Overexpression of Na~+/H~+ of Antiporter (Nhap) Gene Improves Salt Tolerance in Tobacco

Abstract:The Nhap gene was isolated from Synechocystis pcc6803, encoded plasma Na+/H+ antiporter protein, and has an important value in the field of salt tolerance research. In the study, Nhap gene was transferred into tobacco (Nicotiana tobacum ) by Agrobacterium tumefacience-mediated transformation. The results of PCR and RT-PCR detection showed that the transgenic tobaccos were obtained. At the same time, the seed germination, survival rate, chlorophyll content, Na+ content and the salt tolerant character of F1 generation of transgenic tobacco were determined under different NaCl concentrations. The results demonstrated that transgenic tobacco' s seed germination, survivable rate and chlorophyll content were higher than that of control tobacco; Na+ content was decline to the contrary. And transgenic tobacco could normally grow under the treatment of 200 mmol/L NaCl, while the growth of control tobacco was seriously inhibited. From these results, it concluded that Na+/H+ antiporter gene confers tobacco resistant against salt stress.
