Abstract:Objective To develop a screening and confirmation method of UPLC-LTQ Orbitrap MS for 48 chemical medicine including fluoroquinolone,sulfanilamide,tetracycline,malachite green,nitrofuran,olaquindox and carbadox mixed into traditional Chinese herbal medicine for fishery.Methods The samples were extracted by ethyl acetate,cleaned by C18 sorbent,separated by reversed phase chromatography(Velocity:250 μl/min,Cylinder temperature:30 ℃,Sample size:10 μl),and gradient eluted with methanol/water(containing 0.1% formic acid).Final detection was performed by LTQ Orbitrap MS in full scanning mode.The TOXID software combined with database retrieval was used for screening and confirmation of 48 veterinary drugs.Results The limits of detection(LODs) and the limits of quantification(LOQs) of 48 veterinary drugs were 0.006 0-0.21 mg/kg and 0.020-0.72 mg/kg,respectively.The method showed good linearity over the linear range,r2=0.9796-0.9997.The recoveries were 81.9%-114.4%,84.0%-114.3%,80.9%-114.8% for the three spiked levels at 0.25,0.50 and 1.0 mg/kg,respectively.The RSDs were in the range of 1.56%-14.3%.Conclusion The method was rapid,accurate,reliable and precise.It was suitable for the screening and confirmation of chemical medicine adulterated into traditional Chinese herbal medicine.
Rapid Identification of Illegally Addition in Traditional Chinese Antidiabetic Medicine by Field Asymmetric Ion Mobility Spectrometric Technique

Abstract:The field asymmetric ion mobility spectrometric (FAIMS) technique was introduced into the rapid analysis of illegally added ingredients in a traditional Chinese antidiabetic medicine for the first time. The metformin, phenformin and glibenclamide as well as the marketed medicine were detected and identified by a co-developed Thermal Desorption-FAIMS (TD-FAIMS) system. The temperature of the sampling tube for thermal desorption was set as 160 degrees C. The test pressure was 1.07x10(5) Pa. The range of the high field was set from 0% to 100% of the maximum field for the FAIMS spectrometer. The Compensation voltage (CV) scanned from. 6 V to 6 V. Eight groups of fingerprint points contained three variables of CV, E/N and Ion current (AI) were extracted from the spectra of the three reference substances and their combination mixtures. The results indicated that the medicine sample had the same spectrum as the mixture of three reference substances, which gave evidence of illegal addition into the traditional Chinese medicine. The result was confirmed by HPLC analysis by comparing spectrum of the medicine sample with those of metformin, phenformin and glibenclamide. The FAIMS instrument, which features its simplicity, high sensitivity and selectivity and fast response, demonstrates huge potential in the rapid analysis of traditional Chinese medicine.

Abstract:A confirmatory and quantitative method based on a high performance liquid chromatography UV detector (HPLC-UV) and a liquid chromatography tandem mass spectrometry (LC-MS/MS) with an extraction procedure of accelerated solvent extraction (ASE) has been developed for simultaneous determination of 15 kinds of fluoroquinolones in various animal origin food samples. The sample preparation procedures consist of an extraction step with acetonitrile and a cleaning-up step with Oasis HLB cartridge. Parameters for extraction pressure and temperature, cycle of ASE, clean-up, and analysis procedure have been optimized systematically. The recoveries of FQNs spiked in the tissues as the muscle, liver, kidney of swine, bovine, chicken and fish at a concentration range of 10-800μg/kg were found between 70.6% and 111.1% with relative standard deviations (RSD) less than 15% in HPLC. The LOD and LOQ of the HPLC for the 15 FQNs were 3μg/kg and 10μg/kg, respectively, and those of the LC-MS/MS were 0.3 and 1μg/kg, respectively. These rapid and reliable methods can be used to efficiently separate, characterize and quantify the residues of 15 FQNs (Marbofloxacin, Enoxacin, Fleroxacin, Ofloxacin, Pefloxacin, Lomefloxacin, Danofloxacin, Enrofloxacin, Orbifloxacin, Cinoxacin, Gatifloxacin, Sarafloxacin, Difloxacin, Nalidixic Acid, Flumequine) in food of animal origin.

Abstract:Fluoroquinolones (FQs) are among the most important antibiotics used in human and veterinary medicines. A simple and effective analytical method based on reversed-phased liquid chromatography with fluorescence was developed and validated for the simultaneous determination of eight FQs in wastewater at trace level. Aqueous samples were extracted using Anpel (TM) MEP cartridges where they were subsequently eluted by formic acid in methanol. The aqueous extracts were analyzed by gradient elution Liquid Chromatography with Fluorescence Detection (LC-FLD), whose mobile phase was composed of acetonitrile and 10mM tetrabutyl ammonium bromide (TBAB). The limits of detection (LOD) and the relative standard deviation (RSD) were as low as 0.11-1.06 mu g/L and 2-5%, respectively. The presented method was successfully applied to quantify FQs in the influent and effluent of several typical Sewage Treatment Plants (STPs) in Shanghai. For the extraction of 100mL influent and 500mL effluent sewage water samples, recoveries obtained were between 79-109% and 80%-105%, respectively. 7 FQs were occurred and identified in the STPs with the concentrations varying from 7 ng/L to 1 mu g/L. Norfloxacin, ciprofloxacin and lomefloxacin were the most frequently detected antibiotics occurring in the wastewaters. The analytical procedure developed may be used for more in-depth studies on the occurrences and the fate of these commonly used pharmaceuticals in the sewage treatment plants and in the aquatic environment.

Abstract:A speedy and selective ultra-HPLC-MS/MS method for simultaneous determination of deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON), 15-ADON. nivalenol and fusarenon X in traditional Chinese medicines (TCMs) was developed. The method was based on one-step sample cleanup using reliable homemade cleanup cartridges. A linear gradient mobile-phase system, consisting of water containing 0.2% aqueous ammonia and acetonitrile/methanol (90:10, v/v) at a flow rate of 0.4 mL/min, and an Acquity UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m) were employed to obtain the best resolution of the target analytes. [C-13(15)]-DON was used as the internal standard to accomplish as accurate as possible quantitation. The established method was further validated by determining the linearity (R-2 >= 0.9990), sensitivity (LOQ, 0.29-0.99 mu g/kg), recovery (88.5-119.5%) and precision (RSD <= 15.8%). It was shown to be a suitable method for simultaneous determination of DON, 3-ADON, 15-ADON, nivalenol and fusarenon X in various TCM matrices. The utility and practical impact of the method was demonstrated using different TCM samples.

Abstract:A generic procedure, which involved accelerated solvent extraction and homemade cleanup cartridges, has been developed for the extraction and purification of 35 mycotoxins in various traditional Chinese medicine (TCM) matrixes, i.e., rhizomes and roots, seeds, flowers, and grasses and leaves, for subsequent analysis by ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). All target analytes could be simultaneously quantitated in less than 17 min per run, showing narrow symmetrical peaks. The developed method was also successfully applied in routine monitoring programs, which implied a significant reduction of both effort and time, to investigate the contamination of TCMs. Among 60 commercial TCMs analyzed, 50 were positive. The achieved data underpin the practical application of the UHPLC-MS/MS method as a valuable tool for the trace analysis of multiple mycotoxins in TCMs.

Simultaneous Determination of the Four Key Fluoroquinolones and Two Antipsychotics in Fish and Shrimp by LC-MS/MS

Abstract:In this study, we developed and validated a liquid chromatography triple quadrupole tandem mass spectrometry (LC-MS/MS) method used to simultaneously determine levels of four fluoroquinolones (ofloxacin, norfloxacin, lomefloxacin, and pefloxacin) and two antipsychotics (diazepam and methaqualone) in fish and shrimp. The samples were extracted with a mixture of anhydrous sodium sulfate and acetonitrile, and purified by C-18 cartridge solid-phase extraction with an optimized eluent. The MS2 method was applied to recognize the molecular structure of these compounds according to a main fragmentation scheme. The key ions of identification and quantification were deduced from chemical structures. Multiple reaction monitoring was used to quantitatively analyse the compounds of interest. Satisfactory linearities were obtained (R-2 >= 0.99) with the limits of quantitation (LOQs) ranging between 0.03 and 1.96 mu g kg(-1). The recoveries were 74-122%, with a relative standard deviation (RSD) below 4.9% for these compounds at the spiking level of three, five, and ten times the LODs, respectively. The LC-MS/MS method allows precise and sensitive determination of residues of six important banned veterinary drugs in fish and shrimp tissue. This methodological approach solved the problem imposed by the need for two or more analysis methods to analyse the compounds of interest described in this study.

Abstract:A rapid and sensitive analytical method for the simultaneous determination of four fluoroquinolones, four tetracyclines and six sulfonamides in chicken muscle using ultra-performance liquid chromatography coupled to tandem mass spectrometry (UPLC–MS–MS) has been developed and validated. Samples were extracted with McIlvaine buffer-acetonitrile, defatted with n-hexane, and analyzed by UPLC–MS–MS. Solvent delay technique was applied in the analysis to remove the non-volatile phosphate and carry out farther on-line SPE clean-up. Satisfactory recoveries (55–110%) of all the veterinary drugs were demonstrated in 1, 10 and 20 μg kg−1 spiked levels with the overall RSD for intra- and inter-day of 14 analytes less than 18%. The LOD and LOQ were 0.3 and 1.0 μg kg−1, respectively. Quantitative results of 103 real samples indicated that the present method was suitable for the quantitative analysis of real samples.

Abstract:MRM→IDA→EPI→spectral library search model was used to establish a HPLC-electrospray ionization tandem mass spectrometry method for the multi-residue detection of fluoroquinolones in pig tissues.The quantitative and qualitative analysis of the target analyses were achieved by LC-MS/MS combining with spectral library search function.Tissue samples were extracted with N,N-Dimethylformamide and acetonitrile,purified with Waters HLB column,The analytical method in the present study was well validated and good results were obtained with respect to precision,repeatability and spiked recovery.The limit of detection of residues was 0.1～0.5μg/kg for fluoroquinolones,and with a linear range from 1.0 to 5000 μg/kg.The mean recovery was of 81.6%～114% and RSD was of 1.1%～11.2%.

Abstract:A simple, rapid and sensitive liquid chromatographic method with programmable fluorescence and ultraviolet detection was developed and validated for simultaneous determination of seven fluoroquinolones (marbofloxacin, ofloxacin, ciprofloxacin, lomefloxacin, danofloxacin, enrofloxacin and difloxacin) and four sulfonamides (sulfadiazine, sulfapyridine, sulfathiazole and sulfadimidine) in chicken muscle in a single run. The tissue sample was extracted with phosphate buffer (pH 6.0) and cleaned-up with a solid phase extraction cartridge. The mean recoveries for each drug in chicken muscle ranged from 78.0 to 105.2% with a relative standard deviation below 9.3% at 0.2–400 ng g −1 fortification levels. The limit of quantification was 0.2–4.0 ng g −1 for fluoroquinolones and 15.0 ng g −1 for sulfonamides.

Abstract:BACKGROUND: Micellar liquid chromatography - fluorescence detection was used to determine the antibiotics flumequine, marbofloxacin, difloxacin, and sarafloxacin in porcine, bovine, poultry, ovine, caprine, rabbit, and equine meat, to verify compliance with EU Regulation 37/2010 with regard to the occurrence of veterinary drugs in food.RESULTS: The analytes were isolated from the matrix by ultrasonication-assisted leaching in a micellar solution, and the supernatant was filtered and directly injected. The fluoroquinolones were resolved in < 19 min using a C18 column, with an isocratic mobile phase of 0.05 mol L-1 sodium dodecyl sulfate - 8% 1-butanol - 0.5% triethylamine buffered at pH 3. The limits of quantification (0.01-0.05 mg kg-1 ) were below the maximum residue limits (0.15-0.4 mg kg-1 ). The method was validated by EU Commission Decision 2002/657/EC guidelines.CONCLUSION: The method shows practical advantages such as simplicity, low cost, eco-friendliness, safety, and applicability for routine analysis, and is useful for surveillance programs. © 2018 Society of Chemical Industry.

Abstract:A new method was developed for the determination of six fluoroquinolone antibiotics including fleroxacin, levofloxacin, ciprofloxacin, lomfloxacin, enrofloxacin, and sparfloxacin in chicken breast muscle, in which the extraction and clean-up were performed in one step by microwave irradiation. The mixture of ACN containing 0.3% v/v phosphoric acid/water pH 3 (70: 30, v/v) was used as the extraction solution and hexane was used as the clean-up solution. The extract was analyzed by liquid chromatography-tandem mass spectrometry system. The RSDs of intra-and inter-day obtained are in the range of 1.0-10.4 and 3.8-13.6%, respectively. In the three fortified levels of chicken breast muscle (20, 100, and 500 ng/g), the recoveries of fluoroquinolone antibiotics ranging from 66.0 to 97.2% are obtained. The LODs are in the range of 2.7-6.7 ng/g. This method simplifies the process of the sample preparation and reduces the operation errors.

Abstract:An ultra-performance liquid chromatography coupled with tandem mass spectrometry (UPLC/MS/MS) method was developed to screen and confirm multi-class veterinary drug residues in pig tissues including pig kidney, liver and meat. Twenty-one drugs of two different classes including seven tetracyclines and four types of quinolones (quinoline, naphthyridine, pyridopyrimidine and cinoline) were determined simultaneously in a single run. The homogenized sample tissues were extracted with EDTA-McIlvaine buffer solution and further purified using a polymer-based Oasis HLB solid-phase extraction (SPE) cartridge. An ACQUITY UPLC (TM) BEH C-18 column was used to separate the analytes followed by tandem mass spectrometry using an electrospray ionization source. MS data acquisition was performed in the positive ion multiple reaction monitoring mode, selecting two ion transitions for each target compound. Recovery studies were performed at different fortification levels. The overall average recoveries from pig muscle, kidney, and liver fortified with quinolones and tetracyclines at three levels ranged from 80.2 to 117.8% based on the use of matrix-fortified calibration with the coefficients of variation ranging from 2.1 to 17.8% (n=6). The limits of quantitation (LOQs) of quinolones and tetracyclines in different tissues ranged from 0.03-4.50 mu g/kg and 0.16-10.00 mu g/kg, respectively. The effects of the extraction solvent, SPE cartridge, elution solvent and sample matrix on the analyte recovery as well as the effects of the mobile phase composition and column temperature on the chromatographic behavior were also studied. Abstract:Objective:To develop a method for simultaneous determination of 13 chemicals added illegally in anti-rheumatic traditional Chinese medicines(TCMs) by rapid resolution liquid chromatography coupled with tandem mass spectrometry(RRLC-MS/MS).Methods:Separation was performed on Agilent Eclipse Plus C18(2.1 mm×10 mm:1.8 μm) column by gradient elution with acetonitrile and 0.1% formic acid aqueous solution as the mobile phase.The chemicals detected in samples were identified by comparison of sample peaks with the retention time,mass spectra and the product ions with those of reference substances.Results: The developed RRLC-MS/MS method was interfered by the adjuvants.The peaks detected could be separated from each other.The range of detection limits(LODs) for 13 chemicals was 0.0031-0.9471 ng.Conclusion:The developed RRLC-MS/MS method was not convenient,accurate and sensitive,which could be used for rapid screening of above-mentioned chemicals added illegally in TCMs.
Stimultaneous Determination of 15 Quinolones in Aquatic Products by Liquid Chromatography-Tandem Mass Spectrometry

Abstract:An effective multiresidual high-performance liquid chromatographic method coupled with mass-spectrometric detection was developed for the trace analysis and validation of 15 quinolones(oxolinic acid,nalidixic acid,flumequine,fleroxacin,ofloxacin,enoxacin,norfloxacin,ciqrofloxacin,enrofloxacin,lomefloxacin,danofloxacin,orbifloxacin,difloxacin,sarafloxacin and sparfloxacin) in aquatic products.The targets were extracted from homogenised tissues using acetonitrile and the extracts were subjected to a stong cation exchange(SCX) solid phase extraction(SPE) cartridges.The linear range was 0～100 μg/L and the correlation coefficients were 0.9924～0.9992.The average recoveries of 15 quinolones and the relative standard deviations ranged from 79.9%～93.8% and 4.8%～14.6%,respectively,in spiked tissue samples at concentration levels ranging from 0.002 mg/kg to 0.04 mg/kg of each quinolone.This method is suitable for aquactic products.

Abstract:A rapid and robust multiresidue method was developed for the analysis of seven quinolones (ciprofloxacin, danofloxacin, enrofloxacin, sarafloxacin, difloxacin, oxolinic acid, and flumequine) in animal muscle tissues by HPLC-FLD. The samples were extracted with phosphate aqueous solution and cleaned with HLB-SPE. The analytes were separated using formic acid aqueous solution-acetontrile system as mobile phase with a linear gradient elution program, and determined by programmable fluorescence detection. The linear range was 0.3–1000 μg l−1 with correlation coefficients (r) greater than 0.9989. The limits of detection were 0.1–0.3 μg kg−1, and limits of quantification were 0.3–1.0 μg kg−1. The mean recoveries for each analyte in chicken and pig muscles ranged from 70.4% to 105.8% with relative standard deviation below 9.3% at 1–100 μg kg−1 fortification levels. The method is convenient, fast, safe, sensitive, and accurate for determining residual quinolones in actual samples.
Highly-sensitive detection of eight typical fluoroquinolone antibiotics by capillary electrophoresis-mass spectroscopy coupled with immunoaffinity extraction

Abstract:Fluoroquinolones (FQs) are a class of synthetic antibiotics with a broad active spectrum towards mycoplasma and bacteria (both Gram positive and negative). The contamination of environmental water samples with FQs has been a concern due to the extensive use of them in the veterinary and pharmaceutical industries. Reliable and simple analytical methods are needed for the monitoring of these FQs. An off-line procedure of immunoaffinity extraction (IAE) followed by capillary electrophoresis-mass spectroscopy (CE-MS) has been developed for simultaneous determination of eight typical FQs in environment water. A broad-specific polyclonal antibody was home-prepared and immobilized on a Sepharose 4B stationary phase for sample affinity extraction and the enrichment factor of our columns was about 100. CE-MS was subsequently applied to FQs separation and quantification. The immunoaffinity column was evaluated in the parameters of selectivity, binding capacity, elution protocol, and recovery. Under the optimized conditions, the developed method was successfully applied to analyze FQs spiked with 2.5 nM to 30 nM of environmental water from a local lake, providing satisfactory recoveries from 72% to 112%, and LODs and LOQs ranging from 1.2 nM to 5.0 nM and 4.0 nM to 17 nM, respectively.

Abstract:Shuanghuanlian oral liquid, a traditional Chinese medicine preparation, is a mixture of three herbs (Flos Lonicerae, Radix Scutellariae and Fructus Forsythiae). In this study, the quantitative analysis of three main active compounds, chlorogenic acid, forsythin and baicalin in samples from different manufacturers was performed rapidly by high-performance liquid chromatography coupled with photodiode array detection followed by Contour Projection coupled to stepwise regression treatment of the obtained three-dimensional spectra in which the partial overlap between adjacent target components existed. The method was validated for linearity (R>0.9940), precision (RSD<1.25%), recovery (92.20-102.50%), limit of detection (0.01-0.02 mu g/mL) and limit of quantification (0.03-0.07 mu g/mL). The results indicated that the combination of the three-dimensional spectra of traditional Chinese medicine and Contour Projection-stepwise regression offered an accurate, simple, low-cost and eco-friendly way for the rapid quantitative analysis of Shuanghuanlian oral liquid samples.

Abstract:A novel, simple and rapid high performance liquid chromatographic method has been developed and validated for the determination of sinafloxacin, a new fluoroquinolone, in rat plasma using 96-well protein precipitation, fused-core C(18)-silica column (4.6mmx50mm, 2.7microm) packed with a new solid support, which is made of 2.7microm particles that consist of a 1.7microm solid core covered with a 0.5microm thick shell of porous silica.The chromatographic separation was achieved with a mobile phase of 20:80 (v/v) of acetonitrile and phosphate buffer (pH=3.0) at a flow rate of 1mlmin(-1). Fluorescence detection was employed with lambda(ex) 295nm and lambda(em) 505nm. Lomefloxacin was used as internal standard (IS). The total analysis time was as short as 3min. The method was sensitive with a limit of detection (LOD) of 2ngml(-1), with good linearity (R(2)=0.9996) over the linear range of 5-500ngml(-1). The intra-day and inter-day precision was less than 5.8% and accuracy ranged from 100.3% to 103.5% for quality control (QC) samples at three concentrations of 10, 50 and 400ngml(-1).The fused-core C(18)-silica column method offered high sample throughput, low injection volume and low consumption of organic solvents. The method was successfully employed in the pharmacokinetic study of sinafloxacin formulation product after tail vein injection to healthy rats.
