In July of 2017 and 2018, a leaf spot of B. semperflorens was observed in many flower nurseries in Harbin, Heilongjiang province, China. Disease symptoms were irregular, gray, sunken leaf spots (0.2 to 2.4 cm in diameter), which expanded to blights with concentric rings, and eventually the whole leaves dried up to die. Ten diseased tissues from infected leaves of five nurseries (two plants per nursery) were surface disinfected in 0.5% NaOCl for 5 min, rinsed three times in sterile distilled water, and cultured on potato dextrose agar at 26°C in the dark for 3 days. Eight fungal cultures were isolated, and all were morphologically similar. Hyphal tips were transferred and subcultured on potato carrot agar (PCA) with a light/dark (12 h/12 h) cycle for 5 days. Colonies on PCA were olive-green with white margins and black pigments under the colony and abundant aerial hyphae. Conidia were black, brown, obclavate to obpyriform, and formed in long sparsely branched chains on simple short conidiophores that measured 4.8 to 15.7 μm in width and 7.1 to 31.8 μm in length (n = 50) and had one to four transverse and zero to three longitudinal septa. According to these morphological characteristics, the fungus was identified as Alternaria spp. (Simmons 2007). Genomic DNA was extracted from the mycelium of representative isolate QHT3. The internal transcribed spacer region (ITS), translation elongation factor 1-α gene (TEF), and glyceraldehyde°3-phosphate°dehydrogenase (GPD) were amplified and sequenced using primers ITS1/ITS4 (White et al. 1990), EF1-728F/EF1-986R (Carbone and Kohn 1999), and Gpd1/Gpd2 (Hubmann et al. 2011). The consensus sequences (GenBank accession nos. MN264615 for ITS, MN256108 for TEF, and MN977123 for GPD) were aligned using BLASTn in GenBank and were 99.7, 100, and 100% identical to Alternaria tenuissima strains ZZ9-5-1 (accession no. MN907695.1, 574/579 nucleotides), CC17 (MG013473.1, 281/268 nucleotides), and GA24 (MK451973.1, 398/620 nucleotides), respectively. To confirm pathogenicity of QHT3, B. semperflorens plants were grown in 20-cm pots containing a commercial potting mix (one plant per pot). At five- to eight-leaf stage, three plants were inoculated by spraying 15 ml of the spore suspension (4 × 106 spores/ml) over the whole plant. Three plants treated by spraying 15 ml of sterile distilled water served as a control. All plants were placed in a greenhouse (16 h/8 h light/dark, 22 to 28°C, 70% relative humidity) and covered with a plastic bag after inoculation. After 48 h, the plastic bag was removed from each plant. All inoculated leaves developed symptoms similar to those observed in the greenhouse 8 days after inoculation, whereas no symptoms were observed on the control leaves. The fungus was reisolated from the diseased inoculated leaves and identified by morphology and molecular biology. There has already been a report about A. tenuissima causing leaf necrosis on Polygonatum cyrtonema in China (Li and Guo 2020). To our knowledge, this is the first report of A. tenuissima causing leaf spot of B. semperflorens in China. The incidence rate of the disease in the nurseries is up to 70% and seriously reduced the ornamental and economic value of B. semperflorens in Harbin. Due to the importance of B. semperflorens, the distribution of A. tenuissima needs to be investigated and monitored for effective disease management strategies to be developed.The author(s) declare no conflict of interest.References:Carbone, I., and Kohn, L.
