Construction of lentiviral vector plasmid containing EEF1A2 gene and its stable expression in SW1990 cell

Abstract:The full length EEF1A2 gene was synthesized by PCR method and cloned to lentivirus plasmid pGLV5-EF1a-EGFP/Puro to construct recombinant expression plasmid pGLV5/EEF1A2.After recombinant plasmid was identified by PCR,restriction enzymes and DNA sequence analysis,the pGLV5/EEF1A2 plasmid and three packaging plasmids(pVSV-G,pGag/Pol,pRev) were cotransfected into 293T cells to produce a replicationin competent lentivirus.The lentivirus was added to pancreatic cancer cell SW1990 and selected with puromycin for the stable transfected EEF1A2/SW1990 cell.Real-time PCR and Western blot results showed the expression of EEF1A2 was significantly upregulated in EEF1A2/SW1990 cells compared to parental cells(P < 0.05),and MTT assay showed enhanced proliferation ability of EEF1A2/SW1990 cells compared to parental cells(P < 0.05).The lentiviral expression system of EEF1A2 gene and stable transfected EEF1A2/SW1990 cells were established successfully.
