Abstract:Hybrid rice production includes two main processes: multiplication of male-sterile line and production of hybrid seed. During these processes, certain factors may make the male-sterile line fertile and decrease the purity of hybrid seed. Additional problems include contamination with fertile seed originating from pollination with exotic pollen or from physical mixture of seed. Therefore, developing a rapid, economic and convenient system for ensuring seed purity is a prerequisite for hybrid seed production. To achieve this objective, novel kinds of cytoplasmic male sterile (CMS) line, temperature-sensitive genic male sterile (TGMS) line and temperature/photoperiod-sensitive genic male (T/PGMS) line were developed with green-revertible leaf color marker. Male sterile, lines of all three types displayed the following major characteristics: a) Leaf color marker could be easily distinguished from normal green plants, but was only expressed at the seedling stage; b) They were insensitive to temperature with regard to the marker phenotype and expressed the marker unchanged under various environmental and growing conditions:, c) A single recessive gene controlled the marker trait; d) They maintained sterility characteristics as their corresponding parent did. e) Expression of the marker trait not only did not have negative effects on major agronomic trait of malesterile line itself, but also did not affect the hybrid F-1 plants derived from these lines. Practical application demonstrated that these lines could be used as an ideal screening marker for eliminating and measuring off-types among male-sterile line and hybrid rice plants.
