The protection of anti-TNFR1 neutralizing antibody toward the hepatocytes of the acute hepatic failure mice

Abstract:Objective To study the roles of anti-TNFR1 neutralizing antibody for hepatocytes of the acute hepatic failure mice.Methods Seventy-two male mice were divided into three groups randomly. The model group were induced of TNF-α and D-Ga1N. The treatment group were administered anti-TNFR1-Ab,TNF-α and D-Ga1N. The histopathological change was observed by HE and TUNEL. Results Profuse apoptosis and necrosis emerged in hepatocytes when tested by TUNEL and HE method. However, in the treatment group, the liver injury was remarkably ameliorated based on the serum transaminase levels and histological change.Conclusion Anti-TNFR1-Ab treatment provides the basis for a novel therapeutic modality in which an unfavorable apoptotic process can be inhibited in acute hepatic failure mice.
