Transcription Analysis and Prokaryotic Expression of Chicken Il-2 Receptor Gamma Chain Gene

Abstract:A full-length cDNA of chicken interleukin-2 receptor gamma chain(chIL-2Rγ) gene was amplified from chicken spleen mononuclear cells by RTPCR.The cDNA fragment of chIL-2Rγ gene contained an ORF of 1047bp,encoding a 37.8ku of peptide of 348 amino acids.It was predicted that the mature chIL-2Rγ peptide contained four conserved cysteine residues,one WSXWS motif and seven N-linked glycosylation.ChIL-2Rγ had low similarity to those of other animals at the amino acid level,ranging from 21.4% to 38.2%.Tissue transcription analysis indicated that chIL-2Rγ mRNA was found in cerebrum,cerebellum,medulla spinalis,bursa of fabricius,thymus,spleen,bone marrow,cecal tonsil,stomachus glandularis,stomachus muscularis,jejunum,yolk sac diverticulum,ileum,cecum,rectum,heart,kidney,lung,liver,skeletal muscle and skin,but was not detected in duodenum and testis.A prokaryotic expression vector pET28a-exchIL-2Rγ was constructed successfully and expressed,and the expressed protein was identified.
