in the biomimic reactions.

Abstract:Aminoacyl adenylate pentacoordinated phosphorus compounds were analyzed by field desorption (FD) and fast atom bombardment (FAB) mass spectrometries, together with the B/E linked scan technique, and their mass spectral fragmentation pathways were investigated. For the five bonds (one P-N bond, three P-O bonds and one mixed anhydride bond P-O-CO), the cleavage usually occurred more on the P-N bond, the mixed anhydride bond and the O-C bond adjacent to the P-O bond, and less on the P-O bond. Ion YH+, corresponding to water loss from protonated 2',3'-O-isopropylidene-adenosine, was the base peak. The results reflect the structural characteristics of aminoacyl adenylate pentacoordinated phosphorus compounds. Copyright (C) 1999 John Wiley & Sons, Ltd.

Abstract:On account of the broad utilities of organophosphorus compounds, the development of highly efficient and concise phosphination methods is significantly important and urgent. Herein, we disclose a novel method for the synthesis of phosphorylated heterocycles: versatile intermediate propargylamines serving as a new type of radical acceptors incorporated in P-radicals via a photocatalytic strategy. This reaction proceeds through a cascade phosphinoylation/ cyclization/oxidation/aromatization pathway using readily available starting materials under mild conditions of light with excellent atom economy, catalyzed by AgOAc or fac-Ir(ppy)(3). One of the phosphorylated quinolines was selected, as an example, as an electron-transporting material for fabricating phosphorescence organic light-emitting diodes displaying excellent electroluminescence performances with a maximum external quantum efficiency of 21.9% with negligible efficiency roll-off ratios.

Abstract:Biological phosphates can coordinate metal ions and their complexes are common in living systems. Dynamics of mutual oxygen atom exchange in the tetrahedral group in complexes has not been investigated. Here, we present a direct experimental proof of the exchange ("phosphonate rotation") in model Ln(III) complexes of monophosphonate H4dota analogue which alters phosphorus atom chirality of coordinated phosphonate monoester. Combination of macrocycle‐based isomerism with P‐based chirality leads to several diastereoisomers. The (non)‐coordinated oxygens were distinguished through 17O‐labelled phosphonate group and their mutual exchange was followed by various NMR techniques and DFT calculations. The process is sterically demanding and occurs through bulky bidentate (k2‐PO2)– coordination. It was observed on complexes of large Ln(III) ions in twisted‐square antiprism diastereoisomers. The process energy increases for smaller Ln(III) ions (298ΔG‡(exp./DFT) = 51.8/52.1 and 61.0 / 71.5 kJ mol−1 for La(III) and Eu(III), respectively). These results are helpful in design of such complexes for MRI and protein paramagnetic NMR probes. It demonstrates usefulness of 17O NMR to study solution dynamics in complexes involving phosphorus acid derivatives. The results may inspire use of this method to study dynamics of phosphoric acid derivatives (as e.g. phosphorus acid‐based inhibitors of metalloenzymes) in different areas of chemistry.

Abstract:The mechanism of phosphoserine's bio-mimic reaction involving the formation of hexa-coordinate intermediate was studied by MNDO method. From literature, the intramolecular carboxylic-phosphoric mixed anhydride 2 could be formed from starting material phosphoserine 1 by a non-synchronous concerted reaction pathway and the transition state energies were 172 kJ/mol. In this paper, it was showed that the penta-coordinate phosphorus 2 could in turn be converted into hexa-coordinate phosphorus 3 by two steps. In the first step, the acidic proton in the intermediate 2 dissociated from P-O-H bond to form 2', in which there is a weak hydrogen bond between O1 on phosphorus and the hydroxyl group on the serine side chain. Then the hydrogen of the hydroxyl group could be transfered to O1 on phosphorus through a hydrogen-bond-bridged transition state, In the second step, the oxygen O6 anion on the serine side chain could attack the phosphorus and the molecule geometry was changed into octahedral structure of hexa-coordinate phosphorus intermediate 3, The hydrogen-bond-bridge structure lowered the transition state energy to 148. 5 kJ/mol. These results could be used to explain the formation of hexa-coordinate phosphorus intermediate and the co-participation of the carboxyl group and hydroxyl group of serine side chain during its bio-mimic reaction.

Abstract:Interaction of phosphate with the surfaces of clays and metal oxyhydroxides is important for nutrient cycling in natural and agricultural systems. We examined the specific adsorption of phosphate to boehmite (gamma-AlOOH) by solid-state P-31 NMR spectroscopy, which yields evidence for the presence of two bridging bidentate surface complexes differing in protonation. For samples prepared along the sorption isotherm at pH 5, distinct phosphate environments are observed as two major peaks in P-31 NMR spectra (chemical shifts of 0 and -6 ppm) that show little change in relative intensity with adsorbate loading. Both peaks correspond to rigid phosphate in close proximity to H, as indicated by P-31{H-1} cross-polarization magic-angle-spinning (CP/MAS) data, and yield nearly identical P-31{Al-27} dephasing curves in rotational echo adiabatic passage double resonance (REAPDOR) experiments. The REAPDOR results indicate that both phosphate environments have similar coordination to Al and are best fit by dephasing curves simulated for bridging bidentate configurations. The two resolved phosphate species exhibit distinct P-31 chemical shift anisotropy (CSA) and intensity variations with pH. the peak near 0 ppm being dominant at pH > 7. P-31 CSA's from quantum chemical calculations of hydrated bidentate cluster models with varying protonation state show that the CSA for monoprotonated phosphate is unique and closely matches that for the peak at -6 ppm. The CSA for the peak at 0 ppm is consistent with both di- and nonprotonated phosphate, but assignment to the latter is suggested based on the dominance of this peak in samples prepared at high pH and with trends in P-31 NMR chemical shifts.

Abstract:N-phosphorylamino acids are chemically active species that have many biomimic activities. alpha-COOH in amino acids and peptides behaviors rather differently than beta-COOH in many biochemical processes and takes a more important role in the origin of life. Activity differences between alpha-COOH and beta-COOH in the peptide formation of phosphoryl amino acids are studied by 1D, 2D NMR techniques and by ab initio and density functional theory (DFT) calculations in this paper. Phosphoryl dipeptide is formed directly from phosphoryl aspartic acids without any coupling reagents. Only the alpha-dipeptide ester is observed by 1D H-1, C-13, and P-31? NMR and 2D NMR. In the ab initio and DFT calculations, the pentacoordinate phosphorane intermediates containing five-membered rings are predicted to be more favored than those with six-membered rings. Both the experimental results and the theoretical calculations suggest that only the alpha-COOH group is activated by N-phosphorylation in N-phosphorylaspartic acid under mild conditions.
