Experimental Study of the Preventive Effect of Polysaccharide Sulfate on Steroid-induced Osteonecrosis

Abstract:Objective To investigate preventive effect of polysaccharide sulfate(PSS) on the steroid-induced osteonecrosis in high risk cohort.Methods Twenty adult health rabbits were divided into two groups:Animals in the model group were treated with horse serum and dexamethasone injection in a conbination way to develop the animal model of steroid-induced osteonecrosis.PSS group were treated horse serum and dexamethasone in a conbination way like the animals that in the model group,then began to receive PSS orally on the day when dexamethasone was injected.Serological tests [CHOL,TG,HDL-C,LDL-C,Lp(a),and AT-Ⅲ,TXB2,6-Keto-PGF1α,t-PA,PAI,D-dimer] were mea-sured before treatment and 24 hours or 4 weeks after the final treatment.Meanwhile,the histopathologi-cal changes of the bone samples were observed under light microscope,and the diameter of the largest fat cell intraosseous was measured,the rate of osteonecrosis occurrence in each group was calculated.The percentage of the bone area and fat area of the femoral head under light-microscope in both groups were evaluated by computer,then compared with each other to find the difference between them.Re-sults CHOL,TG,LDL-C and TXB2,PAI,D-dimer levels in the PSS group were lower than that in the animal model group,while AT-Ⅲ and t-PA levels in the PSS group were higher than that in the animal model group.On the other hand,the rate of osteonecrosis occurrence(25% vs 83.33%),the diameter of the largest fat cell(29.84 μm vs 45.54 μm) and the percentage of fat area of the femoral head(16.35% vs 30.45%) in the PSS group were lower than that in the animal model group,while the percentage of bone area(45.57% vs 36.64%) in the PSS group was higher than that in the animal model group(P< 0.05).Conclusion PSS has the effects of decreasing blood lipid,anti-coagulation and increasing the fibrinolysis.And PSS has partly a preventive effects on the steroid-induced osteonecrosis.
