Effect of T-2 toxin on serum levels of IL-6,IL-1β and TNF-α in rat Kaschin-Beck Disease model at selenium deficiency conditions

Abstract:Objective Assaying the concentrations of IL-6,IL-1β and TNF-α in serum from rat with KBD in order to investigate the effects of T-2 toxin at the low selenium conditions.Methods All Sprague-Dawley(SD) rats were randomly divided into two groups and keeping for one month.When the low-selenium status of rats were determined,they were randomly divided into six groups,control group included.The levels of supernatant IL-6,IL-1β and TNF-α in serum were detemined by ELISA.Results The levels of IL-6,IL-1β and TNF-α in serum of KBD group were significantly higher than those in the control subjects.Serum IL-6 levels from low selenium and T-2 toxin group were increased almost five-fold when compared to that of the normal rats from a control group.In addition,Serum IL-1β and TNF-α levels were also increased one or two-fold times.There exists a remarkable significance(P<0.01) in IL-6,IL-1β and TNF-α levels compared to the normal rats.IL-6,IL-1β and TNF-α levels in the low-selenium group and the conventional +low T-2 group,there was no difference(P>0.05);there is no difference between the two groups of conventional +high/low T-2 groups(P>0.05);the two groups of low-selenium +high / low T-2 were significantly different(P<0.01);low-selenium + high/low groups compared with other intervention groups were significantly different(P<0.01).Conclusions IL-6,IL-1β and TNF-α play important roles in the development and progression of pathogenesis in KBD rats.The high T-2 toxin toxicity was stronger.Low selenium is necessary in increased concentration of IL-6,IL-1β and TNF-α.T-2 toxin is associated with presence and severity of Kaschin-Beck disease,and there was a dose-dependent relationship with T-2 toxin.Selenium has some protective effects in high or low T-2 toxin group but it does not inhibit T-2 toxin on the role of the development of the disease,especially the high T-2 toxin had more serious effects in the acute phase of low selenium.
