Effect of "freezing-cultivation" cycle on compositions of membrane fatty acids of Lactobacillus plantarum

Abstract:"Freezing-cultivation" cycle treatment is applied to the cold-sensitive strain of Lactobacillus plantarum.The objective of this paper is to investigate the influence of cold stimulation on the membrane fatty acids compositions and the survival rate of this strain.The membrane fatty acids were analyzed with GC-MS,and the survival rate was evaluated by standard plate count method.Firstly,L.plantarum B1 was most sensitive strain among four lactobacilli strains after these strains were frozen at-20 ℃ for 24 h according to standard plate count.Secondly,the unsaturated/saturated fatty acids(U/S) ratio of the sensitive strain of L.plantarum B1 was increased by 60.391% after 10 times of "freezing-cultivation" cycle.And the percentage of C16:0 and C18:0 was decreased,but that of C18:2 was increased.After four cycles of freezing-thawing,the viable cells number of L.plantarum B1 treated by "freezing-cultivation" cycle was increased by one order of magnitude compared with that of the original cells.The treatment of "freezing-cultivation" cycle can change the compositions of the membrane fatty acids.It can be suggested that the resistance improvement of survival cells treated by "freezing-cultivation" cycle to freezing-thawing is related to the raising of U/S ratio.
