Abstract:Background Human defensin peptides with broad-spectrum antimicrobial activity have been implicated in the human defence response towards microbial invasion. Two families of defensins designated alpha- and beta-defensins, respectively, have been identified. Little is known about the expression of both defensin families in human peripheral blood. The purpose of this study was to examine the expression of alpha- and beta-defensin genes in human peripheral blood. Material and methods Fifty-one healthy blood donors were screened for defensin expression. Blood from defensin responders was stimulated by lipopolysaccharide or heat-inactivated Pseudomonas aeruginosa ex vivo. Levels of mRNA were assessed by semiquantitative RT-PCR. Southern blot analysis and sequencing were used to confirm the identity of defensin gene transcripts. Western blotting analysis was used to detect the expression of defensin peptides. Results beta-defensin was undetected in human peripheral blood without stimulation. Following stimulation by lipopolysaccharide or heat-inactivated bacterial cells, the majority (88.2%) of healthy individuals had a detectable expression for beta-defensin-1 gene and 39.2% for beta-defensin-2 gene, compared with none for beta-defensin-3. beta-defensin-1 and -2 mRNAs in the stimulated human peripheral blood of responders became detectable at 3 h and showed a maximum at 6 h following induction by 100 ng mL(-1) of lipopolysaccharide or bacterial cells. In contrast, human alpha-defensins 1-3 mRNA are constitutively expressed in peripheral leukocytes but not up-regulated by lipopolysaccharide or bacterial cells. Conclusions In human peripheral blood, beta-defensin-1 and -2 genes were transiently transcribed and translated following the induction of lipopolysaccharide or heat-inactivated bacterial cells, whereas a-defensins 1-3 genes were constitutively transcribed, and beta-defensin-3 gene was not expressed. The inducible expression of beta-defensin-1 and -2 genes showed interindividual variability.

Abstract:Human beta-defensin-2 (hBD-2), a small cationic peptide, exhibits a broad range of antimicrobial activity. It has been found to play important roles in innate and adaptive immune responses against microbial invasion. For the purposes of this study, hBD-2 gene was cloned from the lesions of human condyloma acuminatum. An expression vector was constructed and transformed into E. coli. hBD-2 was expressed as a fusion protein in both the soluble and insoluble forms, which was further confirmed by western blotting analysis.

Abstract:Human defensin is a family of cationic antimicrobial peptides in human being. During the last two decades a series of endogenous alpha-and beta-human defensins have been discovered. They are important components of the first barrier in human's body against the invasion of various microorganisms, and they are thought to play an important role in linking the innate and adaptive defense system of human being. The recent advances in the research of human defensins were reviewed, including their discovery, molecular and genetic properties, expression regulation, and mechanisms of antimicrobial activity. The possibility to produce human defensins via genetic engineering was also discussed. And the application outlook of human defensins in medicine and curing patients infected with antibiotics-resistant microbials was presented.

Abstract:Human defensins are a family of cationic antimicrobial peptides with molecular weights of 4–5kDa, containing a conserved six disulphide-linked cysteine motif. During the last two decades a series of endogenous α- and β-human defensins were discovered. They exhibit a broad range of antimicrobial properties and are thought to be ideal therapeutic agents because of their potential ability to circumvent the problems of acquired resistance often observed with other antimicrobial therapies. Because of their appealing medical and pharmaceutical potential there has been an emphasis on human defensins in medical and molecular biology research in recent years. This paper aims to present a comprehensive review of recent advances in the study of human defensins including their discovery, classification, molecular properties, expression, mechanisms of action and potential medical applications. In addition, the advances in producing human defensins via genetic engineered cells are summarized from research works in our group (besides host cells including E. coli, B. subtilis and yeast systems, the cell-free protein synthesis system was also employed to express human β-defensin-2) along with other related published works. The present challenges and prospects for the potential application of human defensins are also discussed.

Abstract:Defensins are small, multifunctional cationic peptides. They typically contain six conserved cysteines whose three intramolecular disulfides stabilize a largely beta-sheet structure. This review of human alpha-defensins begins by describing their evolution, including their likely relationship to the Big Defensins of invertebrates, and their kinship to the beta-defensin peptides of many if not all vertebrates, and the beta-defensins found in certain non-human primates. We provide a short history of the search for leukocyte-derived microbicidal molecules, emphasizing the roles played by luck (good), preconceived notions (mostly bad), and proper timing (essential). The antimicrobial, antiviral, antitoxic, and binding properties of human alpha-defensins are summarized. The structural features of alpha-defensins are described extensively and their functional contributions are assessed. The properties of HD6, an enigmatic Paneth cell alpha-defensin, are contrasted with those of the four myeloid alpha-defensins (HNP14) and of HD5, the other alpha-defensin of human Paneth cells. The review ends with a decalogue that may assist researchers or students interested in alpha-defensins and related aspects of neutrophil function.

Abstract:Defensins play a vital role in the early stage of infection before adaptive immune responses are generated. Thus far, only limited detailed genomic data for avian defensin genes have been described. Here, using bacterial artificial chromosome libraries, we found that a 95kb and 177kb sequences in the golden pheasant (Chrysolophus pictus, Galliformes) and hwamei (Garrulax canorus, Passeriformes) corresponds to 16 and 30 defensin genes, respectively. Fluorescence in situ hybridization assigned defensin gene clusters in the golden pheasant and hwamei to chromosomes 2q and 3q, respectively. In combination with the previous chicken (Gallus gallus, Galliformes) and zebra finch (Taeniopygia guttata, Passeriformes) defensin clusters, the comparative genomic analysis revealed that lineage-specific duplications and deletions have given rise to clearly different genomic structures. On the basis of genomic characteristics, we further found that transposable elements act as agents of evolution, causing direct and indirect copy number variations in defensin genes via duplications. Tissue expression analysis showed that the Passeriformes-specific duplicated AvBD1 and AvBD3 genes are mainly expressed in the upper and lower respiratory tract, tongue, and spleen. Our analyses indicate that the duplication-and-deletion of defensin genes conformed to the birth-and-death evolutionary process and that transposable elements induced the duplication of defensin genes. Moreover, the respiratory system-specific expression pattern of Passeriformes-specific expanded AvBD1 and AvBD3 genes suggests their important role in maintaining the singing trait of songbirds. The understanding of the genomic structure, expression, and evolution of defensin genes can provide a crucial immunological foundation to study and prevent avian diseases.

Abstract:Background: Defensins are key effector molecules of innate immunity that can contribute towards the diagnosis and monitoring of chronic obstructive pulmonary disease (COPD). The present study was conducted to investigate the role of alpha-defensins in patients with COPD by quantifying serum and sputum samples. Methods: A total of 180 patients were enrolled and divided into four groups, and sputum and serum values of alpha-defensins were assessed. The sensitivity, specificity, and accuracy of sputum alpha-defensin as a diagnostic biomarker were evaluated to assess its utility in diagnosing COPD. Results: The mean value of sputum alpha-defensins was found to be statistically significant amongst the four groups (P < 0.001). The highest levels were found in subjects with AECOPD (385.76 ± 116.62 ng/mL). Sputum alpha-defensins were found to be negatively correlated with FEV 1 values (rho = -0.31, P < 0.001). Conclusion: Sputum alpha-defensins can be used as a potential marker for predicting acute exacerbation of COPD. In addition, they could serve as an indicator of disease severity in COPD patients.

Abstract:Human beta-defensin-2 (hBD-2) is a cysteine-rich cationic low molecular weight antimicrobial peptide, which exhibits a broad range of antimicrobial activity without observed acquired resistance. In this work, multiple copies of the hBD-2 gene were linked in tandem and the expression of the multiple joined genes in two fusion expression system, pET28a(+) and pGEX-4T-2, was examined. Using plasmid pET28a(+) with one, two, and four copies of the hBD-2 gene, the expressed level was relatively low, whereas much higher with plasmid pGEX-4T-2, and the fusion products, most of which in insoluble form, account for approximately 26% of the total insoluble cellular proteins.

Abstract:Severe sepsis is an infection-initiated, inflammatory syndrome which complicated with organ dysfunction. Genetic factors affect the incidence and outcome of severe sepsis. Human beta-defensins, cysteine-rich cationic antimicrobial polypeptides with 3 disulfide bridges, play an important role in innate immunity and adaptive immunity in sepsis. Our previous studies have shown that human beta-defensin 1 gene was transiently transcribed following the induction of LPS or heat-inactivated bacterial cells and the inducible expression of beta-defensin1 gene showed interindividual variability. The purpose of this study was to evaluate whether single-nucleotide polymorphisms (SNP) in the 5′-untranslated region of the HBD-1 gene are associated with the susceptibility to severe sepsis or with the outcome of severe sepsis. Two hundred and fifty patients with severe sepsis who had been treated at the intensive care unit of the university hospital were studied. One hundred seventy postoperative patients without sepsis served as controls. The DNA fragment containing the three SNPs (-52G/A, -44C/G and -20G/A) in the 5′-untranslated region of the HBD-1 gene were amplified and sequenced. There was a significant difference in the distribution of genotype and alleles of -20G/A SNP and -44C/G SNP between severe sepsis and controls. While, there was no significant difference in the distribution of genotype and alleles of -52G/A SNP between severe sepsis and controls. Further, there was no significant difference in the distribution of genotype and alleles of -20G/A SNP, -44C/G and -52G/A SNP between survivors and non-survivors. -20G/A SNP and -44C/G SNP is associated with the susceptibility to severe sepsis.

Abstract:Lipopolysaccharide (LPS) is an important pathological factor involved in serious inflammatory diseases and male reproductive impairments. Emerging evidence demonstrates that antimicrobial peptides possess protective activity in response to LPS-induced inflammation. However, the LPS-binding and/or immunosuppressive activity of beta-defensins (DEFBs) has been underestimated. In the present work, we characterized a novel human defensin, DEFB114, which was expressed predominantly in the epididymis and gingival cells at the RNA level. Homogenous recombinant DEFB114 peptides were prepared and characterized using mass spectrometry. DEFB114 protein exhibited a broad spectrum of antimicrobial activity with salt sensitivity against typical pathogenic microbes (i.e. Escherichia coli, Staphylococcus aureus, and Candida albicans). Interestingly, DEFB114 demonstrated novel LPS-binding activity in vitro and inhibited TNF-alpha release in RAW264.7 cultures through the inhibition of MAPK p42/44 when challenged with LPS. Moreover, DEFB114 could also rescue the LPS-induced reduction of human sperm motility in vitro and protect D-galactosamine-sensitized C57BL/6 mice from LPS-induced lethality in vivo. The protective activity of DEFB114 on RAW264.7, human sperm, and the D-galactosamine-sensitized mice was disulfide bond-dependent because alkylated DEFB114 lost its activity. The low cytotoxicity of the DEFB114 peptide toward human erythrocytes is indicative of its potential therapeutic use in the treatment of LPS-induced inflammation, LPS contamination, and potentially septic shock.

Abstract:s-defensins are antimicrobial peptides secreted by epithelial cells that can bind to cellular receptors. CCR2 and CCR6 are the two cellular receptors known to bind human s-defensin (hBD) 2 and -3. Both of these receptors are of crucial relevance in HIV infection. CCR6 is expressed, often in concert with CCR5, on cells that are highly susceptible to HIV infection: memory T cells, Th17 cells, α4s7+ cells, and defects in CD4+CCR6+ cells have been associated with faster AIDS progression. CCR2 is expressed on monocytes and macrophages, cells that are reservoirs of HIV infection and that are known to mediate central nervous system damage. Our studies show that hBD2, hBD3, and CCR6 ligand MIP-3α/CCL20 inhibit HIV infection via CCR6 by increasing expression of the antiviral protein APOBEC3G. This increase is due to a transcriptional mechanism mediated by intracellular signaling. hBD2 also inhibits HIV replication in macrophages that express CCR2. Our findings suggest novel therapeutic and preventive approaches that exploit CCR6 and CCR2-mediated intracellular signaling to inhibit HIV infection in highly susceptible cells.
Expression of human beta-defensin-2 with multiple joined genes in Escherichia coli.

Abstract:Human beta-defensin (HBD)-2 is a small cationic peptide with a broad range of antimicrobial activity. In this study, multiple copies of the hBD2 gene were linked in tandem, and a number of different Escherichia coli expression vectors were evaluated, including pQE-30, pBV220, pET-28a(+), and pGEX-4T-2. No expression of multiple joined genes was detectable in the pQE-30 expression system, whereas in pBV220 with one or two joined hBD2 genes and in pET-28a(+) with one, two, or four copies, target proteins were expressed at a low level. Only when pGEX-4T-2 was applied as expression plasmid with one or two joined hBD2 genes were target proteins expressed in high level, and the expressed fusion proteins account for 26 and 16% of the total insoluble proteins, respectively. In the pGEX-4T-2 and pET-28a(+) expression systems, the effects of multiple joined genes on the growth of host strains and plasmid stability were examined. Host cells containing plasmid carrying fewer copies of hBD2 genes were faster in cell growth. Plasmid stability decreased with an increase in multiple joined genes, which was especially noticeable in the pET-28a(+) system. Furthermore, the presence of glucose in culture medium brought about a positive effect on plasmid stability when using pET28-nhBD2 as expression vectors.

Abstract:Human β‑defensin 6 (DEFB106) is an antimicrobial peptide expressed in the epididymis, testis and lung, which indicates that DEFB106 may be involved in innate immunity and fertility. However, as a β‑defensin, this protein has not been well characterized. Using an intein‑mediated fusion expression system, the recombinant DEFB106 was expressed and purified (yield, 3‑5 mg/l) under optimized conditions. The purified protein was characterized using mass spectrometry and circular dichroism spectroscopy. The measured molecular weight was consistent with its theoretical value and the predominant secondary structure was β‑sheet, the common structure of β‑defensin family members. The purified DEFB106 showed antimicrobial activity against not only Escherichia coli (E. coli) and Candida albicans (C. albicans) SC5314, but also Staphylococcus aureus (S. aureus) CMCC26003. Furthermore, it exhibited a high affinity for heparin and lipopolysaccharide. In addition, it was determined that native DEFB106 was located in the epididymis, bone marrow and skin. These observations may aid in the determination of the physiological and pathological functions of DEFB106.

Abstract:Objective Human beta defensins(HBD) are antimicrobial peptides mainly generated by the epithelia of human body,Allicin is a antimicrobial drug made in China,our research is to investigate what might be the mechanism involved in improving the immunity of human body. Methods We used RPMI 1640 which included 5 μg/mL Allicin to culture the lung cancer cell line A549,and detected the mRNA expression levels of different kinds of HBD(HBD1~3). Results The HBD3 expression level could be improved,and there were no effects on the expression level of HBD1 and HBD2. Conclusion Allicin can improve the expression level of HBD3 in the cell line A549,that might be one of the reasons which allicin involve in the adjustment of human immunity.It further confirmed that there were different signalling pathways between HBD3 and HBD2.

Abstract:In this work, taking human beta-defensin-2 (HBD2) as a demonstrative molecule, the strategies for high efficient production of functional human beta-defensins in E. coli were studied. Fusion mature HBD2 (TrxA-mHBD2) showed high solubility and productivity without the need for lowering the cultivation temperature. The solubility of target fusion protein could attain 81.3% even at 37 degrees C with a volumetric productivity as high as 235 mg/L in a rich medium MBL at the same temperature and reached 346 mg/L at 28 degrees C. The His-Tag in the fusion protein enabled the application of affinity chromatography separation to obtain high purity of the overexpressed recombinant fusion protein. After digestion by enterokinase, purification via cationic exchange chromatography, and desalting by ultrafiltration, mature HBD2 product was obtained with a purity of 95% in an overall recovery of 29.2%. The antimicrobial activity of the recombinant mature HBD2 and the influence factors were tested using E. coli K12D31 as a sensitive strain.
