Abstract:An array of ultra-long-chain fatty acid sugar alcohol monoesters, with behenic acid as an acyl representative and sugar alcohols altered from ethylene glycol to glycerol, erythritol, pentaerythritol, arabitol, xylitol and sorbitol, were enzymatically synthesized in high purity and selectivity. The molecular structures of the synthetic compounds were confirmed by H-1 NMR, FT-IR and MS analysis, and the thermal properties were primarily characterized by DSC analysis. The molecular packing and thermal properties of synthetic sugar alcohol monobehenates (SAMBs) were investigated by Temp-Ramp-FT-IR. For in vivo application purposes, the enzymatic lipolysis of synthesized SAMBs was examined by a PPL (porcine pancreatic lipase)-mediated in vitro digestion test, and improved resistance of most SAMBs to enzymatic lipolysis, in comparison to glycerol monopalmitate, was observed. FT-IR spectroscopic analysis of the thermotropic phase transitions of the synthetic SAMBs indicated that the thermal collapse temperatures do not vary significantly as the polar head alters, suggesting their thermostabilities are largely governed by hydrophobic interactions among the alkanyl chains, while the size, properties and volume of the polar heads may determine the packing patterns. Systemic mapping of the structure-property-function relationship of SAMBs revealed the potential of these compounds for multipurpose applications. Ethylene glycol and glycerol monobehenates enable orthorhombic packing, and could find applications in cosmetic formulation, whereas sorbitol monobehenate is capable of forming stable surfactant-free nanoparticles, which could be excellent excipients for solid lipid nanoparticles for use as delivery cargo for drugs and food ingredients.

Abstract:There has also been a growing interest in the application of carbohydrates as pharmaceutical drugs in the field of invasive bacterial diseases, inflammatory diseases, and viral infections. Chemical oligosaccharide synthetic procedures often require tedious protection and deprotection steps. Enzymatic synthesis offers tremendous benefits because of its high regio- and stereoselectivities, stability, organic solvent compatibility, and low cost. Glycosyltransferase-catalyzed synthesis of carbohydrates has been recognized as one of the most practical approaches for its high efficiency and specificity under very mild conditions. The two main obstacles in the enzymatic synthesis of carbohydrates are the inadequate availability of glycosyltransferases and the high cost of sugar nucleotides, which serve as intermediates. Significant progress in the study of the enzymatic biosynthesis of complex carbohydrates has been made with developments in protein purification, molecular genetics, and new methods of enzymological analysis. Bioinformatics provide a large amount of putative candidates for carbohydrate active enzymes. The combined enzymatic and genetic approach has overcome formidable obstacles ubiquitous to the study of glycans, and has begun to yield new information that definitively addresses basic enzymological issues relevant to these complex carbohydrates. In fact, complex carbohydrates representing the glycan chain of gangliosides such as GT2, GM1, and GD1a were synthesized using glycosyltransferases. Although traditional cell-free enzymatic synthesis of oligosaccharides will continue to be used in laboratory scale synthesis, the future direction for large-scale preparation of complex oligosaccharides and polysaccharides will be whole-cell systems. Further development of these enzymatic methods will allow synthetic biochemists to create important molecular tools for biochemical, biophysical, and medical applications.

Abstract:Acid hydrolysis and enzymatic hydrolysis are the main methods for releasing glycosidically bound volatiles (GBV). However, acid hydrolysis yields a strong pungent odor, and enzymatic hydrolysis is time consuming. In the present study, a new method, ultrasound hydrolysis, is reported to release GBV. This method is simple, environmentally friendly, fast and effective. Large differences were observed in the released aglycones and glycosyls between ultrasound and enzymatic hydrolysis of GBV. More types of aglycones were released under ultrasound than enzymatic hydrolysis. Alcohols and esters were the main aglycones under enzymatic hydrolysis, and terpenoids, esters and aldehydes were the main aglycones under ultrasound hydrolysis. The glycosyls released under ultrasound hydrolysis were mannose, glucose and sucrose, and those released under enzymatic hydrolysis were galactose and sucrose. The present study gives a new insight into a hydrolytic method for GBV by using ultrasound hydrolysis and can provide a reference method for fruit juice aromatization.
A Novel Procedure for the Synthesis of 1,2:5,6-Di-o-isopropylidene-d-glucofranose by Ultrasound

Abstract:In the paper, synthesis of 1,2:5,6-di-O-isopropylidene-D-glucofranose (DAG) assisted by ultrasound were performed in the presence of ferric chloride as catalyst. The effects of some reaction parameters (e.g. power and frequency of ultrasound, reaction temperature, mass ratio of the reactant) on the yield were investigated. It was found that the reaction is greatly accelerated by the ultrasound in comparison to the control runs under mechanical stirring. The optimal ultrasound output power and frequency were found to be 200 W and 24 kHz at 40 °C with a 1:30 mass ratio of glucose to acetone. Under these optimal conditions, the yield of DAG reached 75.8%.

Abstract:Ultrasound application is a method of enhancing the performance of biocatalysts. Enzymes performance is strongly influenced by the nature of the reaction medium. Ultrasounds have the potential to significantly influence the activity of the enzymatic processes, provided that the energy input is not too high to inactivate the enzyme. In the present paper a systematic study on ultrasound-assisted enzymatic esterification for aroma esters preparation is described. Thus, by ultrasound assisted enzymatic esterification i-amyl acetate was obtained. The method is efficient, mild, and environmentally benign. Significant improvements were obtained in comparison to conventional method. The results show a favourable perspective of the ultrasound technique to improve the process efficiency and reduce the reaction time. The commercial aroma esters synthesis will be potentially realized due to this ultrasound-promoted esters synthesis method.

Abstract:Enzymatic hydrolysis of starch is an important process in the food industry. In the present work, ultrasound was introduced in glucoamylase pretreatment, starch pretreatment and mixed reaction system treatment to enhance starch hydrolysis efficiency. These different processes were studied to explore the mechanism of ultrasound in promoting enzymatic reactions. The hydrolysis degree of starch was determined via measuring the reducing sugar yield. Ultrasound caused enzyme inactivation under high temperatures, high ultrasonic power and longtime treatment, especially at high temperatures exceeding 65 degrees C. Ultrasound pretreatment of starch before enzymolysis led to the furtherance of starch hydrolysis degree. Meanwhile, sonicating the mixed enzymatic reaction system below 65 degrees C promoted starch hydrolysis significantly, inducing more than five- fold growth in the degree of starch hydrolysis as much as the ultrasound pretreatment caused. Molecular weights analysis conducted by the MALLS system reflected the enormous damage of starch molecules caused by ultrasound. The amylose contents and chain length distributions of samples were separately analyzed by iodine binding method and size exclusion chromatography. The results of the two experiments illustrate that ultrasound could promote the enzymatic hydrolysis of amylopectin, which is harder for glucoamylase to hydrolyze compared to amylose. Abstract:The present work investigates the synergistic impact of glucoamylase and ultrasound on starch hydrolysis. The extent of starch hydrolysis at different reaction parameters (ultrasonic intensity, temperature, reaction time) was analyzed. The hydrolysis extent increased with the reaction time and reached a maximum value under ultrasonic intensity of 7.20 W/mL at 10 min. Ultrasound did not alter the optimum enzymatic temperature but speeded up the thermal inactivation of glucoamylase. The evaluation of enzymatic kinetics and starch degradation kinetics indicated a promotion of the reaction rate and enzyme-substrate affinity. According to the thermodynamic results, sonoenzymolysis reactions require less energy than enzymolysis reactions. The measurement of molecular weight, solubility, thermal properties, and structures of the substrates revealed that sonoenzymolysis reaction generated greater impacts on starch properties. The molecular weight and radii of gyration decreased by 80.19% and 90.05% respectively while the starch solubility improved by 136.50%. (C) 2017 Published by Elsevier Ltd.

Abstract:Abstract This work is focused on the study of the esterification parameters for the ultrasound assisted synthesis of isoamyl acetate catalyzed by lipase Lipozyme 435 in a continuous loop reactor. Investigating the influence of different parameters shows that a higher concentration of ester (462 mg/g mixture) can be obtained at a temperature of 50 °C, flow rate 0.16 mL/min. The best ultrasonication conditions are: sonication applied continuously for a short time (20 min), ultrasound power 32 mW and amplitude 20%. The enzyme can be successfully reused tree times without loss of enzyme activity. Reaction kinetics for isoamyl acetate ultrasound assisted production showed that satisfactory reaction concentration (close to the equilibrium concentrations) could be reached in short reaction times (2 h). Ultrasound assisted enzymatic esterification is consequently a cleaner and a faster process.

Abstract:Comparative studies of enzymatic acylation of troxerutin by the alkaline protease from Bacillus subtilis under ultrasound and shaking were carried out in nonaqueous media. Using divinyl dicarboxylates (CH2CH–OOC–(CH2)n–COO–CHCH2, n=2, 3, 4, 7, 8, 11) featuring different chain length as acyl donors, troxerutin was regioselectively acylated at B ethoxyl group, whether under ultrasound or shaking. Ultrasonic treatment increased the reaction rate and led to high conversion. Several factors, such as pre-irradiation on the enzyme, the power and frequency of the ultrasound, operation manner, as well as the length of the acyl donors were investigated. Using enzyme pre-irradiated for 8h, the conversion of troxerutin was increased to 87.3% compared with 56.3% obtained from the untreated enzyme. Experimental results also showed that continual ultrasound caused greater rate acceleration than interval ultrasound. Powers of 100, 150 and 200W, frequencies of 40, 80 and 100kHz all showed significant improvement on the transesterification, with the greatest effect observed at 150W, 80kHz. The acceleration effect increased as the chain length of the acyl donors decreased from C13 to C4.
Enzymatic Synthesis of Glucose-Based Fatty Acid Esters in Bisolvent Systems Containing Ionic Liquids or Deep Eutectic Solvents

Abstract:Sugar fatty acid esters (SFAEs) are biocompatible nonionic surfactants with broad applications in food, cosmetic, and pharmaceutical industries. They can be synthesized enzymatically with many advantages over their chemical synthesis. In this study, SFAE synthesis was investigated by using two reactions: (1) transesterification of glucose with fatty acid vinyl esters and (2) esterification of methyl glucoside with fatty acids, catalyzed by Lipozyme TLIM and Novozym 435 respectively. Fourteen ionic liquids (ILs) and 14 deep eutectic solvents (DESs) were screened as solvents, and the bisolvent system composed of 1-hexyl-3-methylimidazolium trifluoromethylsulfonate ([HMIm][TfO]) and 2-methyl-2-butanol (2M2B) was the best for both reactions, yielding optimal productivities (769.6 and 397.5 µmol/h/g, respectively) which are superior to those reported in the literature. Impacts of different reaction conditions were studied for both reactions. Response surface methodology (RSM) was employed to optimize the transesterification reaction. Results also demonstrated that as co-substrate, methyl glucoside yielded higher conversions than glucose, and that conversions increased with an increase in the chain length of the fatty acid moieties. DESs were poor solvents for the above reactions presumably due to their high viscosity and high polarity.
Selective and Catalyst-free Oxidation of D-Glucose to D-Glucuronic acid induced by High-Frequency Ultrasound

Abstract:This systematic experimental investigation reveals that high-frequency ultrasound irradiation (550 kHz) induced oxidation of D-glucose to glucuronic acid in excellent yield without assistance of any (bio)catalyst. Oxidation is induced thanks to the in situ production of radical species in water. Experiments show that the dissolved gases play an important role in governing the nature of generated radical species and thus the selectivity for glucuronic acid. Importantly, this process yields glucuronic acid instead of glucuronate salt typically obtained via conventional (bio)catalyst routes, which is of huge interest in respect of downstream processing. Investigations using disaccharides revealed that radicals generated by high frequency ultrasound were also capable of promoting tandem hydrolysis/oxidation reactions.

Abstract:Synthesis of ethyl acetate (EA) using lipase as a catalyst, as an environmentally-friendly process, has attracted considerable attention. However, the slow rate of enzymatic reaction and higher cost limit its applications. Ultrasound irradiation (UI) is known as an effective tool for accelerating the rate of enzymatic reaction under mild conditions. To attain maximum yield, it is crucial to understand the effect of UI on EA synthesis and the underlying mechanism. Therefore, the present study investigated the effect of UI on lipase-catalyzed ethyl acetate synthesis. The results clearly demonstrated that UI could remarkably accelerate the process of lipase-catalyzed synthesis of ethyl acetate compared with mechanical shaking. The optimal conditions of ultrasound-assisted reaction by using Novozyme 435® were 6% enzyme loading, 4:1 molar ratio of ethanol to acetic acid, 150 W & 28 kHz ultrasonic power and frequency, respectively. Bi-bi ping-pong model was employed to give a deep insight into the underlying mechanism of the beneficial effect of UI. It was found that Vmax under UI was elevated by 143% compared with that under mechanical shaking. Application of UI could decrease Kacetic acid and Ki ethanol while increase Kethanol and Ki acetic acid . In conclusion, the present study addresses that UI could remarkably accelerate the process of lipase-catalyzed synthesis of EA compared with mechanical shaking, and kinetic model results explained why ultrasound can significantly speed up lipase catalyzed synthesis of ethyl acetate, which may help to improve the industrial application potential of ethyl acetate biosynthesis.

Effect of Ultrasonic Time on the Structural and Physico-Chemical Properties of Hemicelluloses from Eucalyptus Grandis.

Abstract:Effect of sonication on the extractability and physico-chemical properties of hemicelluloses from Eucalyptus grandis using 5% KOH solution at 50 degrees C for 3 h has been comparatively studied. The results showed that the yield of hemicelluloses increased from 2.6 to 19.6% as the ultrasonic time was extended from 5 to 35 min. The highest yield of hemicelluloses (95.2%) was achieved at 30 min ultrasonic time. Xylose was the dominant sugar (82.94-84.96%) of all the hemicellulosic fractions. Furthermore, the hemicelluloses obtained by ultrasound-assisted extractions had slightly lower molecular weights (74,510-66,770 g/mol) and thermal stabilities, but higher contents of xylose (83.95-84.96%). The increased yield of ultrasonically extracted hemicelluloses, which have preserved their main structural properties, confirmed the great potential of ultrasound-assisted extraction to separate hemicelluloses from Eucalyptus grandis at an industrial level.
Effect of ultrasound on the kinetics of anti-solvent crystallization of sucrose

Abstract:The effect of ultrasound on the kinetics of anti-solvent crystallization of sucrose was studied. The influence of temperature, stirring rate, supersaturation and ultrasonic power on the anti-solvent crystallization of sucrose was investigated. The relationship between infrared spectral characteristic band of sucrose and supersaturation was determined with an online reaction analyzer. The crystal size distribution of sucrose was detected by a laser particle-size analyzer. Ultrasound accelerated the crystallization process, and had no impact on the crystal shape. Abegg, Stevens and Larson model was fitted to the experimental data, and the results were the following: At 298.15 K, the average size of crystals was 133.8 μm and nucleation rate was 4.87 × 109 m-3·s-1 without ultrasound. In an ultrasonic field, the average size was 80.5 μm, and nucleation rate was 1.18 × 1011 m-3·s-1. Ultrasound significantly reduced the average size of crystals and improved the nucleation rate. It was observed that the crystal size decreased with the increase of stirring rate in silent environment. When the stirring rate increased from 250 to 400 rpm, the average size decreased from 173.0 to 132.9 μm. However, the stirring rate had no significant impact on the crystal size in the ultrasonic field. In addition, the activation energy of anti-solvent crystallization of sucrose was decreased, and the kinetic constant of nucleation rate was increased due to the effect of ultrasound. In the ultrasonic field, the activation energy was reduced from 20422.5 to 790.5 J·mol-1, and the kinetic constant was increased from 9.76 × 102 to 8.38 × 108.

Ultrasound affects the selectivity and activity of immobilized lipases applied to fatty acid ethyl ester synthesis

Abstract:Hydrophobic carriers can be used to improve the activity, stability and other properties of enzymes. Physical agents, like ultrasound, may also contribute to improving the dispersion and collision of the reagent molecules, decreasing the reaction time and intensifying the catalytic process. However, its effect on the enzyme activity and reaction selectivity is still not entirely understood. Here, enzyme modulation of immobilized lipases was studied under pulsed ultrasound irradiation in fatty acid ethyl ester (FAEE) synthesis for biodiesel production. Novozym 435® and two commercial lipases from Thermomyces lanuginosus and Rhizomucor miehei, immobilized on Octadecyl-Sepabeads were used as a biocatalyst in the transesterification reaction of vegetable oils and ethanol. The use of ultrasound associated with catalysis by the Novozym 435 increased the production of FAEE by about three times (from 8.9 to 26.4%) using soybean oil and changes were observed in the profile of the products. From the sonicated reaction, ethyl-palmitate production decreased from 23.4 to 11.7%, while the ethyl-linoleate content rose from 47.5 to 59.2%. On the other hand, the T. lanuginosus lipase was less affected by sonication with the overall production of FAEE increasing from 17.2 to 24.1%, with ethyl-palmitate and ethyl-linoleate content changing from 16.2 to 17.5% and 55.0 to 47.8%, respectively. Although the changes in the production yield are not too high, the main idea here was to show that ultrasound modulates the lipase activity as well as its respective selectivity. Thus, ultrasound, is responsible for changing the ethyl ester production, which can be applied to many other biochemical processes to improve or modulate their synthesis yield.
Mechanistic investigations in ultrasound-assisted xylitol fermentation

Abstract:This study has investigated ultrasound-assisted xylitol production through fermentation of dilute acid (pentose-rich) hydrolysate of sugarcane bagasse using free cells of Candida tropicalis. Sonication of fermentation mixture at optimum conditions was carried out in ultrasound bath (37 kHz and 10% duty cycle). Time profiles of substrate and product in control (mechanical shaking) and test (mechanical shaking + sonication) fermentations were fitted to kinetic model using Genetic Algorithm (GA) optimization. Max. xylitol yield of 0.56 g/g and 0.61 g/g of xylose was achieved in control and test fermentations, respectively. The biomass yield also increased marginally (∼17%) with sonication. However, kinetics of fermentation increased drastically (2.5×) with sonication with 2× rise in xylose uptake and utilization by the cells. With comparative analysis of kinetic parameters in control and test experiments, this result was attributed to enhanced permeability of cell membrane that allowed faster diffusion of nutrients, substrates and products across cell membrane, higher enzyme-substrate affinity, dilution of toxic components and reduced inhibition of intracellular enzymes by substrate.

Abstract:Cellulose is the most abundant and renewable biopolymers on earth. The hydrophilic nature of cellulose endows cellulosic materials with good compatibility with polar matrices, but it also leads to their poor dispersion in non-polar matrices. Regulating the surface hydrophobicity of cellulosic materials via surface esterification can greatly extend their applications. However, the existing surface esterification methods for cellulosic materials are time-consuming and costly, which makes them less attractive for practical application. Here we found that when vinyl esters were used as esterifying agents, various alkalis can be used to catalyze the ultrafast surface esterification of cellulose materials in aqueous media at room temperature within seconds or minutes, which is highly attractive for the surface esterification of nanocellulose in suspension state. Surface acetylated cellulose nanofibers (ACNF) and surface acetylated cellulose nanocrystals (ACNC) with acetyl group content (Ac%) up to 10.7% and 6.5%, respectively, were successfully prepared within 5 min in aqueous diethylamine solution. Additionally, this method can also be used to achieve the rapid acetylation of phenol hydroxy group in aqueous media.

Abstract:This study reports an enhancement of ethanol yield in Saccharomyces cerevisiae with low-intensity ultrasonic irradiation using fixed mode frequency generated by a self-developed six-frequencies (20, 23, 25, 28, 33, 40 kHz) ultrasonic device in our group. After sonication treatment, the ethanol production potential was determined. Under the optimal conditions of ultrasonic treatment (ultrasonic frequency 28 kHz, power density 180 W/L, and treatment time 24 h), the maximum ethanol yield increased by 34.87% compared to the control. Transcriptome sequencing showed that the ultrasonic treatment had expressional regulations on genes involved in pyruvate metabolism, glycolysis, pentose phosphate pathway, glucose transport, and reducing power production. The quantitative real-time polymerase chain reaction (qRT-PCR) further confirmed the changes in gene expression (up- or down-regulation). Metabolomics revealed that ultrasonic treatments increased intracellular glucose and nicotinamide adenine dinucleotide (NADH) contents, which are key metabolites for ethanol synthesis. Besides, ultrasonic treatments decreased the acetate and its derivatives resulting in lowered reverse consumption of pyruvate and thus promoted ethanol synthesis. These changes in gene expression and metabolites content might be the main reason why the ethanol yield in Saccharomyces cerevisiae increased after ultrasonic irradiation.

Abstract:BACKGROUND Today, ultrasound is increasingly utilized in enzyme modification. Strongly dependent on the specific operational conditions, the modification effect brought by ultrasound can be activation and inactivation of enzymes. This work aims to study the ultrasound mechanisms under different conditions, to investigate the respective roles of free radical effect and mechanical effect in pectinase activation and inactivation, and to reveal the influence of pectinase concentration on the ultrasound-modification effect. RESULTS When ultrasound was introduced to a liquid system, generation of free radicals was positively correlated with ultrasound intensity and treatment duration, but negatively correlated with temperature. Thiourea with a concentration of 4 mmol L(-1)was selected as a free radical scavenger to effectively shield ultrasound free radicals. The highest enzyme activity of pectinase solutions at 0.1, 1.0, and 10.0 mg mL(-1)was obtained at the same ultrasound intensity of 4.50 W mL(-1)and time of 15 min, where the enzyme activity was increased by 68.24%, 20.98% and 18.83%, respectively. Furthermore, the addition of thiourea enhanced the enzyme activity at each tested ultrasound intensity and time, especially those exceeding the best conditions; it also eliminated the redshift phenomenon that was previously presented in the fluorescence spectra of pectinase samples. CONCLUSION Pectinase concentrations did not change the optimum ultrasound conditions for enzyme modification, but pectinase with a low concentration was more vulnerable to ultrasound treatment. During modification, ultrasound mechanical effects dominated in the pectinase activation, while free radical effects dominated in the inactivation process. (c) 2020 Society of Chemical Industry

Abstract:With the extensive application of enzyme-catalyzed reactions in numerous fields, improving enzymatic efficiency has attracted wide attention for reducing operating costs and increasing output. There are three targets throughout enzymatic reactions: the enzyme, substrate, and mixed reaction system. Ultrasound has been known to accelerate enzymatic reactions by acting on different targets. It can modify both enzyme and substrate macromolecules, which is helpful for enhancing enzyme activity and product yields. The synergistic effect of ultrasound and enzymes is widely reported to increase catalytic rates. The present review discusses the positive effect induced by ultrasound throughout the enzymatic process, including ultrasonic modification of enzymes, ultrasound assisted immobilization, ultrasonic pretreatment of substrates, and ultrasound assisted enzymatic reactions.
