Abstract:An efficient and general method for the highly stereoselective transformation of (1S,3S)-cis-1,3-disubstituted 1,2,3,4-tetrahydro-β-carbolines (THBCs) into (1S,3R)-trans-1,3-disubstituted THBCs is described. The method contains the following three steps: the enantiomerically pure (1S,3S)-cis-1,3-disubstituted THBCs 1 were first converted into (1S,3S)-cis-1,2,3-trisubstituted THBCs 2 by N-1-naphthylmethylation/benzylation; (1S,3S)-cis-1,2,3-trisubstituted THBCs2 were then converted into (1S,3R)-trans-1,2,3-trisubstituted THBCs 3 in high yields and with high stereoselectivities via a base-catalyzed epimerization at C-3; (1S,3R)-trans-1,2,3-trisubstituted THBCs 3 were subsequently converted into (1S,3R)-trans-1,3-disubstituted THBCs 4 after reductive removal of the 1-naphthylmethyl/benzyl group. In addition, as an application of this method, an improved and highly stereoselective synthesis of the PDE5 inhibitor tadalafil (Cialis®) starting from natural and less expensive l-tryptophan was developed.

Abstract:The first synthesis of tadalafil 1 (Cialis) from l-tryptophan is described. The title compound 1 was synthesized via seven steps from l-tryptophan methyl ester hydrochloride in 42.3% overall yield. Two characteristic steps involved in this synthesis are the base-catalyzed epimerization of the C-3 position of (1S,3S)-1,2,3-trisubstituted-tetrahydro-β-carboline 3a and the acid-catalyzed epimerization of the C-1 position of (1S,3R)-1,3-disubstituted-tetrahydro-β-carboline 5. The (S)-configurations at C-1 and C-3 were inverted to (R)-configurations during the epimerization reactions. The base-catalyzed epimerization of C-3 of (1S,3S)-1,2,3-trisubstituted-tetrahydro-β-carbolines 3a–3e was also studied in detail.

Abstract:The acid-catalyzed Pictet–Spengler reaction of d-tryptophan methyl ester with piperonal in acetic acid has been reported, the best stereoselectivity (cis/trans=92:8) was obtained with benzoic acid as the catalyst. The Pictet–Spengler reaction of d-tryptophan methyl ester hydrochloride with piperonal in various solvents has been extensively studied, the solvent-dependence of stereoselectivities could be principally attributed to the solubility-difference between cis and trans products 5-HCl in the used solvent, the best stereoselectivity (cis/trans=99:1) was obtained using nitromethane or acetonitrile as the solvent. A base-catalyzed epimerization at 12a-position of Cialis 1 (tadalafil) in a DMSO-containing solvent was also exploited. Cialis, 12a-epi-Cialis 2, deuterium-labeled 3,3,12a-d3-Cialis 3, and 3,3,12a-d3-12a-epi-Cialis 4 were efficiently synthesized from d-tryptophan methyl ester hydrochloride.

Abstract:An efficient one-pot multistep strategy has been developed, comprising auto-oxidative difunctionalization of alkenes, oxidation of sulfides, and a further reduction of peroxides for the synthesis of complex beta-hydroxysulfone derivatives from phenthiols and alkenes. This method has several advantageous characteristics, including readily available substrates, low-cost and environmental benign reagents, nontoxic and renewable solvents, and mild reaction conditions. The application of this transformation to the multigram-scale preparation of the anticancer drug bicalutamide is accomplished.

Abstract:A facile one-pot synthesis of 2,5-disubstituted-1,3,4-thiadiazoles was achieved by ultrasonic irradiation and phase transfer catalysis for the first time and eight novel 2,5-disubstituted-1,3,4-thiadiazoles were synthesized. This method has the advantages of mild conditions, short time, good yields, convenience, economy and environmental friendliness. The structures of the target molecules were characterized by IR, H-1 NMR,C-13 NMR, 1D NOE, MS spectra and elemental analysis. The experiments of the biological activity for the target molecules were accomplished. The experimental results of the inhibiting activity against COX-2 indicated that the compound 5f had the highest inhibition rate, up to 95.59%, and yet the compound 5g exhibited weakly inhibiting activity. The target compounds did not possess anticonvulasional activity.

Abstract:A novel phosphodiesterase type-5 inhibitor, Tadalafil in a total yield of 64%, was synthesized from D-tryptophan by Pictet-Spengler condensation, N-acylation reaction and cyclization. Separation of cis-and trans-product of Pictet-Spengler condensation was accomplished by their difference of solubility in isopropanol to avoid resolution of diastereoisomer.

Abstract:A series of 2-amino-4-aryl-5-oxo-5,6,7,8-tetrahydro-4H-chromene-3-carbonitrile derivatives (4a similar to 4g) were prepared by using 1,3-cyclohexanedione, aromatic aldehydes and malononitrile in anhydrous alcohol under microwave irradiation without catalyst. Compared with conventional heating method, the method of microwave irradiation possesses advantages including short time, high yield and environmental friendliness, etc. The structures of these compounds were characterized by IR, (1)H NMR spectra and elemental analysis.

Abstract:There are two methods in the literature for the preparation of 12-oxo-1. 15-pentadecanolide(1) from α-ni-trocyclododecanone: four-step reaction sequence and four-step one pot reaction sequence. The total yield of the latter is higher than the former. The reason for lower total yield of the four-step reaction sequence was studied. On this basis , 1 was synthesized by four-step two pot reaction sequence from α-nitrocyclododecanone in total yield of 77%.

Abstract:Oligostilbenoids represented a family of natural products, which contained one or several multifused benzofuran substructures and displayed promising biological activities towards cancer as well as immunological therapeutic targets. A convergent-divergent strategy featuring Rh-catalyzed carboacylation/aromatization cascade reaction based on C-C activation of benzocyclbutenones had been conceived. penta-Methyl amurensin H and diptoindolnesin G were successfully synthesized without any protecting groups, constituting a new entry towards oligostilbenoids' natural product synthesis. The synthesis completed within 10 steps for both natural products, suggesting conciseness and efficacy of the C-C activation in complex natural product synthesis. (C) 2019 Elsevier Ltd. Abstract:Owing to the massive importance of dihydropyrimidine (DHPMs) scaffolds in the pharmaceutical industry and other areas, we developed an effective and sustainable one-pot reaction protocol for the synthesis of ( R / S )-2-thioxo-DHPM-5-carboxanilides via the Biginelli-type cyclo-condensation reaction of aryl aldehydes, thiourea and various acetoacetanilide derivatives in ethanol at 100 °C. In this protocol, taurine was used as a green and reusable bio-organic catalyst. Twenty-three novel derivatives of ( R / S )-TDHPM-5-carboxanilides and their structures were confirmed by various spectroscopy techniques. The aforementioned compounds were synthesized via the formation of one asymmetric centre, one new C–C bond, and two new C–N bonds in the final product. All the newly synthesized compounds were obtained in their racemic form with up to 99% yield. In addition, the separation of the racemic mixture of all the newly synthesized compounds was carried out by chiral HPLC (Prep LC), which provided up to 99.99% purity. The absolute configuration of all the enantiomerically pure isomers was determined using a circular dichroism study and validated by a computational approach. With up to 99% yield of 4d , this one-pot synthetic approach can also be useful for large-scale industrial production. One of the separated isomers (4 R )-(+)-4 S developed as a single crystal, and it was found that this crystal structure was orthorhombic.

Abstract:Multi-component condensation of 2-naphthol, aromatic aldehydes, and cyclic 1,3-dicarbonyl compounds catalyzed by ionic liquid [NSPTEA][HSO 4 ] was accomplished for the synthesis of a series of 12-aryl-8,9,10,12-tetrahydrobenzo[a]xanthen-11-ones under solvent-free conditions. High yields, ease recovery, short reaction time and reusability of catalyst are significant advantages. ZrOCl 2 ·8H 2 O was also found to act as an effective catalyst towards this transformation.
