Abstract:Objective To investigate the effect of quercetin on the proliferation and apoptosis of rat mesangial cells.Methods Rat mesangial cells were incubated with different concentration of quercetin.Cell proliferation was assessed by MTT colorimetric assay.Cell cycle and apoptosis were analyzed by flow cytometry.Results Administration of 5-30μmol/L quercetin into culture medium could significantly inhibit mesangial cell proliferation in a dose and time-dependent manner;subsequently increased the cells in G_0/G_1 phase and decreased the cells in S phase.High dose of quercetin also could induce mesangial cells to apoptosis.Conclusion Quercetin could significantly inhibit mesangial cell proliferation and make it arrest in G_0/G_1 phase and induce the cell apoptosis.
Artemisinin Induces ROS-mediated Caspase3 Activation in ASTC-a-1 Cells

Abstract:Artemisinin (ART), an antimalarial phytochemical from the sweet wormwood plant or a naturally occurring component of Artemisia annua, has been shown a potential anticancer activity by apoptotic pathways. In our report, cell counting kit (CCK-8) assay showed that treatment of human lung adenocarcinoma (ASTC-a-1) cells with ART effectively increase cell death by inducing apoptosis in a time- and dose-dependent fashion. Hoechst 33258 staining was used to detect apoptosis as well. Reactive oxygen species (ROS) generation was observed in cells exposed to ART at concentrations of 400 μM for 48 h. N-acetyl-L-cysteine (NAC), an oxygen radical scavenger, suppressed the rate of ROS generation and inhibited the ART-induced apoptosis. Moreover, AFC assay (Fluorometric assay for Caspase3 activity) showed that ROS was involved in ART-induced caspase3 acitvation. Taken together, our data indicate that ART induces ROS-mediated caspase3 activation in a time-and dose-dependent way in ASCT-a-1 cells.

Abstract:Objective To study the effects of simvastatin on proliferation and apoptosis in rat mesangial cells in vitro, and to investigate the signal pathways involved in apoptosis induced by simvastatin. Methods Cultured mesangial cells were treated with simvastatin. Proliferation of mesangial cells was examined by MTT assay. Simvastatin-treated apoptotic mesangial cells were observed by electron microscopy. Propidium iodide (PI) staining and flow cytometry were employed for quantitative measurement of apoptosis. Caspase-3 activation was determined by CaspGLOW Green Caspase-3 Staining Kit. Results (1)Simvastatin significantly inhibited proliferation of mesangial cells compared with control (P<0.05). (2)Under electron microscopy, mesangial cells with condensed chromatin, shrunken plasma, marginated nuclear chromatin or apoptotic body were observed in simvastatin-treated mesangial cells. Quantitative analysis of apoptotic mesangial cells showed that simvastatin did induce an increased apoptosis rate of rat mesangial cells in a dose-dependent manner. (3)Caspase-3 expression was activated by simvastatin in a dose- and time-dependent manner. Conclusions Proliferation inhibition and apoptosis induction of mesangial cells are involved in the mechanisms of the cholesterol-independent effects of simvastatin on the renal protection. The latter may be induced by activation of caspase-3.
Inhibiting Effect of Dihydroartemisinin on Proliferation of Rat Mesangial Cells in Culture

Abstract:Objective: To investigate the inhibiting effect of dihydroartemisinin on proliferation of rat mesangial cells in vitro. Methods: Rat mesangial cell line was cultured in vitro, incubating with different concentrations and different time of DHA. Cell proliferation was assessed by MTT colorimetric assay. Results: DHA could significantly inhibit the proliferation of mesangial cells, with a relatively low LD 50, 19.7?μmol/L. The inhibiting effect shows a dose-dependent and time-dependent manner. Inhibition at 100?μmol/L of DHA is the most significant among the different concentrations of DHA (0?μmol/L to 100?μmol/L). And mesangial cells are the most significantly inhibited after 72 hours of incutation with DHA, among the different time courses (0?hour to 72?hours) (P0.01). Conclusion: Dihydroartemisinin could significantly inhibit mesangial cells proliferation.

Abstract:Objective:To study the effects of artesunate(Art) on the proliferation and apoptosis of rat hepatic stellate cells T6 (HSC-T6).Method:HSC-T6 was cultured in vitro.Morphological changes of HSC-T6 after the drug treatment were observed by an inverted microscope.The effects of Art with different concentrations on the proliferation of HSC-T6 were detected at different times by CCK-8.A flow cytometry with double staining of Annexin and PI was used to detect the apoptosis rate of HSC-T6.Result:The results shown by the inverted microscope suggested the number decrease of the adherent cells with shorter and thinner synapses,and the cell surface wrinkled and the shape changed from star to round.Art with the concentration of 12.5-200μg·ml~(-1) during 0-72 h exhibited notable anti-proliferative effect on HSC-T6(P <0.01) with time-and dose-dependence.The results of flow cytometry showed that the apoptosis rate of HSC-T6 was significantly enhanced with the increase of the Art dose and the acting time(P <0.05).Conclusion:Art can inhibit the proliferation and promote the apoptosis of HSC-T6.
Effect of Caspase Inhibitor Ac-DEVD-CHO on Apoptosis of Vascular Smooth Muscle Cells Induced by Artesunate

Abstract:Numerous studies have shown that the proliferation and apoptosis of vascular smooth muscle cells play a key role in restenosis. Artesunate is a triterpenoid with a peroxide structure and its antimalarial, antitumor, and antiangiogenetic activities can inhibit the proliferation and apoptosis of multifarious cells. Apoptosis is caused by the activation of a series of intracellular proteolytic enzymes, among which caspase-dependent apoptosis was the earliest to be recognized. The purpose of this article is to study the effects of caspase-3 inhibitor Ac-DEVD-CHO on proliferation and apoptosis of vascular smooth muscle cells induced by Artesunate and to explore the mechanism of Artesunate-induced apoptosis of vascular smooth muscle cells. By using the method based on methyl thiazolyl tetrazolium to observe the effects of Artesunate on the growth and proliferation of vascular smooth muscle cells; observing the change in cell shape before and after Artesunate administration by transmission electron microscopy; detecting the changes in cell cycle and apoptosis rates before and after drug administration by flow cytometry; detecting the activity of caspase-3 in the caspase apoptosis pathway by the Western Blot method, we found that Artesunate inhibits the growth and proliferation of vascular smooth muscle cells in a dose- and time-dependent manner within the concentration range of 7.5–120 μg/mL, and the inhibition rate of Artesunate can be as high as 89.49 % at a concentration of 120 μg/mL after acting for 72 hours; vascular smooth muscle cells show a typical apoptosis peak due to the effects of higher concentration of Artesunate. Compared with the control group, the higher-concentration group shows major variability, Ac-DEVD-CHO, however, can significantly decrease this induction; it has been detected by Western Blot that Artesunate can induce caspase-3 activity dramatically in vascular smooth muscle cells, but this activation may be remarkably inhibited by Ac-DEVD-CHO.

Abstract:The present study was aimed to investigate the effect of dihydroartemisinin on the colon cancer cell proliferation and apoptosis. The results from MTT assay revealed a concentration and time dependent relation between the inhibition of SW 948 cell viability and dihydroartemisinin addition. The viability of SW 948 cells was reduced to 45 and 24% on treatment with 30 and 50µM, respectively concentrations of dihydroartemisinin after 48h. Morphological examination of SW 948 cells showed attainment of rounded shape and cluster formation on treatment with dihydroartemisinin. Western blot analysis showed a significant increase in the activation of caspase-3 and expression of cleaved PARP by dihydroartemisinin treatment. The activation of PPARγ was increased significantly in SW 948 cells by treatment with dihydroartemisinin. Compared to control, the migration potential of SW 948 cells was reduced significantly (p<0.005) and the expression levels of MMP-2 and -9 inhibited by dihydroartemisinin at 50µM concentration. In the dihydroartemisinin treatment group colon tumor formation was significantly inhibited on treatment with 20mg/kg doses of dihydroartemisinin after 30days. Therefore, dihydroartemisinin inhibits colon cancer growth by inducing apoptosis and increasing the expression of PPARγ. Thus dihydroartemisinin can be used for the treatment of colon cancer.

Abstract:OBJECTIVE:To evaluate the effects of artemisinin on proliferation, cell cycle and apoptosis of gallbladder cancer cells. METHODS:Gallbladder carcinoma cell lines(GBC-SD and NOZ)were cultured in vitro. The effects of artemisinin in different concentration on proliferation of the two cell lines in vitro were examined using MTT assay. The cell cycle distribution of GBC-SD and NOZ cells 24 h after treatments with artemisinin(20 μmol/L) were examined using flow cytometry. The apoptosis of GBC-SD and NOZ cells 24 h after treatments with artemisinin (20 μmol/L) were examined using Annexin V/PI staining.The expressions of p-ERK1/2, CDK4, cyclin D1, p16, cytochrome C and caspase-3 were examined by Western blot assay. t-test and one way ANOVA were used to evaluate the differences between two groups and more than two groups, respectively. RESULTS:The cell proliferation was significantly inhibited by artemisinin, the IC50 of artemisinin against GBC-SD and NOZ cells were 14.05 μmol/L and 12.42 μmol/L, respectively.Artemisinin induced cycle arrest, and G1 population of GBC-SD and NOZ cells increased to 74.60% and 78.86%. Cell apoptosis and apoptotic population of GBC-SD and NOZ cells were increased to 15.67% and 16.51% after dealt with artemisinin, respectively. In addition, expression of p16 was increased, and expressions of p-ERK1/2, CDK4 and cyclin D1 were down-regulated by artemisinin(all P<0.05). Cytochrome C was released from mitochondria to cytoplasm leading to the activation of caspase-3 and PARP after dealt with artemisinin(P<0.05). CONCLUSION:The inhibition effect of artemisinin on the proliferation gallbladder cancer cells is accompanied by down-regulation of ERK1/2 signaling pathway, G1 phase arrest and triggering caspase-3-mediate apoptosis.

Abstract:BACKGROUND: Activation of hepatic stellate cells (HSCs) plays an important role in the development of cirrhosis through the increased production of collagen. p53, the "guardian of the genome", is a transcription factor that can bind to promoter regions of hundreds of genes where it either activates or suppresses gene expression. Thereby, p53 serves as a tumor suppressor by inducing cell cycle arrest, apoptosis, senescence and DNA repair. Artesunate is a derivative of Artemisinin, Scholars had found it had more extensive pharmacological effects past 10 years. However, little is known about the expression of p53 in the effects of Artesunate on induction of apoptosis and inhibition of proliferation in rat HSCs. METHODOLOGY/PRINCIPAL FINDINGS: Isolated and cultured rat primary HSCs in the flask for 10 days to make cells activated. HSCs were divided into two groups: experimental groups and control groups, experimental groups included with various concentrations of Artesunate (125, 150, 175, 200, 225 µmol/L) for 24, 48 and 72 hours. Analysis of MTT revealed that activated HSCs treated with various concentrations of Artesunate (150-225 µmol/L) were inhibited on dose and time-effect relationships; Concentration of hydroxyproline in supernatant was detected by digestive method; Analysis of flow cytometry demonstrated that Artesunate could arrest cell cycle in G1 and induce apoptosis; The nuclear morphological changes in apoptotic cells were evaluated with DNA staining by Hoechst 33258 dye; The expression of p53 were up-regulated showed by western blotting and RT-PCR. CONCLUSION: Artesunate could inhibit HSCs proliferation in dose-dependent and time-dependent manners in vitro through increase the expression of p53.

[Artesunate Suppresses Cell Proliferation of THP-1 Cells by Inducing Apoptosis].

Abstract:OBJECTIVE:To investigate whether Artesunate(ART) can inhibit the proliferation of THP-1 cells and to explore the potential mechanism of its anti-leukemia effect.METHODS:THP-1 cells were treated with 5 concentrations of Artesunate for 24 h, 48 h or 72 h. The viability of cells was detected with CCK-8 assay, apoptosis was assessed by using flow cytometry, and the STAT3, Caspase3 and Caspase8 protein levels were measured with Western blot .RESULTS:Compared with the control group, ART significantly inhibited the proliferation of THP-1 cells in a dose-dependent manner (r=0.9829, P<0.05). ART also increased the apoptosis of THP-1 cells. The results of Western blot showed that after treated with ART, the STAT3 protein expression in THP-1 cells was significantly down-regulated (P<0.01), and the expressions of Caspase3, cleaved Caspase3 and Caspase8 proteins were up-regulated(P<0.01).CONCLUSION:Artesunate can inhibit the proliferation of THP-1 cells, which may relate with the down-regulation of STAT3 expression and the activation of Capase3 and Caspase8.

Abstract:Objective: To observe the effect of Dihydroartemisinin on the proliferation and apoptosis in human colon carcinoma cell line SW480 in vitro and investigate the mechanism.Methods: The inhibition of proliferation of SW480 cells was determined by MTT test.Morphologic changes were observed under optical microscope.Distribution of cell cycle and apoptosis was analyzed using flow cytometry through PI and AnnexinV/PI double-1abeled staining.The expression of cell apoptosis-associated gene protein was detected with immunohistochemical technique.Results: Dihydroartemisinin could inhibit the proliferation of SW480 cells and the inhibition was depended on the exposure dose and time.After treated with Dihydroartemisinin for 48 h,apoptotic cells and morphologic changes could be found under optical microscope.The analysis of cell cycle indicated that Dihydroartemisinin blocked cells at G0/G1 phases,showed an increase of G0/G1 phase and a decrease of S phase.The expression of survivin was down-regulated,and caspase-3 expression was up-regulated.Conclusion: Dihydroartemisinin could inhibit the proliferation of the colon carcinoma cell line SW480 by regulating the cell cycle through arresting cells at G0/G1 phase and inducing apoptosis.The anti-tumor effects of Dihydroartemisinin may be related to the down-regulation of the expression of survivin protein and up-regulation of caspase-3.

Abstract:To investigate the effects of an allelochemical artemisinin extracted from Artemisia annua (A. annua) on cell growth, death mode, and microcystin-LR (MC-LR) changes of Microcystis aeruginosa (M. aeruginosa), a series of morphological and biochemical characteristics were studied. The results showed that artemisinin could inhibit the growth of M. aeruginosa and reduce the content of phycobiliprotein. Under the allelopathy of artemisinin, algae cells deformed due to swelling, which caused cell membranes to rupture and cell contents to leak. FDA/PI double-staining results showed that 15.10–94.90% of algae cells experienced the death mode of necrosis-like. Moreover, there were 8.35–14.50% of algae cells undergoing programmed cell death, but their caspase-3-like protease activity remained unchanged, which may mean that algae cells were not experiencing caspase-dependent apoptosis under artemisinin stress. Attacked by artemisinin directly, both intracellular and extracellular MC-LR increased sharply with the upregulation of mcyB, mcyD, and mcyH. The upregulation multiple of mcyH suggested that M. aeruginosa could accelerate transportation of algal toxin under adverse conditions of artemisinin. Artemisinin not only can inhibit the growth of M. aeruginosa but it also causes the accelerated release and increase of microcystin-LR. These imply that the application of artemisinin should be reconsidered in practical water bodies.

Abstract:To investigate the effect and mechanism of 1,25-dihydroxyvitamin D3 on Caspase-3 expression of rat mesangial proliferative glomererulonephritis.Methods:Clean SD rats were randomly divided into control group and the glomerulonephritis model groups,which was subdivided into the glomerulonephritis group and 1,25-(OH)2D3 group.Immune-histochemistry was used to detect the expression of Caspase-3 in glomerular and renal interstitium,to compare the differences between the groups.Results:1)There was no expression of Caspase-3 in renal glomeruli,in the glomerulonephritis group,Caspase-3 presented expression in Mesangium,P0.01;compared with the control group,the expression of Caspase-3 in 1,25-(OH)2D3 group was improved,but the difference was not statistically significant,P0.05;2)Caspase-3 in normal renal tubular epithelial cells had a small amount of expression.compared with the control group,Caspase-3 of the glomerulonephritis group in renal proximal tubules and distal tubules with time continuation of expression was significantly increased,and the difference was statistically significant(P0.01);Given to the intervention of 1,25-(OH)2D3,after three days,Caspase-3 expression became higher than the glomerulonephritis group,but after 7 days and 14 days treatment,the expression was significantly lower than nephritis group,with statistically significant difference(P0.05).1,25-(OH)2D3 may participate in the regulation of caspase-3,then enhance inflammation recoveredy;but it may reduce renal tubule epithelial cells apoptosis in rat MsPGN model,prevent renal fibrosis,and delay the progression of rat renal glomerular disease.

Abstract:Abstract Background Bone marrow-derived mesenchymal stem cell (BMSC) transplantation is one of the new therapeutic strategies for treating ischemic brain and heart tissues. However, the poor survival rate of transplanted BMSCs in ischemic tissue, due to high levels of reactive oxygen species (ROS), limits the therapeutic efficacy of this approach. Considering that BMSC survival may greatly enhance the effectiveness of transplantation therapy, development of effective therapeutics capable of mitigating oxidative stress-induced BMSC apoptosis is an important unmet clinical need. Methods BMSCs were isolated from the 4-week-old male Sprague Dawley rats by whole bone marrow adherent culturing, and the characteristics were verified by morphology, immunophenotype, adipogenic, and osteogenic differentiation potential. BMSCs were pretreated with artemisinin, and H 2 O 2 was used to induce apoptosis. Cell viability was detected by MTT, FACS, LDH, and Hoechst 33342 staining assays. Mitochondrial membrane potential (ΔΨm) was measured by JC-1 assay. The apoptosis was analyzed by Annexin V-FITC/PI and Caspase 3 Activity Assay kits. ROS level was evaluated by using CellROX® Deep Red Reagent. SOD, CAT, and GPx enzymatic activities were assessed separately using Cu/Zn-SOD and Mn-SOD Assay Kit with WST-8, Catalase Assay Kit, and Total Glutathione Peroxidase Assay Kit. The effects of artemisinin on protein expression of BMSCs including p-Erk1/2, t-Erk1/2, p-c-Raf, p-p90 rsk , p-CREB, BCL-2, Bax, p-Akt, t-Akt, β-actin, and GAPDH were measured by western blotting. Results We characterized for the first time the protective effect of artemisinin, an anti-malaria drug, using oxidative stress-induced apoptosis in vitro, in rat BMSC cultures. We found that artemisinin, at clinically relevant concentrations, improved BMSC survival by reduction of ROS production, increase of antioxidant enzyme activities including SOD, CAT, and GPx, in correlation with decreased Caspase 3 activation, lactate dehydrogenase (LDH) release and apoptosis, all induced by H 2 O 2 . Artemisinin significantly increased extracellular-signal-regulated kinase 1/2 (Erk1/2) phosphorylation, in a concentration- and time-dependent manner. PD98059, the specific inhibitor of the Erk1/2 pathway, blocked Erk1/2 phosphorylation and artemisinin protection. Similarly, decreased expression of Erk1/2 by siRNA attenuated the protective effect of artemisinin. Additionally, when the upstream activator KRAS was knocked down by siRNA, the protective effect of artemisinin was also blocked. These data strongly indicated the involvement of the Erk1/2 pathway. Consistent with this hypothesis, artemisinin increased the phosphorylation of Erk1/2 upstream kinases proto-oncogene c-RAF serine/threonine-protein kinase (c-Raf) and of Erk1/2 downstream targets p90 ribosomal s6 kinase (p90 rsk ) and cAMP response element binding protein (CREB). In addition, we found that the expression of anti-apoptotic protein B cell lymphoma 2 protein (BcL-2) was also upregulated by artemisinin. Conclusion These studies demonstrate the proof of concept of artemisinin therapeutic potential to improve survival in vitro of BMSCs exposed to ROS-induced apoptosis and suggest that artemisinin-mediated protection occurs via the activation of c-Raf-Erk1/2-p90 rsk -CREB signaling pathway.

Abstract:AIM: To investigate the effects of artesunate on proliferation and apoptosis in human hepatocelluar carcinoma cell line HepG2 and to study the sensitizing effect of artesunate on HepG2 cells to chemotherapeutic drugs.METHODS: The proliferation of HepG2 cells was determined by the assay of cell counting kit-8(CCK-8) and the colony formation test.The morphology of HepG2 cells with Hoechst 33258 staining was observed under fluorescent microscope.Annexin V/propidium iodide(PI) was used to analyze the apoptosis and the cell cycle.The sensitizing effects of artesunate on HepG2 cells to chemotherapeutic drugs [5-fluorouracil(5-FU),carboplatin and epirubicin] were determined by CCK-8 assay.RESULTS: When treated with artesunate for 48 h,the proliferation of HepG2 cells was significantly inhibited in a dose-dependent manner.The IC50 was 19.2 μmol/L.Compared with the control cells,the colony formation of HepG2 cells treated with artesunate for 7 days was significantly inhibited.The nuclear fragmentation,karyopyknosis,chromosomal condensation,cell shrinkage,and attachment loss in HepG2 cells treated with artesunate were observed.The cells in G2 phase increased obviously,and the percentages of hypodiploid cells and early apoptotic rates were significantly higher in artesunate treatment groups than those in control group.The IC50 of 5-FU,carboplatin and epirubicin combined with artesunate was 3.33,2.02 and 1.71 times sensitized as compared with control group,respectively.CONCLUSION: Artesunate effectively inhibits proliferation and induces apoptosis of HepG2 cells.Aresunate also sensitizes HepG2 cells to chemotherapeutic drugs.

Abstract:Objective: To investigate the inhibition of artemisinin on acute leukemia K562 cell proliferation. Methods: K562 cells were cultured in vitro together with a series of artemisinin concentrations. The inhibitory rate of the cellular growth was measured using MTT assay. The cell apoptosis was assessed by flow cytometry (FCM) and Hoechst 33258 fluorescence staining. The telomerase activities before and after the cell apoptosis were assayed using TRAP-PCR-ELISA. Results: Axtemisinin had a significant inhibition of the telomerase activity and the cell growth in both time- and dose-dependent manner. Artemisinin resulted in the cell apoptosis. Conclusion: Artemisinin has antiproliferation and apoptosis inducing effects on K562 cells by reducing the telomerase activity.

Abstract:Objective To investigate the apoptosis and cell growth inhibition in vitro effects of artesunate(ART) on human gastric adenocarcinoma MKN45 cell line,and to explore the underlying mechanism of this action.Methods After MKN45 cell lines in vitro were treated with various concentrations of ART,cell growth inhibition ratio and apoptosis ratio were determined by the methyl thiazolyl tetrazolium(MTT) method and flow cytometry assay,respectively.The expression levels of apoptosis related proteins(Bcl-2,Bax,caspase-9 and caspase-3) after MKN45 cell lines treated with various concentrations of ART were detected by Western Blotting.Results The proliferation of MKN45 cells were significantly inhibited after treated with ART for 48 hours and this inhibitory action was dose-dependent(P0.05).The value of IC50 was 13.41 μmol·L-1.The proliferation and survival ability of MKN45 cells were inhibited by 13.41 μmol·L-1ART,and this inhibitory action was positive correlated with time-and dose-dependent(P0.05).With the increase of concentration of ART,the apoptosis ratio of MKN45 cell was also elevated(P0.05).ART could down-regulate the expression level of Bcl-2 and up-regulate Bax,caspase-9 and caspase-3 protein levels.This regulation effect was obviously enhanced when the concentration of ART was increased.Conclusion ART can inhibit the growth and induce apoptosis of human qastric adenocarcinoma MKN45 cell line in vitro.

Abstract:Objective To investigate the effects and the underlying mechanisms of artesunate(ART)on the growth and apoptosis of epidermal carcinoma cells.Methods The proliferation,cell cycle and apoptosis of A431 cells(a cell line of malignant epidermal carcinoma)were detected with MTT assay and flow cytometer(FCM).HaCaT cells(an immortalized keratinocyte cell line)served as normal control,and diamminedichloroplatinum(DDP)as drug positive control.Deferoxamine mesylate(DFOM)was employed to block the effect of ART.Results ART showed obviously inhibitory effect on the proliferation of A431 cells,and had lesser toxicant to HaCaT cells compared with DDP.When 60 μmol/L ART(IC50)was used to treat A431 cells for 48 h,the cells was arrested in S phase [(67.60±4.12)%,P<0.05];and their apoptotic rate was increased to(19.87±0.03)%(P<0.01).However,when the A431 cells was pretreated with 60 μmol/L DFOM for 4 h followed with the treatment as above mentioned,there was no change in cell cycle distribution,and the apoptotic rate was(7.37±0.02)%(P<0.01).Conclusion ART significantly suppresses the proliferation of A431 cells by arresting the cells in S phase and inducing cell apoptosis in a dose-dependent manner.Fe2+ might mediate the apoptosis of A431 cells induced by ART.ART has lower toxicant to A431 cells than DDP.

Abstract:Objective To investigate the role and mechanism of dihydroartemisinin in inducing apoptosis in glioma GL261 cells.Methods GL261 cells were treated with different concentrations of dihydroartemisinin(0,30,60 and 90 μmol/L).The morphological changes of GL261 were observed by an inverted microscope,and the viability of GL261 cells was measured by CCK-8 assay.Annexin V PI assay was used to study the percentage of apoptotic GL261 cells.Laser scanning confocal microscopy was applied to examine the expression and subcellular localization of activated caspase-3 in the GL261 cells,and Western blotting was carried out to detect the expression of activated caspase-3 and Bcl-xL.Results Dihydroartemisinin reduced the viability and induced apoptosis of GL261 cells in a dose-dependent manner(P<0.05).Treatment with 30 μmol/L dihydroartemisinin for 12 h could induce the activation of caspase-3 and co-localization with the nucleus in GL261 cells.Dihydroartemisinin could up-regulate the level of activated caspase-3 and down-regulate the level of Bcl-xL in GL261 cells in a dose-dependent manner(P<0.05).Conclusion Dihydroartemisinin induces apoptosis in GL261 cells.Anti-apoptotic protein Bcl-xL and the executor caspase-3 are involved in this process.
