Intracerebroventricular Injection of Streptozotocin Caused Brain Energy Deficiency and Spatial Memory Impairment in Rats

Abstract:Objective To testify whether dysfunction of insulin/insulin receptor transduction system may cause brain energy deficiency and spatial memory impairment in rats. Methods Rats randomized into groups to receive intracerebroventricular (icv) injection of streptozotocin (STZ) or normal saline (NS) bilaterally at 1st and 3rd days. After 21 days of the operation, enzyme activities of succinate dehydrogenase, cytochrome-oxydase, Na~+- K~+-ATPase, and Ca~(2+)-Mg~(2+)-ATPase were measured. High performance liquid chromatography (HPLC) method was used to determine ATP, ADP and AMP concentrations. Morris water maze was used to assay spatial memory impairment. Results Rats received icv injection of STZ bilaterally at 1st and 3rd days showed decrease activities of succinate dehydrogenase, cytochrome-oxydase, Na~+-K~+-ATPase, Ca~(2+)-Mg~(2+)-ATPase and decreased ATP concentration with spatial memory impairment. Conclusions Introcerebroventricular injection of STZ caused brain ATP deficiency with spatial memory impairment, suggested dysfunction of insulin/insulin receptor transduction system may play an important role in the pathogenesis of sporadic Alzheimer disease.
