Diagnostic value of detection of hypermethylation of p16 gene in peripheral lung carcinoma

Abstract:Objective To detect the hypermethylation of p16 gene in plasma DNA from patients with peripheral lung cancer and its clinical value on the diagnosis of peripheral lung cancer. Methods Using a methylation-specific polymerase chain reaction, the status of methylation of the p16 gene was investigated in plasma DNA and matched pathological tissues excised in operation from 31 cases of peripheral lung cancer and 25 cases of benign lung nodi patients. Results Hypermethylation of the p16 gene was presented in 20 cases of the plasma samples and 22 cases of the tumor tissues from 31 pateints with peripheral lung cancer but in only 3 cases of plasma and diseased tissues from 25 patients with benign lung diseases(64.5%and 70.9%vs 12.0%, P< 0.05 and 0.05, respectively). However,hypermethylation of the p16 gene in plasma and the corresponding tumor tissues weren't significantly correlated with the clinical stage and pathological type of the tumor. Conclusion It is suggested that the detection of the hypermethylation of p16 gene may be a alternative and useful technique in the diagnosis of peripheral lung carcinoma.
