Expression and significance of TREM-1 in neonatal rats with intrauterine infection

Abstract:Objective To investigate the expression and diagnostic value of triggering receptor expressed on myeloid cells-1(TREM-1) in neonatal rats with intrauterine infection.Methods Thirty SD pregnant rats were randomly divided into two groups of A(normal control,6 rats) and B(intrauterine infection model,24 rats).The intrauterine infection model was established with intraperitonal injection of lipopolysaccharide 0.5 mg/kg,and the specimens from peripheral blood and brain and lung tissues were collected on the 1st,3rd,7th and 14th day after birth.The protein expressions of TREM-1 in the brain and lung tissues were detected by Western blot and immunofluo-rescence.The levels of TREM-1 and TNF-α in peripheral blood were measured by ELISA.ResultsTREM-1 was not positively expressed in group A.Compared with group A,the protein expressions of TREM-1 in the brain and lung tissues and the levels of TREM-1 and TNF-α in peripheral blood were significantly increased on the 1st day,peaked on the 3rd day and gradually decreased on the 14th day after birth in group B.There was a positive correlation between expressions of TREM-1 and TNF-α(r=0.794,P<0.01).Conclusion After intrauterine infection,the expression of TREM-1 in the tissues and peripheral blood of neonatal rats is obviously upregulated and closely related with TNF-α,which may be taken as a potential indicator for predicting the early prognosis of sepsis.
