Abstract:Brain arteriovenous malformations (BAVM) have high matrix metalloproteinase-9 (MMP-9) expression, the source of which is unclear. We hypothesized MMP-9 production might be due to inflammation in BAVM. Compared to control brain tissues (n=5), BAVM tissue (n=139) had a higher expression (by ELISA) of myeloperoxidase (MPO) (193 +/- 189 vs. 6 +/- 3, ng/mg, P <.001), MMP-9 (28 +/- 32 vs. 0.7 +/- 0.6, ng/mg, P <.001), and IL-6 (102 +/- 218 vs. 0.1 +/- 0.1, pg/mg, P <.001), but not eNOS (114 +/- 87 vs. 65 +/- 9, pg/mg, P=.09). MMP-9 expression in BAVM highly correlated with myeloperoxidase (R-2=.76, P <.001), as well as with IL-6 (R-2=.32, P <.001). In contrast, MMP-9 in BAVM poorly correlated with the endothelial marker, eNOS (R-2=.03, P=.05), and CD31 (R-2=.004, P=.57). Compared to non-embolized patients (n=46), patients with pre-operative embolization (n=93) had higher levels of myeloperoxidase (236 +/- 205 vs. 106 +/- 108, ng/mg, P <.001) and MMP-9 (33 +/- 35 vs. 16 +/- 20, ng/mg, P <.001), however the correlation between MMP-9 and myeloperoxidase was equally strong for both groups (R-2=.69, n=93, P <.001, for both). MMP-9 expression correlated with the lipocalin-MMP-9 complex, suggesting neutrophils as the MMP-9 source. MPO co-localized with majority of MMP-9 signal by immunohistochemistry. Our data suggest that inflammation is a prominent feature of BAVM lesional phenotype, and neutrophils appear to be a major source of MMP-9 in these lesions.
Expression and Significance of Matrix Matalloproteinase and Tissue Inhibitor of Metalloproteinase in Human Astrocytomas

Abstract:Objective:To explore the expression and significance of matrix metalloproteinase and tissue inhibitor of metalloproteinase in astrocytoma of different grades and the relationship with each other.Methods:Fifty seven cases of astrocytomas were adopted in this experiment which included 27 cases of low grade astrocytoma(Ⅰ astrocytoma plus Ⅱastrocytoma)and 30 cases of high grade astrocytoma(Ⅲ astrocytoma plus Ⅳ astrocytoma),and eight cases of normal brain tissues which were taken from internal decompression of brain trauma,followed by immunohistochemical staining by means of SP method(streptavidin peroxidase method) and the LI(Labeling index) can be calculated. All datas were analyzed with SSPS with windows software.Results:LI of MMPs had significant difference between low grade astrocytoma and high grade astrocytoma(P0.01). LI of TIMPs had no difference(P=0.391).Conclusion:The expression of MMPs is closely related to the malignant degree of astrocytoma. The unbalance of MMPs TIMPs is aslo closely related to the malignant degree of pathological grade of astrocytoma.

Abstract:Background and Purpose-Excessive degradation of the vascular matrix by matrix metalloproteinases (MMPs) can lead to structural instability of vessels. In this study we examined the expression of MMPs and tissue inhibitors of metalloproteinases (TIMPs) in brain arteriovenous malformations (BAVMs). Methods-We performed gelatin zymography for MMPs and Western blot for MMP-9, MMP-2, TIMP-1, TIMP-2, TIMP-3, and TIMP-4. MMP-9 expression was localized by immunohistochemistry. Results-We analyzed 37 BAVM specimens and 9 control brain specimens from epilepsy surgery. Thirty-two BAVM patients had embolization treatment before resection. Eighteen BAVM patients had a history of hemorrhage from BAVMs. Neither MMP-2 nor TIMP-2 was detected in BAVMs or control brain specimens. Compared with control brain samples, BAVM samples had higher levels of total MMP-9, active MMP-9, pro-MMP-9, TIMP-1, and TIMP-3. TIMP-4 levels were higher in the control brain than in BAVM specimens. MMP-9 was localized to the endothelial cell/peri-endothelial cell layer and infiltrating neutrophils of BAVMs. BAVMs with venous stenosis greater than or equal to50% had higher expression of MMP-9 than BAVMs with venous stenosis <50%. There was no apparent association between total MMP-9, pro-MMP-9, or active MMP-9 levels and (1) feeding artery pressure, (2) pattern of draining vein (exclusively deep venous drainage versus any superficial drainage), and (3) BAVM size. Conclusions-We found increased levels of MMP-9 and TIMPs in BAVMs. Abnormal balance of MMP-9 and TIMPs may contribute to vascular instability of BAVMs.

Abstract:Astrocytomas represent the majority of cerebral gliomas. Studies show that the anti-inflammatory protein Annexin-A1 (ANXA1) is associated with the tumor invasion process and that its actions can be mediated by the receptor for formylated peptides (FPR). Therefore, we evaluated the expression of ANXA1, the receptor FPR2 and matrix metalloproteinases 2 and 9 (MMP-2 and MMP-9) in brain astrocytomas. Detection of proteins was performed in sections of diffuse astrocytomas (grade II), anaplastic astrocytomas (grade III) and glioblastomas (GBM, grade IV) and quantifications were made by densitometry. Our analyses showed increased expression of ANXA1 in astrocytomas of all grades, but especially in GBM. The expression of FPR2 is similar to that found for ANXA1, being higher in GBM. Immunostaining for MMPs is also stronger as the degree of malignancy increases, especially with respect to MMP-9. The positive correlation between ANXA1/FPR2 and ANXA1/MMP-9 was observed in all tumors studied. The data indicate the possible action of ANXA1 and FPR2 on the development and progression of astrocytomas, related to increased expression of MMP-9. Thereby, ANXA1 and FPR2 are involved in the biology and malignancy of diffuse astrocytic tumors.

Abstract:Objective Meningioma of intracranial tumors is one of the common benign tumor of neurosurgery.The study has in contrast of the different levels of meningiomas magnetic resonance imaging (MRI) features matrix metalloproteinase (MMP)-9 expression,further study of the different meningioma MRI and molecular pathological basis.Methods 60 patients after surgical treatment had been confirmed by pathology.34 cases of meningioma grade Ⅰ,17 cases of meningiomas grade Ⅱ,9 cases of meningio mas grade Ⅲ.Using superconducting 3.0 T magnetic resonance imaging system underwent MRI plain and enhanced scan.Using of immunohistochemistry to detect meningiomas of MMP-9 expression.Results Positive expression of 34 cases of Word Health Organization (WHO) grade Ⅰ meningiomas was 8.82％,31 cases were negative,2 cases had weak expression,and 1 case had neutral expression;Positive expres sion of 17 cases of WHO grade Ⅱ meningioma was 41.00％,10 cases were negative,5 cases had weak expression,and 2 cases had neutral expression;Positive expression of 9 cases of WHO grade Ⅲ meningiomas was 100.00％,and 9 cases had strong expression.Immunohistochemistry results show:the expression of MMP-9 significant difference (x2 =68.880,P =0.001) in all levels of meningiomas.meningiomas expression of grade Ⅲ was significantly higher statistically significant than grade Ⅱ (x2 =26.000,P =0.001),meningiomas expression of grade Ⅱ was significantly higher statistically significant than grade Ⅰ (x2 =7.550,P =0.023).Lowest expression in normal meningeal tissue,meningiomas expression of grade Ⅲ of MMP-9 expression in the highest,MMP-9 protein expression levels tended to increase with menin gioma level increasing.Conclusion Meningiomas expression of grade Ⅲ is the highest in expression of MMP-9.With meningioma level increases,MMP-9 expression corresponding increase.The meningioma high expression of MMP-9 promotes peritumoral brain edema formation,with a synergistic effect on tumor proliferation and infiltration.
Matrix Metalloproteinase-9 Inhibition Attenuates Vascular Endothelial Growth Factor-Induced Intracerebral Hemorrhage

Abstract:Background and Purpose - Human brain arteriovenous malformation tissue displays increased levels of vascular endothelial growth factor (VEGF) as well as matrix metalloproteinase (MMP)- 9, a tissue protease associated with various intracerebral hemorrhage (ICH). We hypothesized that increased MMP- 9 was associated with ICH induced by vascular endothelial growth factor hyperstimulation and that this effect could be attenuated by nonspecific MMP inhibition.Methods - We used a mouse model with adenoviral vector- mediated vascular endothelial growth factor transduction in the brain. The association of MMP- 9 expression and the brain tissue hemoglobin levels, an index of ICH, after stereotactic injection of adenoviral vector- mediated vascular endothelial growth factor into caudate putamen was assessed. A dose - response study with adenoviral vector- mediated vascular endothelial growth factor and a time course study at both 24 and 48 hours postinjection were performed. Effects of minocycline, a nonspecific MMP inhibitor, and pyrrolidine dithiocarbamate, an upstream regulator of MMPs, on MMP- 9 activity and thereby the degree of ICH were also tested.Results - Adenoviral vector- mediated vascular endothelial growth factor at the higher dose and at 48 hours induced MMP- 9 levels 6- fold (n = 6, P = 0.02) and increased brain tissue hemoglobin (43.4 +/- 11.5 versus 30.3 +/- 4.1 mu g/mg, n = 6, P = 0.003) compared with the adenoviral vector control. Immnunostaining was positive for MMP- 9 around the cerebral vessels and the hemorrhagic areas. Minocycline and pyrrolidine dithiocarbamate administration suppressed vascular endothelial growth factor- induced MMP- 9 activity (n = 6, P = 0.003 and P = 0.01, respectively) and the associated increases in hemoglobin levels (n = 5 - 6, P = 0.001 and P = 0.02, respectively).Conclusions - Vascular endothelial growth factor- induced ICH is associated with increased MMP- 9 expression. Suppression of MMP- 9 by minocycline or pyrrolidine dithiocarbamate attenuated ICH, suggesting the therapeutic potential of MMP inhibitors in cerebral vascular rupture.

Abstract:To explore the expression of matrix metalloproteinase 9 (MMP-9), type IV collagen (Col IV) and vascular endothelial growth factor (VEGF) in adamantinomatous craniopharyngioma (ACP) and analyze the correlation between the level of these markers and adamantimous craniopharyngiomas recurrence. Expressions of MMP-9, Col IV and VEGF were tested by immunohistochemistry (IHC) in 40 cases of ACP, including 24 cases of primary group and 16 cases of recurred group. The expression level of MMP-9 and VEGF in recurred group were significantly higher than primary group (93.7% vs. 41.7%, P < 0.05, 87.5% vs. 45.8%, P < 0.05, respectively). The expression of Col IV in the recurred group was significant different from the primary group (Z = −2.619, P < 0.05). MMP-9, Col IV and VEGF may be the potential specific bio-marker related to the recurrence of ACP.
Increased expression of serum Matrix Metalloproteinase-9 in patients with moyamoya disease

Abstract:Background: Moyamoya disease is a chronic occlusive cerebrovascular disease with unknown etiology characterized by an abnormal vascular network at the base of the brain, which can manifest both as ischemic stroke and as cerebral hemorrhage. It was also reported that the patients with moyamoya disease are more vulnerable to cerebral hyperperfusion such as postoperative hemorrhagic complication after extracranial-intracranial bypass surgery despite its low flow revascularization. However, the underlying mechanisms of its pathologic angiogenesis and the occurrence of hemorrhage are undetermined. Excessive degradation of the vascular matrix by MMPs, proteolytic enzymes that degrade all the components of extracellular matrix, can lead to instability of the vascular structure and can thereby cause bleeding. The MMPs also play an important role in tissue remodeling including angiogenesis in both physiologic and pathologic condition. Methods: We examined the serum levels of MMP-2 and MMP-9 in 16 cases with definitive moyamoya disease by enzyme-linked immunosorbent assay and compared them with those from healthy controls. Results: The serum MMP-9 level was significantly higher in moyamoya disease (40.18 ng/mL) than in healthy controls (13.75 ng/mL, P = .0372). There was no difference in serum MMP-2 level between moyamoya disease (646.65 ng/mL) and healthy control (677.60 ng/mL). Immunohistochemistry on the surgical specimens showed significant increase in MMP-9 expression within the arachnoid membrane of moyamoya disease. Conclusion: The increased expression of MMP-9 may contribute to pathologic angiogenesis and/or to the instability of the vascular structure and could thereby cause hemorrhage in moyamoya disease.
Matrix metalloproteinase 9: An emerging biomarker for classification of adherent vestibular schwannoma

Abstract:Background: The progression of vestibular schwannoma (VS) is intricately linked with interactions between schwannoma cells and the extracellular matrix. Surgical resection of VS is associated with substantial risks as tumors are adherent to the brainstem and cranial nerves. We evaluate the role of matrix metalloproteinase 9 (MMP9) in VS and explore its potential as a biomarker to classify adherent VS. Methods: Transcriptomic analysis of a murine schwannoma allograft model and immunohistochemical analysis of 17 human VS were performed. MMP9 abundance was assessed in mouse and human schwannoma cell lines. Transwell studies were performed to evaluate the effect of MMP9 on schwannoma invasion in vitro. Plasma biomarkers were identified from a multiplexed proteomic analysis in 45 prospective VS patients and validated in primary culture. The therapeutic efficacy of MMP9 inhibition was evaluated in a mouse schwannoma model. Results: MMP9 was the most highly upregulated protease in mouse schwannomas and was significantly enriched in adherent VS, particularly around tumor vasculature. High levels of MMP9 were found in plasma of patients with adherent VS. MMP9 outperformed clinical and radiographic variables to classify adherent VS with outstanding discriminatory ability. Human schwannoma cells secreted MMP9 in response to TNF-α which promoted cellular invasion and adhesion protein expression in vitro. Lastly, MMP9 inhibition decreased mouse schwannoma growth in vivo. Conclusions: We identify MMP9 as a preoperative biomarker to classify adherent VS. MMP9 may represent a new therapeutic target in adherent VS associated with poor surgical outcomes that lack other viable treatment options.
Relationship between the Expression of Matrix Metalloproteinases and Their Tissue Inhibitors in Patients with Brain Tumors

Abstract:Matrix metalloproteinases (MMPs) and their inhibitors (TIMPs) play critical roles in regulating processes associated with malignant behavior. These endopeptidases selectively degrade components of the extracellular matrix (ECM), growth factors, and their receptors, contributing to cancer cell invasiveness and migratory characteristics by disrupting the basal membrane. However, the expression profile and role of various matrix metalloproteinases remain unclear, and only a few studies have focused on differences between diagnoses of brain tumors. Using quantitative real-time PCR analysis, we identified the expression pattern of ECM modulators (n = 10) in biopsies from glioblastoma (GBM; n = 20), astrocytoma (AST; n = 9), and meningioma (MNG; n = 19) patients. We found eight deregulated genes in the glioblastoma group compared to the benign meningioma group, with only MMP9 (FC = 2.55; p = 0.09) and TIMP4 (7.28; p < 0.0001) upregulated in an aggressive form. The most substantial positive change in fold regulation for all tumors was detected in matrix metalloproteinase 2 (MNG = 30.9, AST = 4.28, and GBM = 4.12). Notably, we observed an influence of TIMP1, demonstrating a positive correlation with MMP8, MMP9, and MMP10 in tumor samples. Subsequently, we examined the protein levels of the investigated MMPs (n = 7) and TIMPs (n = 3) via immunodetection. We confirmed elevated levels of MMPs and TIMPs in GBM patients compared to meningiomas and astrocytomas. Even when correlating glioblastomas versus astrocytomas, we showed a significantly increased level of MMP1, MMP3, MMP13, and TIMP1. The identified metalloproteases may play a key role in the process of gliomagenesis and may represent potential targets for personalized therapy. However, as we have not confirmed the relationship between mRNA expression and protein levels in individual samples, it is therefore natural that the regulation of metalloproteases will be subject to several factors.

Coexpression of angiogenic factors in brain arteriovenous malformations.

Abstract:OBJECTIVE: Brain arteriovenous malformations (BAVMs) are structurally unstable blood vessels that display an angiogenic phenotype, possibly maintained by concerted effects of key angiogenic factors. Therefore, we hypothesized that there are close correlations among key angiogenic factors in BAVMs and that levels of key angiogenic factors are associated with BAVM clinical characteristics that are linked with vascular instability. METHODS: We measured the expression of angiopoietin-2 (Ang-2), matrix metalloproteinase-9, vascular endothelial growth factor (VEGF), and platelet-derived growth factor-AA and -BB by use of enzyme-linked immunosorbent assay in 27 BAVM surgical specimens. Tissues were also collected from 14 structurally normal brain specimens obtained during epilepsy surgery. RESULTS: Ang-2, VEGF, and matrix metalloproteinase-9 were highly expressed in BAVM samples (11.9 &PLUSMN; 15.5 ng/mg protein, 137 &PLUSMN; 102 pg/mg protein, and 379 &PLUSMN; 455 pg/mg protein, respectively). Platelet-derived growth factor-BB was detectable in 7 of 21 BAVM samples. There were close correlations between Ang-2 and VEGF (R-2 = 0.79). Higher Ang-2 and VEGF levels seemed to be associated with draining vein characteristics linked with BAVM hemorrhage but not with feeding artery pressure or BAVM size. In the structurally normal brain specimens, levels of Ang-2, VEGF, and matrix metalloproteinase-9 were low (0.8 &PLUSMN; 2.3 ng/mg protein, 38 &PLUSMN; 25 pg/mg protein, and 52 43 pg/mg protein, respectively). CONCLUSION: There were close correlations among angiogenic factors in BAVMs. Concerted effects of angiogenic factors may maintain the angiogenic phenotype in BAVMs and thereby determine the clinical course of BAVMs.

Abstract:Background The precise mechanisms responsible for the development and growth of intracranial arteriovenous malformations (AVMs) remain unclear. Osteopontin (OPN) is a phosphorylated glycoprotein with diverse functions. This study aimed to analyze the expression of OPN in human brain AVMs.Methods The AVM nidus was surgically obtained from patients with AVM, whereas control brain artery specimens were surgically obtained from patients with epilepsy. Reverse transcription-polymerase chain reaction (RT-PCR) was used to examine the expression of OPN mRNA in biopsy specimens. OPN protein expression was localized by immunohistochemistry. The statistical differences between different groups were assessed by two-way analysis of variance (ANOVA).Results We analyzed 36 brain AVM specimens and 8 control brain artery specimens. Eleven patients with brain AVM received embolization treatment, and five underwent gamma knife radiotherapy before resection. Nineteen patients with brain AVM had a history of hemorrhage from AVMs. The expression of OPN mRNA was significantly higher in AVMs than that in the control specimens (25.76 +/- 2.71 vs. 21.46 +/- 2.01, P <0.01). There was no statistically significant difference in the extent of OPN mRNA expression between the AVM group with and that without history of hemorrhage (26.13 +/- 2.45 vs. 25.34 +/- 2.99) or gamma knife radiotherapy (24.39 +/- 2.10 vs. 24.53 +/- 1.85). However, the difference between the AVM group with and that without embolization treatment history was statistically significant (24.39 +/- 2.10 vs. 28.80 +/- 1.13, P <0.01). Relationship between the expression of matrix metalloproteinase-9 and angiogenesis in glioma and its clinical significance

Abstract:Objective: To explore the role and significance of matrix metalloproteinase-9 (MMP-9) in angiogenesis through observing the relationship between the expression of MMP-9 and microvessel density (MVD) in glioma. Methods: The expressions of MMP-9 and CD34 in 10 cases of normal brain tissues and 58 cases of glioma (14 cases of grade I, 20 cases of grade II, 15 cases of grade III, and 9 cases of grade IV) were detected by immunohistochemical streptavidin-peroxidase technique. The positive cells of MMP-9 and the positive microvessels were examined under binocular light microscope. Results: The positive expression of MMP-9 in glioma was located in the tumor-cell cytoplast and endothelial cells. The positive rate of MMP-9 in glioma of grade I, II, III and IV was 42.9%, 65.0%, 86.7% and 88.9%, respectively. The expression of MMP-9 was obviously higher than that of normal brain tissues (P<0.01) and positively correlated with glioma malignancy (rs = 0.597, P<0.05). MVD was correlated with glioma malignancy (H = 47.865, P<0.05). The expression of MMP-9 was significantly correlated with MVD (rs = 0.897, P<0.01). Conclusion: The expressions of MMP-9 and MVD are correlated with glioma malignancy, which may be helpful in judging the malignancy, invasion and prognosis. MMP-9 plays an important role in angiogenesis of glioma and accelerates glioma malignancy development by promoting angiogenesis.

Abstract:Matrix metalloproteinases are a major group of enzymes that regulate cell-matrix composition. They are essential for pathological processes including artherosclerosis,tumor migration,imflammation and so on.MMP-9 plays a key role in cerebral vascular diseases.It can degrade the laminin and fibronectin which are the major components of the basal lamina,then break down the blood-brain barrier which is high permeability. It shows that MMP-9 is relevant to edema and hemorrhagic transformation of cerebral infarction.Its functional mechanism in cerebral infarction turns it into a target for therapy of acute ischemic stroke.

Abstract:Memory formation requires learning-based molecular and structural changes in neurons, whereas matrix metalloproteinase (MMP) 9 is involved in the synaptic plasticity by cleaving extracellular matrix proteins and, thus, is associated with learning processes in the mammalian brain. Because the mechanisms of MMP-9 transcription in the brain are poorly understood, this study aimed to elucidate regulation of MMP-9 gene expression in the mouse brain after fear learning. We show here that contextual fear conditioning markedly increases MMP-9 transcription, followed by enhanced enzymatic levels in the three major brain structures implicated in fear learning, i.e. the amygdala, hippocampus, and prefrontal cortex. To reveal the role of AP-1 transcription factor in MMP-9 gene expression, we have used reporter gene constructs with specifically mutated AP-1 gene promoter sites. The constructs were introduced into the medial prefrontal cortex of neonatal mouse pups by electroporation, and the regulation of MMP-9 transcription was studied after contextual fear conditioning in the adult animals. Specifically, -42/-50- and -478/-486-bp AP-1 binding motifs of the mouse MMP-9 promoter sequence have been found to play a major role in MMP-9 gene activation. Furthermore, increases in MMP-9 gene promoter binding by the AP-1 transcription factor proteins c-Fos and c-Jun have been demonstrated in all three brain structures under investigation. Hence, our results suggest that AP-1 acts as a positive regulator of MMP-9 transcription in the brain following fear learning.

Abstract:Matrix Metalloproteinases (MMPs), which are endopeptidase reliant on zinc, are low in embryonic tissues but increases in response to a variety of physiological stimulus and pathological stresses. Neuro-glial cells, endothelial cells, fibroblasts, and leucocytes secrete MMPs, which cleave extracellular matrix proteins in a time-dependent manner. MMPs affect synaptic plasticity and the development of short-term memory by controlling the size, shape, and excitatory synapses' function through the lateral diffusion of receptors. In addition, MMPs influence the Extracellular Matrix proteins in the Peri-Neuronal Net at the Neuro-glial interface, which aids in the establishment of long-term memory. Through modulating neuronal, and glial cells migration, differentiation, Neurogenesis, and survival, MMPs impact brain development in mammals. In adult brains, MMPs play a beneficial role in physiological plasticity, which includes learning, memory consolidation, social interaction, and complex behaviors, by proteolytically altering a wide variety of factors, including growth factors, cytokines, receptors, DNA repair enzymes, and matrix proteins. Additionally, stress, depression, addiction, hepatic encephalopathy, and stroke may all have negative effects on MMPs. In addition to their role in glioblastoma development, MMPs influence neurological diseases such as epilepsy, schizophrenia, autism spectrum disorder, brain damage, pain, neurodegeneration, and Alzheimer's and Parkinson's. To help shed light on the potential of MMPs as a therapeutic target for neurodegenerative diseases, this review summarizes their regulation, mode of action, and participation in brain physiological plasticity and pathological damage. Finally, by employing different MMP-based nanotools and inhibitors, MMPs may also be utilized to map the anatomical and functional connectome of the brain, analyze its secretome, and treat neurodegenerative illnesses.
Chronic cerebral hypoperfusion induces MMP-2 but not MMP-9 expression in the microglia and vascular endothelium of white matter

Abstract:White matter lesions are closely associated with cognitive impairment and motor dysfunction in the aged. To explore the pathophysiology of these lesions, the authors examined the expression of matrix metalloproteinase-2 (MMP-2) and MMP-9 in the white matter in a rat model of chronic cerebral hypoperfusion. After bilateral clipping of the common carotid arteries, myelin staining revealed demyelinating changes in the optic tract and the corpus callosum on day 7. Zymographic analyses indicated an increase in the level of MMP-2, but not MMP-9, after the hypoperfusion. Immunohistochemical analyses revealed the presence (most abundantly on day 3) of MMP-2-expressing activated microglia in the optic tract and corpus callosum. In contrast, the capillary endothelial cells expressed MMP-2 later. IgM-immunoreactive glial cells were absent in the sham-operated animals, but were present in the hypoperfused animals by day 3, reflecting the disrupted blood-brain barrier. These findings suggest that the main sources of the elevated MMP-2 were the microglia and the endothelium, and that these cells may contribute to the remodeling of the white matter myelin and microvascular beds in chronic cerebral hypoperfusion.
