EFFECTS OF HOMOCYSTEINE AND S-ADENOSYLHOMOCYSTEINE ON ATHEROSCLEROSIS IN APOE-DEFICIENT MICE

Abstract:Objective To explore the effects of homocysteine(Hcy) and S-adenosylhomocysteine(SAH) on atherosclerosis(AS) in ApoE-deficient mice.Method Five groups of ApoE-deficient mice were fed different diets for 8w to induce changes in their plasma Hcy and SAH concentrations.These included a control diet(control),the control diet supplemented with methionine(M+),the M+ diet deficient in B vitamins(M+B-),the M+ diet supplemented with B vitamins(M+B+) and the control diet deficient in B vitamins(B-).Results Compared with the controls,mice fed the B-diet had moderate elevation in their plasma Hcy levels.However,their plasma SAH concentration and AS lesion areas were not different.In contrast,the mice fed the M+B+ diet had larger AS lesion areas and elevated plasma SAH concentrations but their plasma Hcy concentration did not differ from that of the control group.Plasma SAH concentrations,but not the plasma Hcy levels,were consistently correlated with aortic sinus lesion areas.Conclusion The extent of AS in ApoE-deficient mice fed high dietary methionine was independent of the plasma Hcy concentration.Plasma SAH is more closely related to AS,and thus is a better biomarker of AS than Hcy.
