Differentail expression of EphB1 mRNA and protein in colorectal cancer and its clinicopathologic significance

Abstract:The expression of EphB1 in colon cancer cell lines and colorectal cancer tisseus were detected and the relationship between the expression levels of EphB1 and significance of clinicopathology was analyzed.The methylation status of CpG island in promoter region was analyzed.In order to look for new target gene of the treatment of colorectal cancer,the role of EphB1 in tumorigenesis of colorectal cancer was evaluated.Using quantitative real-time reverse transcriptase-polymerase chain reaction(RT-PCR) and immunohistochemistry to detect expression of EphB1 in 5 colon cancer cell lines and 61 colorectal cancer specimens with each paired uninvolved normal mucosa.The methylation status of the EphB1 promoter-associated CpG island was assessed by methylation-specific polymerase chain reaction(MSP).The EphB1 transcript was differentially expressed in 5 colon cancer cell lines,HT29 was low,and SW480 was high.The expression of EphB1 mRNA and protein in colorectal carcinoma specimens paired with its normal mucosa were different respectively.Expression of the EphB1 transcript was under-expressed in 31.1%(19/61),and over-expressed in 52.5%(32/61) of the tested samples correspondingly.There was significant difference between EphB1 mRNA and tumors histological differentiation(P=0.008).EphB1 staining was detected in the normal mucosa and some of the cancer cells.Down-expression was in 62.3%(38/61),and up-expression was in 24.6%(15/61).There were significant difference between EphB1 protein and tumors histology(P=0.033),invision depth(P=0.038),sex(P=0.028),and tumor location(P=0.039).No methylation of CpG island was found in DNA samples with under-expression of EphB1 mRNA using MSP.These results show that EphB1 expression may play a tumorsuppression role in determining the tumorigenesis and development in colorectal cancer because of its down-expression was association with the pathological differentiation and invision depth.
