Grafting Tests Showed the Lateral Branching Development Was Inhibited by a Graft-transmissible Signal in Cucumber

Abstract:In this study, from a collection of breeding lines for fresh-eaten cucumber cultivars, we chose two different branchless lines S61 and S92 to primarily explore the molecular mechanisms of the cucumber branching. Reciprocal grafting in branching line S06 and branchless lines showed that the branching phenotypes of scions could be reversed by one of branchless rootstock S92; however the other branchless line S61 couldn’t suppress the branching phenotypes of scions. Decapitated experiments showed that lateral buds were generated from the leaf axils of S92, but no lateral buds were produced in the leaf axils of S61. Further histological experiments revealed that only floral meristems in the region of leaf axil of S61 and lateral bud meristem in the same region of S92 can be detected. The branchless phenotype of S61 is caused by the obstacle in lateral meristem initiation whereas the lateral branching development of S92 was inhibited by a graft-transmissible signal.
