Using 13C-Leucine As a Tracer to Message Whole Body Protein Metabolism in Rabbits after Burn Injury

Abstract:Objective To build a rabbit hypermetabolic model by immersing partly the body of the animal into boiling-water,and study their whole body protein kinetics by using a primed constant infusion of L-1-13C-leucine a week later. Methods Twenty healthy New Zealand White rabbits were divided at random into two groups: the burn group and the control group. After 3 days of acclimatization,a full thickness burn of 15% of the body surface was established to the back of the rabbits of the burn group by immersing into a boiling-water bath;and for the control group,only anaesthetics and antibiotics were applied. A week later,left carotid artery,right jugular vein catheterization and trachea intubation were performed in each rabbit. Then a primed,continuous 2-h infusion of NaH13CO3 (10 μmol·kg-1 prime,10 μmol·kg-1·h-1 infusion) was given via the jugular vein ,and followed immediately by a primed,continuous 3-h infusion of L-1-13C-leucine (4.2 μmol·kg-1 prime,4.2 μmol·kg-1·h-1 infusion). Blood samples and expired air samples were collected after different time intervals,to determine 13C-KIC enrichment and 13CO2 enrichment by GC-MS and IRIS respectively. At last,whole body protein synthesis and breakdown rates were calculated according to given equations. Results Compared to the control group,the leucine flux and oxidation of the burn group were significantly elevated (+92.3%,+175%,respectively),indicating an accelerated proteolysis and a more intense negative body protein balance (both P0.01); however,the rate of whole body protein synthesis did not differ significantly between the two groups(P=0.15),while it tended to elevate somewhat in the burn group (4.1±1.5 g·kg-1·d-1 vs 2.7±0.9 g·kg-1·d-1). Conclusions A rabbit hypermetabolic model was successfully established,characterized by an accelerated whole body proteolysis and a negative nitrogen balance after burn injury. By using 13C-leucine as a tracer,the method used to determine whole body protein metabolism seems practical.
