Effect of allo-human bone marrow mesenchymal stem cells on the expression of IL-1, TNF-α、TGF-β1 in patients with rheumatoid arthritis

Abstract:Objective To study the effect of allo-human bone marrow mesenchymal stem cells (bMSCs) on the expression of IL-1,TNF-α and TGF-β1 in patients with rheumatoid arthritis (RA) in vitro. Methods bMSCs were isolated from bone marrow of healthy volunteers,purified by density gradient centrifugation and cultured in vitro. Mononuclear cells from peripheral blood of patients with RA were isolated,and were cocultured with bMSCs and mononuclear cells from healthy controls in vitro. The expressions of IL-1 m-RNA,TNF-α m-RNA and TGF-β1 m-RNA in the cells were detected by RT-PCR. Results After the peripheral blood mononuclear cells (PBMCs) from RA patients were co-cultured with bMSCs for 7 days,there was a reduction in expressions of IL-1 and TNF-α and an in-crease in expression of TGF-β1,with statistically significant difference as compared with those found in untreated RA PBMCs (P<0.05). Similar change was not noted in co-cultured normal PBMCs when compared with untreated normal ones (P>0.05). For TGF-β1 in both RA or normal PBMCs after 7 days' co-culture,an increased expression was found in co-culture system compared to untreated PBMCs,with statistically significant difference (both P<0.05). Conclusion Human bone marrow MSCs may have modulatory effects on main cytokines of patients with RA in vitro. bMSCs may be playing a role in pathogenesis of RA and can be useful for treatment of RA.
