Abstract:A green and fully enzymatic procedure for the resolution of chiral alcohols through lipase/esterase-catalyzed acylation and subsequent lipase-catalyzed aminolysis using anhydrous ammonia was demonstrated. Both enantiomers can be obtained in high ee values (up to >99%) under ambient reaction conditions. The solvent and acyl donors can be recycled, and the enzyme can be reused for up to five times.

Abstract:Metal-catalyzed enantioselective allylation, which involves the substitution of allylic metal intermediates with a diverse range of different nucleophiles or S(N)2'-type allylic substitution, leads to the formation of C-H, -C, -O, -N, -S, and other bonds with very high levels of asymmetric induction. The reaction may tolerate a broad range of functional groups and has been applied successfully to the synthesis of many natural products and new chiral compounds.

Abstract:Highly stereo- and enantioselective synthesis of (E)-δ-hydroxymethyl-anti-homoallylic alcohols is reported. This reaction provides synthetically useful homoallyl alcohols in high yield with good to high enantioselectivity. Abstract:Aminoalcohols are expressed in natural products, blockbuster pharmaceuticals, agrochemicals, and key building blocks for innumerable society-benefitting applications. Historically their synthesis relies on polar-bond retrosynthetic analysis and often suffers from extensive protecting/functional group manipulation chemistry that hinders their direct access. Here we show how a simple serine-derived chemical cassette can be leveraged in a series of electrocatalytic transformations to dramatically simplify the way such structures can be made enantio- and chemoselectively in a modular fashion. This chemistry can be used to rapidly create libraries of useful building blocks and the radical retrosynthetic logic employed can be applied to truncate the synthesis of known structures. This methodology can be used on large scale as well as demonstrated with a unique flow reactor setup.

Abstract:The asymmetric synthesis of multisubstituted allylic amino acid derivatives was accomplished by the allylic alkylation of a chiral glycine-based nickel complex with vinylethylene carbonates. High enantioselectivities and diastereoselectivities were obtained under mild reaction conditions. The gram-scale synthesis was carried out with a good yield and high enantioselectivity, indicating that the method is a highly efficient route to chiral multisubstituted allylic amino acid derivatives.

Abstract:An efficient three-step method has been developed for enantioselective synthesis of chiral alpha-aminoxy acids from aldehydes. In this method, chiral propargylic alcohols are obtained by asymmetric addition of terminal alkynes to aldehydes. The hydroxyl group then converted to aminoxy group via Mitsunobu reaction and oxidative cleavage of the internal carbon-carbon triple bond produce the carboxylic acid group. This represents a convenient approach to the general asymmetric synthesis of alpha-aminoxy acids with the advantages of substantial overall yields (37-73%) and high enantioselectivities (81-99% ee).

Abstract:A chiral aldehyde/palladium catalysis-enabled asymmetric α-allylation of NH(2)-unprotected amino acid esters with 1,3-disubstituted allyl acetates is described in this work. With the utilization of different chiral phosphine ligands, both the anti- and syn-selective allylation reactions are achieved enantioselectively. A series of α,α-disubstituted amino acid esters bearing two adjacent chiral centers are produced in moderate-to-excellent yields, diastereoselectivities, and enantioselectivities.

Abstract:A practical catalytic method for enantioselective addition of an allene unit to aldimines is disclosed. Transformations are promoted by an in-situ-generated B-based catalyst that is derived from a simple, robust, and readily accessible (in multigram quantities) chiral aminoalcohol. A range of aryl-, heteroaryl-, and alkyl-substituted homoallenylamides can be obtained in 66-91% yield and 84:16 to >99:1 enantiomeric ratio through reactions performed at ambient temperature and in the presence of 0.1-3.0 mol% of the chiral catalyst and a commercially available allenylboron reagent. The catalytic protocol does not require strict anhydrous conditions, can be performed on gram scale, and promotes highly selective addition of an allenyl unit (vs a propargyl group). The utility of the approach is demonstrated through development of succinct approaches to syntheses of anisomycin and epi-cytoxazone.

Abstract:We report herein the development of α-borylmethyl-(Z)-crotylboronate reagent and the application in highly stereo- and enantioselective syntheses of (E)-δ-hydroxymethyl-syn-homoallylic alcohols. Starting from 1,4-pentadiene, α-borylmethyl-(Z)-crotylboronate was synthesized in two steps with high Z-selectivity and enantioselectivity. Subsequent aldehyde allylboration with the developed boron reagent gave highly enantioenriched (E)-δ-hydroxymethyl-syn-homoallylic alcohols upon oxidative workup.

Abstract:A general and highly efficient method for asymmetric sequential hydrogenation of alpha,beta-unsaturated ketones has been developed by using an iridium/f-Ampha complex as the catalyst, furnishing corresponding chiral alcohols with two contiguous stereocenters in high yields with excellent diastereo- and enantioselectivities (up to 99% yield, >20 : 1 dr and >99% ee). Control experiments indicated that the C=C and C=O bonds of the enones were hydrogenated sequentially, and the final stereoselectivities were determined by the dynamic kinetic resolution of ketones. Moreover, DFT calculations revealed that an outer sphere pathway was involved in both reduction of C=C and C=O bonds of enones. The synthetic utility of this method was demonstrated by a gram-scale reaction with very low catalyst loading (S/C = 20 000) and a concise synthetic route to key chiral intermediates of the antiasthmatic drug CP-199,330.

Abstract:A palladium-catalyzed asymmetric decarboxylative allylation of azlactones with 5-vinyloxazolidine-2,4-diones as allylpalladium precursors is developed, which provides a facile access to diverse (Z)-trisubstituted unnatural allylic amino acid derivatives in high yields with excellent enantioselectivities, exclusive Z-selectivities and regioselectivities towards linear products. Significantly, this method was proven to be workable in the scale-up synthesis of (Z)-trisubstituted chiral unnatural allylic amino acid derivatives, which were further transformed into ring-opening products while retaining the enantioselectivity and geometric control.

Abstract:The development of catalytic enantioselective aldol reactions has been at the forefront of advancements in contemporary asymmetric methodology. Despite significant progress, the catalytic enantioselective synthesis of certain synthetically important motifs such as the ubiquitous aliphatic syn-diol remains a challenge. Herein, we report the application of a benzyloxy-functionalized malonic acid half thioester as an activated ester equivalent in a highly enantioselective glycolate aldol reaction. This robust method operates at ambient temperature, tolerates air and moisture, and generates CO2 as the only by-product. The synthetic applicability of the method is demonstrated by the large-scale enantiodivergent synthesis of -benzyloxy-β-hydroxybutyric acid thioester and its subsequent conversion to diverse polyoxygenated building blocks, deoxy-sugars, and the natural product (–)-angiopterlactone B.

Abstract:(S)-(−)-2-Amino-1,1,3-triphenylpropanol 3c, which is readily available from l-Phe, was found to be effective in catalyzing the addition reaction of an alkynylzinc reagent to aromatic ketones with up to 80% ee of the thus produced chiral tertiary propargylic alcohols. Unlike previous reports, 3c does not require the use of an additional central metal (other than zinc itself) and the pre-preparation of an alkynylzinc. This has greatly simplified the experimental procedure for this reaction.
