Abstract:Lung cancer is one of the most common malignant tumors,with high morbidity and mortality,which is easy to produce the resistance to radiotherapy and chemotherapy.We still lack of effective therapies at present.With the development of Molecular Biology,gene therapy might be a promising therapeutic target for lung cancer,which has attracted more and more attention in medical field.The occurrence and development of lung cancer is a complex process,related to multiple genes,including the activation of proto-oncogenes and inactivation of anti-oncogenes.There are many methods of gene therapy,each of them has merits and defects.Combined treatment had a good effect.This review summarized several methods of gene therapy,in order to provide convenience to scholars in science research and clinical therapy.

Abstract:Lung cancer is one of the most common malignant tumors,is the leading cause of the tumor-related death in the world.The standard therapy is currently the chemotherapeutic regimes with the basis of platinum.If the patients' conditions advance,the choice of thrapy is limitted.In recent years,researchers have paid more attention to immunotherapy,this kind of treatment contains tumor vaccines,programmed death-1(PD-1),cytotoxic T lymphocyte antigen-4 (CTLA-4) and so on.The research data have supported the immunotherapy effect.This article reviews the mechanism of anti-tumor vaccines,PD-1,CTLA-4 inhibitors,shows some results about immunotherapy agents in recent series of clinical trials,discusses the biomarkers for predicting the efficacy of immune drugs,introduces the potential problems and directions of future treatment.

Abstract:Gene therapy is of great promise to replace or counteract a defective gene thus curing a disease at its genetic origins.However, there have been many hur-dles to put this simple concept into practice for a long term.Since the end of last century, prosperous devel-opment of genetic engineering and vector technologies have advanced gene therapy in a diversity of diseases, including genetic disorders, cancer, and chronic infec-tions,etc.Recently,clinical trials of gene therapy have achieved breakthrough progress worldwide with remarka-ble therapeutic effects and safety record.These evi-dence demonstrate that gene therapy has the potential to bring innovation into traditional medicine for some pre-viously incurable conditions.In addition, gene editing and stem cell technologies are also contributing to revi-talization and advances of gene therapy.In this context we review the efficacy and safety achievements during the development of gene therapy,as well as ethical con-cerns related to current clinical trials, and further dis-cuss the research hotspots,development prospects,and challenges for gene therapy.

Abstract:Gene therapy is one of the most attractive fields in tumor therapy. In past decades, significant progress has been achieved. Various approaches, such as viral and non-viral vectors and physical methods, have been developed to make gene delivery safer and more efficient. Several therapeutic strategies have evolved, including gene-based (tumor suppressor genes, suicide genes, antiangiogenic genes, cytokine and oxidative stress-based genes) and RNA-based (antisense oligonucleotides and RNA interference) approaches. In addition, immune response-based strategies (dendritic cell- and T cell-based therapy) are also under investigation in tumor gene therapy. This review highlights the progress and recent developments in gene delivery systems, therapeutic strategies, and possible clinical directions for gene therapy.

Abstract:Gene-based therapies for cancer in clinical trials include strategies that involve augmentation of immunotherapeutic and chemotherapeutic approaches. These strategies include ex vivo and in vivo cytokine gene transfer, drug sensitization with genes for prodrug delivery, and the use of drug-resistance genes for bone marrow protection from high-dose chemotherapy. Inactivation of oncogene expression and gene replacement for tumor suppressor genes are among the strategies for targeting the underlying genetic lesions in the cancer cell. A review of clinical trial results to date, primarily in patients with very advanced cancers refractory to conventional treatments, indicates that these treatments can mediate tumor regression with acceptably low toxicity. Vector development remains a critical area for future research. Important areas for future research include modifying viral vectors to reduce toxicity and immunogenicity, increasing the transduction efficiency of nonviral vectors, enhancing vector targeting and specificity, regulating gene expression, and identifying synergies between gene-based agents and other cancer therapeutics.

Abstract:IMPORTANCE OF THE FIELD:Due to the absence of early diagnosis, the highly invasive and metastatic features and the lack of effective therapeutic modalities, the prognosis of patients with pancreatic cancer is poor. Gene therapy is currently regarded as a potential and promising therapeutic modality for pancreatic cancer.AREAS COVERED IN THIS REVIEW:This article summarizes an update of gene therapy approaches and reviews the latest progress in gene delivery systems that have been tested on pancreatic cancer.WHAT THE READER WILL GAIN:The treatment effectiveness of gene combination therapy is better than that of the regulation of single-gene or single gene therapy approaches. Naked DNA is limited because of degradation by intracellular and extracellular nucleases. Virus vectors show high transfection efficiency but are limited due to immunogenicity, inflammatory response and potential carcinogenicity. Non-viral vectors, such as cationic polymers or inorganic nanoparticles, show an important feature that they can be easily modified, and the progress of materials science will provide more and better non-viral vectors, accordingly improving the efficiency and safety of gene therapy, which will make them the most promising vectors for pancreatic cancer.

Abstract:Lung cancer is a leading cause of cancer-related death globally with the highest morbidity and mortality. Non-small cell lung cancer (NSCLC) is the most common histological type of lung cancer. Traditional therapies, even molecular targeted therapy, have improved survival benefit in patients with lung cancer in the past. However, the limitations of these therapies are becoming more and more common. Recently, with the deep uderstanding of immune system and anti-tumor immune reaction, immunotherapy has obtained more and more attention. As key cells of the immune system, T cells play the pivotal role in anti-tumor immune reaction via direct or indirect methods. T cells-based immunotherapy is the critical part of the treatment of NSCLC. Here in this review is to summarize the progress on T cells-based therapy for the treatment of patients with NSCLC. Abstract:The prospect of gene therapy for inherited and acquired respiratory disease has energized the research community since the 1980s, with cystic fibrosis, as a monogenic disorder, driving early efforts to develop effective strategies. The fact that there are still no approved gene therapy products for the lung, despite many early phase clinical trials, illustrates the scale of the challenge: in the 1990s, first generation non-viral and viral vector systems demonstrated proof-of-concept but low efficacy. Since then, there has been steady progress towards improved vectors with the capacity to overcome at least some of the formidable barriers presented by the lung. In addition, the inclusion of features such as codon optimisation and promoters providing long-term expression have improved the expression characteristics of therapeutic transgenes. Early approaches were based on gene addition, where a new DNA copy of a gene is introduced to complement a genetic mutation: however, the advent of RNA-based products that can directly express a therapeutic protein or manipulate gene expression, together with the expanding range of tools for gene editing, has stimulated the development of alternative approaches. This review discusses the range of vector systems being evaluated for lung delivery; the variety of cargoes they deliver, including DNA, antisense oligonucleotides, mRNA, siRNA and peptide nucleic acids; and exemplifies progress in selected respiratory disease indications.

Abstract:In contrast to conventional therapies that require repeated dosing, gene therapy can treat diseases by correcting defective genes after a single transfection and achieving cascade amplification, and has been widely studied in clinical settings. However, nucleic acid drugs are prone to catabolism and inactivation. A variety of nucleic acid drug vectors have been developed to protect the target gene against nuclease degradation and increase the transformation efficiency and safety of gene therapy. In addition, gene therapy is often combined with chemotherapy, phototherapy, magnetic therapy, ultrasound, and other therapeutic modalities to improve the therapeutic effect. This review systematically introduces ribonucleic acid (RNA) interference technology, antisense oligonucleotides, and clustered regularly interspaced short palindromic repeat/CRISPR-associated nuclease 9 (CRISPR/Cas9) genome editing. It also introduces the commonly used nucleic acid drug vectors, including viral vectors (adenovirus, retrovirus, etc.), organic vectors (lipids, polymers, etc.), and inorganic vectors (MOFs, carbon nanotubes, mesoporous silica, etc.). Then, we describe the combined gene therapy modalities and the pathways of action and report the recent applications in solid tumors of the combined gene therapy. Finally, the challenges of gene therapy in solid tumor treatment are introduced, and the prospect of application in this field is presented.

Abstract:Cancer is the main contributor for mortality in the world. Conventional therapy that available as the treatment options are chemotherapy, radiotherapy and surgery. However, these treatments are hardly cell-specific most of the time. Nowadays, extensive research and investigations are made to develop cell-specific approaches prior to cancer treatment. Some of them are photodynamic therapy, hyperthermia, immunotherapy, stem cell transplantation and targeted therapy. This review article will be focusing on the development of gene therapy in cancer. The objective of gene therapy is to correct specific mutant genes causing the excessive proliferation of the cell that leads to cancer. There are lots of explorations in the approach to modify the gene. The delivery of this therapy plays a big role in its success. If the inserted gene does not find its way to the target, the therapy is considered a failure. Hence, vectors are needed and the common vectors used are viral, non viral or synthetic, polymer based and lipid based vectors. The advancement of gene therapy in cancer treatment will be focussing on the top three cancer cases in the world which are breast, lung and colon cancer. In breast cancer, the discussed therapy are CRISPR/Cas9, siRNA and gene silencing whereas in colon cancer miRNA and suicide gene therapy and in lung cancer, replacement of tumor suppressor gene, CRISPR/Cas9 and miRNA.

Abstract:Introduction: Lung cancer commonly occurs at a high incidence worldwide. Application of aerosol gene delivery systems using various kinds of vectors can improve the patient's quality of life by prolonging the survival rate.Areas covered: This review provides a recent update on aerosol gene delivery strategies using various kinds of vectors and gene-modification technologies. Peptide-mediated gene therapy achieves specific targeting of cells and highly improves efficacy. Promoter-operating expression and the CRISPR/Cas9 system are novel gene therapy strategies for effective lung cancer treatment. Furthermore, hybrid systems with a combination of vectors or drugs have been recently applied as new trends in gene therapy.Expert opinion: Although aerosol gene delivery has many advantages, physiological barriers in the lungs pose formidable challenges. Targeted gene delivery and gene-editing technology are promising strategies for lung cancer therapy. These strategies may allow the development of safety and high efficiency for clinical application. Recently, hybrid gene therapy combining novel and specific vectors has been developed as an advanced strategy. Although gene therapy for lung cancer is being actively researched, aerosol gene therapy strategies are currently lacking, and further studies on aerosol gene therapy are needed to treat lung cancer.

Abstract:The p53 gene is one of the genes most closely associated with human tumors and has become a popular target for tumor drug design. Currently, p53-based gene therapy techniques have been developed, but these therapies face challenges such as immaturity, high safety hazards, limited efficacy, and low patient acceptance. However, researchers are no less enthusiastic about the treatment because of its theoretical potential to treat cancer. In this paper, the advances in p53-based gene therapy and related nucleic acid delivery technologies were reviewed and prospected in order to support further development in this field.

New specific target genes and further development of gene technology may bring the break-through in this field.

Abstract:Despite setbacks in the past and apparent hurdles ahead, gene therapy is advancing toward reality. The past several years have witnessed this new field of biomedicine developing rapidly both in breadth and depth, especially for the treatment of cancer, thanks largely to the better understanding of molecular and genetic basis of oncogenesis and the development of new and improved vectors and technologies for gene delivery and targeting. This article is intended to provide a brief review of recent advances in cancer gene therapy using adenoviruses, both as vectors and as oncolytic agents, and some of the recent progress in the development of immunotoxins for use in cancer gene therapy.

Abstract:Lung cancer is a leading cause of cancer-related death worldwide, with a very poor overall five-year survival rate. The intrinsic limitations associated with the conventional diagnosis and therapeutic strategies used for lung cancer have motivated the development of nanotechnology and nanomedicine approaches, in order to improve early diagnosis rate and develop more effective and safer therapeutic options for lung cancer. Cancer nanomedicines aim to individualize drug delivery, diagnosis, and therapy by tailoring them to each patient's unique physiology and pathological features—on both the genomic and proteomic levels—and have attracted widespread attention in this field. Despite the successful application of nanomedicine techniques in lung cancer research, the clinical translation of nanomedicine approaches remains challenging due to the limited understanding of the interactions that occur between nanotechnology and biology, and the challenges posed by the toxicology, pharmacology, immunology, and large-scale manufacturing of nanoparticles. In this review, we highlight the progress and opportunities associated with nanomedicine use for lung cancer treatment and discuss the prospects of this field, together with the challenges associated with clinical translation.

Abstract:Based on the theoretical and clinical development of modern medicines, gene therapy has been a promising treatment strategy for cancer and other diseases. The practice of gene therapy is nearly 27 years old, since the first authorized gene transfer study took place at the National Institute of Health in 1989. However, gene therapy was not readily adopted worldwide, until recently. Several gene therapy clinical trials have been carried out in China since 1998, and medical research in China has flourished. In this report, we review the history of gene therapy in China, focusing on treatment protocol, the administration cycle, dosage calculation, and the evaluation of therapeutic effects, in order to provide more information for the additional development of this promising treatment strategy.
