New Understanding of Mechanisms of Action of 5-Aminosalicylic Acid in Inflammatory Bowel Disease and its Relevant Colorectal Cancer

Abstract:Inflammatory bowel disease (IBD) is classically subdivided into ulcerative colitis (UC) and Crohn’s disease (CD). 5-aminosalicylic acid (5-ASA) is one of the classical anti-inflammatory agents used today for the treatment of IBD. The patients with IBD have a higher risk of developing colorectal cancer, and the long-term use of 5-ASA may significantly reduce the risk of development of colorectal cancer. The conventional hypothesis of anti-inflammatory action of 5-ASA includes modulation of inflammatory cytokine production and inhibition of the biosynthesis of prostaglandins and cytokines. Recent studies showed that 5-ASA may have anti-inflammatory and anti-tumor effects via peroxisome proliferation-activated receptor (PPAR)-γ pathway, this article reviewed the understanding of new mechanisms of action of 5-ASA.
