Abstract:Objective To compare the frequencies of polymorphisms of GSTM1, GSTT1, and GSTP1 in Chinese Uygur with those in Chinese Han. Methods GSTM1 and GSTT1 polymorphisms were analyzed by a polymerase chain reaction(PCR)-multiplex procedure, whereas GSTP1 polymorphism was analyzed by PCR-restriction fragment length polymorphism. Results The frequency of GSTM1 null genotype was 56.1% in Chinese Han and 53.2% in Chinese Uygur, respectively. Whilst the frequency of GSTT1 null genotype in Chinese Han (50.0%) was higher than that of Chinese Uygur (26.6 %). The genotype distribution of GSTP1 was 60.7% I/I, 35.2% I/V and 4.1% V/V in Chinese Han and 51.3% I/I, 40.2% I/V and 8.4% V/V in Chinese Uygur, respectively. Conclusion There is marked ethnic difference in the mutant frequencies of GSTT1 and GSTP1, but not GSTM1, between Chinese Uygur and Chinese Han.

Abstract:Background: The glutathione S-transferase (GST) superfamily comprises multiple isozymes with compelling evidence of functional polymorphisms in various ethnic groups. All these mutations, in particular those in class mu, pi and theta GST, are likely to contribute to interindividual differences in responses to xenobiotics including response to chemotherapy and associated with altered disease. The frequency of common GST mutations in Uygur Chinese is unknown. We investigated the common mutations of GSTM1, GSTT1, and GSTP1 in Uygur (N = 154) Chinese and compare with Han Chinese (N= 196).Method: GSTM1 and GSTT1 polymorphisms were analyzed by multiplexed PCR, and GSTP1 polymorphism was detected by PCR-based restriction fragment length polymorphism (RFLP) analysis.Results: GSTM1 null genotype was found in 53.2% Uygur Chinese, which was close to that in Han Chinese (56.1%) (P=0.592). A significantly lower frequency (P<0.05) of GSTT1 null genotype in Uygur Chinese (26.6%) was observed compared with Han Chinese (50.0%). Uygur Chinese exhibited a GSTP1 genotype distribution of 51.3% I/I, 40.2% I/V and 8.4% V/V, which was different from that in Han Chinese (60.7% I/I 35.2% I/V and 4.1% V/V).Conclusions: There is marked ethnic difference in the frequency of common GSTT1 and GSTP1 mutation, but not GSTM1 mutation, between Uygur and Han Chinese. However, the results from the studies regarding the association between GSTP1 Ile105Val polymorphism and lung cancer risk in the Chinese population have been inconsistent. Thus, we conducted a meta-analysis to investigate the association. Published literature from PubMed, Chinese Biomedical Literature Database, Chinese Wanfang Data, and Chinese National Knowledge Infrastructure databases were searched for eligible publications. Pooled odds ratios (ORs) with 95 % confidence intervals (CIs) were calculated using random or fixed effect model. Ten studies (1,506 cases/1,714 controls) were included in the meta-analysis. The results suggested that GSTP1 Ile105Val polymorphism was marginally associated with lung cancer risk in the Chinese Han population under a multiplicative model (G vs. A, odds ratio (OR) = 1.22, 95 % confidence interval = 1.02–1.46), under a homogeneous codominant model (GG vs. AA, OR = 1.67, 95 % CI = 1.14–2.45), under a heterogeneous codominant model (GA vs. AA, OR = 1.15, 95 % CI = 0.98–1.35), under a dominant model (GG + GA vs. AA, OR = 1.21, 95 % CI = 1.04–1.39), and under a recessive model (GG vs. GA + AA, OR = 1.59, 95 % CI = 1.09–2.31), respectively. Moreover, after adjusted for age, gender, and smoking status, the significant association under dominant model remained (OR = 1.27, 95 % CI = 1.07–1.51). This meta-analysis suggested that there might be an association between GSTP1 Ile105Val polymorphism and lung cancer in the Chinese Han population.

Glutathione S-Transferase Polymorphism in Han Nationality in Shanghai

Abstract:Objective In order to develop a primary exploration of the relationship between glutathione S-transferase (GST) polymorphism and tumor susceptibility or therapeutic correlation in Han nationality, the genetic polymorphism of GST in healthy Han nationality of Shanghai was investigated and the candidate's single nucleotide polymorphism (SNP) was screened. Methods Automatic sequencing with labelled fluorescence was used to screen the SNPs of GSTT1 and GSTM1 in 20 healthy Han people. Results Point mutation was found at 86 057 site between exon 4 and exon 3 of GSTT1 in 4 cases, with guanine G substituted by adenine A. It may be a new candidate's SNP of GSTT1 after compared with genebank's SNP database. G&A heterozygotes could be found at 793 site and 921 site of exon 5 in all cases. Many candidate's SNPs were discovered in all 8 exons of GSTM1 and most of them were heterozygotes. 40% of the examinees showed adenine A deletion in exon 2. All examinees were A&G heterozygotes at 1 383 site and C&G heterozygotes at 1 385 site. At 101 site, 60% of the people examined showed A&T heterozygotes and 40% of the people had adenine A homozygotes. We found deletion of many base pairs or short fragments after 190 bp of exon 2. Conclusions The genetic polymorphism of GST varies greatly in healthy Han people in Shanghai. It remains to be discussed further whether the polymorphisms only occurred in Han nationals and correlated with tumor susceptibility and whether these possible candidate's SNPs could alter amino acid code resulting in variation of the primary structure of protein and alteration of GST acitivity.
[Study on the Genetic Polymorphisms of Human Glutathione-S-transferase A1 in Hakka Population in South China].

Abstract:OBJECTIVE:Glutathione-S-transferase A1 (GSTA1) is one of the major phase II detoxification enzymes in the cytosol, which genetic polymorphisms distribution is different in different ethnic, national and regional population. Up to now, GSTA1 genetic polymorphisms has been rarely reported in China. The purpose of the study was to investigate the distribution of genetic polymorphisms of human GSTA1 in Hakka population in South China.METHODS:Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) were used to identify the genotypes of GSTA1 gene and the data were analyzed with SPSS10.0 software.RESULTS:The GSTA1 genetic polymorphisms were detected in 480 samples. The frequency of GSTA1 * A/ * A,GSTA1 * A/ * B and GSTA1 * B/ * B were 77.1%, 21.7% and 1.2% respectively. And the GSTA1 genetic polymorphism distribution was in accordance with the Hardy-Weinberg equilibrium rule. There were no difference in the GSTA1 genetic polymorphisms among the different groups of age or gender. Logistic regression analysis showed that there were no association between the GSTA1 genetic polymorphism and family history of hypertension, coronary heart disease, stroke, lung cancer and nasopharyngeal cancer, et al.CONCLUSION:The GSTA1 gene existed polymorphism among Hakka in South China.

Abstract:Over 50% prescribed drugs are metabolised by cytochrome P450 3A (CYP3A) and glutathione S-transferase pi (GSTP1) adds a glutathione to the oxidative products by CYP3A, which increases the hydrophilic property of metabolites and facilitates the excretion. Single nucleotide polymorphisms (SNPs) of CYP3A and GSTP1 show a diverse allele and genotype frequencies distribution among the world populations. The present study aimed to investigate the genotype and allele frequency distribution patterns of CYP3A4, CYP3A5, CYP3A7 and GSTP1 polymorphisms among healthy participants in mainland Tibetan, Mongolian, Uyghur, and Han Chinese populations. Blood samples were collected from 842 unrelated healthy subjects (323 Tibetan, 134 Mongolian, 162 Uyghur, and 223 Han) for genotyping analysis. Variant allele frequencies of CYP3A4 rs2242480, CYP3A5 rs776746, CYP3A7 rs2257401, and GSTP1 Ile105Val were observed in Han (0.253, 0.686, 0.312 and 0.188), Tibetan (0.186, 0.819, 0.192 and 0.173), Mongolian (0.198, 0.784, 0.228 and 0.235) and Uyghur (0.179, 0.858, 0.182 and 0.250) respectively. The allele frequency of CYP3A7*1C in Uyghur (0.019) was higher than that in Tibetan (0.002, p < 0.01). There was a strong linkage disequilibrium between CYP3A4 rs2242480, CYP3A5 rs776746, and CYP3A7 rs2257401 among the four ethnic groups. The results might be useful for the precise medication in the Chinese populations.

Abstract:BACKGROUND AIM: To investigate the distribution of glutathione -S-transferase M1 and T1 (GSTM1, GSTT1)genetic polymorphisms in Cantonese people of Guangdong Province,and tentative analysis on the relationship between gene polymorphisms and the family history of some diseases. MATERIAL AND METHODS: The healthy population of 606 people in Xinxing county and Guangzhou city in Guangdong province were selected based on the intension of the study. Polymerase chain reaction (PCR)-2% agarose gel electrophoresis was applied to distinguish the genotype of the GSTM1 and GSTT1. RESULTS: The frequencies of homozygote for GSTM1(-/-) (n=597)and GSTT1(-/-)(n=579)were 56.8% and 42.1%, respectively. The frequency of homozygotes for both GSTM1(-/-) and GSTT1(-/-) was 22.8%(n=570). CONCLUSION: There was no linkage between the GSTM1 and GSTT1 genes. The difference of GSTM1 gene distribution in populations from two different areas was significant. The results of chi-square test showed that there was significant correlation between the distribution of GSTT1 genotype and the family history of coronary artery disease among the study population. GSTT1 null genotype was significantly increased in those who had a history of CHD family.
[Genetic Polymorphism of Glutathione-S-transferase M1 and T1: a Systematic Review in Chinese Population and a Pilot Study in Smear-Positive Pulmonary Tuberculosis Cases of Jilin Province].

Abstract:OBJECTIVE:To investigate the distribution of glutathione-S-transferase M1 (GSTM1) and T1 (GSTT1) genes polymorphisms in Chinese population and smear-positive pulmonary tuberculosis cases of Jilin province.METHODS:Articles about GSTM1 and GSTT1 genes polymorphisms published before 2009 in China were searched. The study population was obtained from fourteen counties (or districts) of Jilin province, which included all cases from November, 2007 to May, 2008, totally 1120. The genotypes of GSTM1 and GSTT1 were detected by multiplex PCR technique.RESULTS:The frequencies of GSTM1 and GSTT1 'null' genotypes and combination M1-T1 'null' genotype acquired from systematic review were 54.2%, 46.8% and 26.2%, respectively, in Chinese Hans they were 53.4%, 44.9% and 25.5%, and in our research they are 57.2%, 20.4% and 13.7%, respectively. No significant differences between the frequencies of males and females as well as among that of different age groups were observed (P > 0.05). The frequency of GSTM1 'null' genotype in our research is slightly higher than that in systematic review (P = 0.016) , and the frequencies of GSTT1 'null' genotype and combination M1-T1 'null' genotype and are significantly lower than those in systematic review (both P < 0.001).CONCLUSION:The frequencies of GSTM1 and GSTT1 'null' genotypes were different among ethnic. The statistical difference between systematic review and our research may due to our large sample size and mostly Southern people in previous studies.

Abstract:Glutathione S-transferases (GSTs) are the major detoxifying Phase II enzyme for eliminating electrophilic compounds. Mutations in GSTM1, GSTP1 and GSTT1 in Caucasian and GSTA1 in Chinese have been found to reduce enzyme activity. However, data on the impact of common genetic polymorphisms of GSTM1 and GSTP1 on enzyme activity in Chinese is lacking. This study aimed to investigate the effect of common GSTP1 and GSTM1 polymorphisms on erythrocyte GST activity in healthy Chinese (n=196). GSTM1 null mutation (GSTM1*0) was analyzed by a PCR-Multiplex procedure, whereas GSTP1 313A→G polymorphism (resulting in Ile105Val at codon 105) was analyzed by PCR-restriction fragment length polymorphism (RFLP) analysis. Erythrocyte GST activity was measured using 1-chloro-2,4-dinitro-bezene (CDNB) as the model substrate. The frequency of GSTM1 null genotype was 54.3% and the frequency of GSTP1-Ile/Ile, -Ile/Val, and -Val/Val genotype was 60.7%, 35.2% and 4.1%, respectively, with a frequency of 21.7% for the 105 valine allele. Age, gender and smoking did not significantly affect the erythrocyte GST activities. The mean erythrocyte GST enzyme activity for GSTP1*-Ile/Val genotype group (3.53±0.63U/gHb) was significantly lower than that for subjects with GSTP1-Ile/Ile genotype (4.25±1.07U/gHb, P=0.004), while subjects with the GSTP1-Val/Val genotype had the lowest enzyme activity (2.44±0.67U/gHb). In addition, the GST activity in carriers of GSTM1*0/GSTP1-Ile/Ile was significantly higher than that of subjects inherited GSTM1*0/GSTP1-Ile/Val or GSTM1*0/GSTP1-Val/Val. However, there is no association between GSTM1 null mutation and reduced enzyme activity. GSTP1 codon 105 mutation led to reduced erythrocyte GST activity in Chinese. A combined GSTP1 and GSTM1 null mutations also resulted in significantly reduced GST activity. Further studies are needed to explore the clinical implications of GSTM1 and GSTP1 polymorphisms.
Genetic polymorphisms of glutathione S-transferase M1 and prostate cancer risk in Asians: a meta-analysis of 18 studies.

Abstract:Background: Many studies have investigated associations between the glutathione S-transferase M1 (GSTM1) null polymorphism and risk of prostate cancer, but the impact of GSTM1 in people who live in Asian countries is still unclear owing to inconsistencies across results. Methods: We searched the PubMed, Web of Science, Scopus, Ovid and CNKI databases for studies of associations between the GSTM1 null genotype and risk of prostate cancer in people who live in Asian countries, and estimated summary odds ratios (ORs) with 95% confidence intervals (95% CIs). Results: A total of 18 case-control studies with 2,172 cases and 3,258 controls were included in this meta-analysis, which showed the GSTM1 null genotype to be significantly associated with increased risk of prostate cancer in people who live in Asian countries (random-effects OR=1.74, 95% CI1.44-2.09, P<0.001). Similar results were found in East Asians (OR=1.41; 95% CI: 1.12-1.78; P=0.004) and Caucasians in Asia (OR=2.19; 95% CI: 1.85-2.60; P<0.001). No evidence of publication bias was observed. Conclusions: This meta-analysis of available data suggested that the GSTM1 null genotype does contribute to increased risk of prostate cancer in people who live in Asian countries.
Comprehensive Analysis of UGT1A1 Genetic Polymorphisms in Chinese Tibetan and Han Populations

Abstract:The study of polymorphism of the UGT1A1 gene has not been reported in the Chinese Tibetan population, and there are no comparisons of genetic polymorphism in the gene between Chinese Han and Tibetan populations. In this study, we directly sequenced the functional regions of the UGT1A1 gene in 200 unrelated healthy Chinese volunteers, detecting 20 variations (including five novel ones). The distributions of allele and genotype frequencies differ between the two groups. UGT1A1*6 is the major reduced functional variant in the populations, and the *27 allele was detected only in the Han group. Differences in the frequencies of the UGT1A1*6/*63 genotype between the Tibetan and Han populations were statistically significant (P = 0.009). Our genetic data might provide fundamental information for the advance of personalized medicine and will facilitate genotype-phenotype studies in larger populations.

Abstract:Objective Human glutathione S-transferases(hGSTs) play a crucial role in the biological detoxification processes of drugs and xenobiotics.In drug metabolic process,the gene polymorphism caused by a homozygous or heterozygous deletion of the GSTM1 and GSTT1 genes was associated with the metabolic speed of glutathione S-transferases.The aim of this study was to investigate the association between gene polymorphisms and the metabolic phenotype of GSTM1 and GSTT1 in normal Chinese Han children,to provide an important theoretical basis for guiding the clinical treatment of drugs which metabolic by liver.Methods Chinese Han healthy children were identified who were randomly sampled from 2005 to 2010 in healthy center of Beijing Children's Hospital.The mean age of them was 5.425 years,ranged from 2 months to 14 years.The participants with positive history of diagnosed cancers,psychiatric disorders,asthma,cataract and cardiovascular disease showing significant association with hGSTs polymorphism were excluded.Demographic and medical information of these children was obtained from the patients' files.Genomic DNA was extracted from peripheral blood by EDTA anticoagulation,using a standard salting-out procedure.The concentration and purity of DNA were estimated spectrophotometrically.Genotyping was carried out by using traditional PCR-RFLP method.Further comparison between our existing data and previously reported frequencies in other ethnic populations,was performed to determine inter-ethnic differences.Results A total of 786 Chinese Han healthy children(486 males and 300 females) were enrolled into the study.In the study population,homozygous deletion(*0/*0) referring to slow metabolism was detected in 59.3%(n=466) for GSTM1 gene and 58.4%(n=459)for GSTT1 gene of the subjects,GSTM1 and GSTT1 homozygous deletions(*0/*0) respectively.The heterozygous deletion(*1/*0) referring to middle metabolism was detected in 34.0%(n=267) for GSTM1 gene and 35.1%(n=276) for GSTT1 gene of the subjects,respectively.The homozygote of non-deletion(*1/*1) referring to fast metabolism was detected in 6.7%(n= 53) for GSTM1 gene and 6.5%(n=51) for GSTT1 gene of the subjects,respectively.There was no co-expression of GSTM1 and GSTT1.And there were no significant statistical differences in gene copy number variation frequency between different genders.The comparisons of GSTM1 and GSTT1 gene genotype frequencies among worldwide populations showed different distribution patterns in Asians,Black and Caucasians.The homozygotes of deletion of the DNA fragments of interest for GSTM1 and GSTT1 gene were more common in Asia population.Conclusions In this study,GSTM1 and GSTT1 slow metabolism(indicated by *0/*0 genotype) was more prone in Chinese Han children.The distributions of GSTM1 and GSTT1 gene polymorphisms varied among different races or regions.It provided an important theoretical basis for formulate treatment plans which suitable for Chinese Han children aiming at the hepatic metabolism drugs.Meanwhile,individualized medical treatment may be fulfilled in Chinese Han children,such as antitubercular treatment.However,observing the frequencies of genetic polymorphism will be insufficient in the absence of any supporting clinical data on catalytic or functional activity concerning the effect of a given genetic variant on the future research,it is planed to present a complete analysis with respect to the role of these variants in influencing the serum levels and therapeutic efficacy of drugs.

Abstract:Objective To investigate the distribution of genetic polymorphisms of human glutathione-S-transferase P1 and M1(GSTP1 and GSTM1) in Hakka population of Meizhou area in South China. Methods Co-amplifying GSTM1 and β-Globin gene to detect the absence or presence of the GSTM1 gene, polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) were used to identify the genotypes of GSTP1 gene,and the data were analyzed with SPSS10.0 software. Results The GSTP1 and GSTM1 genetic polymorphisms were examined in 512 samples. The frequency of null genotype of GSTM1 gene was 62.1%, while the frequency of GSTP1 A/A, GSTP1 A/G and GSTP1 G/G were 69.1%, 28.2% and 2.7% respectively, and the GSTP1 genetic polymorphism distribution was in accordance with the Hardy-Weinberg equilibrium rule. A significant difference was observed in GSTM1 genetic polymorphisms between drinkers and non-drinkers. The drinkers had higher frequency of absence GSTM1 gene. There was also a significant difference in the GSTP1 genetic polymorphisms between smoking group and non-smoking group. Compared with non-smokers, the smokers had a higher frequency of mutate genotype of GSTP1 gene and lower frequency of wild genotype. However there were no differences shown on the frequency of the GSTP1 genotypes in the aspect of cumulative tobacco consumption. Logistic regression analysis results showed that there was a weak association between the GSTM1 genetic polymorphisms and family history of hypertension. Conclusion There are significant differences in the GSTP1 genetic polymorphisms among different smoking status and in GSTM1 genetic polymorphisms between drinkers and non-drinkers. A weak association has been observed between the GSTM1 genetic polymorphisms and family history of hypertension.

Abstract:Objective To investigate the allele frequencies of CYP3A5 gene in Han Chinese,Uygur Chinese,and Kazakh Chinese,and compared with other ethnic groups.Methods CYP3A5 * 3 was tested using the polymerase chain reactionrestriction fragment length polymorphism method.Frequencies of genotypes and alleles were calculated and compared with those in other populations.Results The most common genetic type of CYP3A5 genes is CYP3A5 * 3 / * 3,with the frequency in the Han nationality(50.4%),Uygur(76.1%) and Kazak(70.9%).The sum of frequency of CYP3A5* 1 / * 1 and* 1 / * 3 genotype in Han(49.5%) was statistically higher than that in Uygur(23.9%) and Kazak(28.9%),and there was no difference between the latter two.Conclusion There are marked ethnic differences in the mutant frequencies of CYP3A5 between Chinese and other ethnic groups.The frequency of CYP3A5 polymorphism in Uygur Chinese and Kazakh Chinese was consistent with that reported for the Caucasian population.

Abstract:OBJECTIVETo find out the distributive features of some metabolic genes polymorphisms in Han population of south area of China.METHODSStudy population was obtained from the controls of a community based case-control study, which included 290 blood relatives (inner control) and 404 non-blood relatives (outer control).RESULTSFrequencies of CYP1A1, GSTM1 and GSTT1 polymorphisms had no significant difference among confounding factors, such as sex, living areas, stomach cancer family history and history of tobacco smoking etc. Some controls showed significant difference in age group and alcohol drinking which would be adjusted in analysis of the relationship between polymorphisms and cancers. CYP1A1 Ile/Val and Val/Val genotypes were 33.43% and 5.62% respectively, which were similar to other results from Chinese and Japanese, but higher than those from Caucasians in American, Europe and African-Americans. GSTM1 null allele frequency was 53.48% in our population, which showed difference even among Chinese in different areas. GSTT1 null allele frequency was 45.78%, which was significantly higher than that in Caucasians and African-American.CONCLUSIONThe frequencies of CYP1A1 Ile/Val, Val/Val and GSTT1 null in Han population in south area of China are significantly higher than those in other races, while the ethnic difference of frequency of GSTM1 null is less.

Abstract:OBJECTIVE:To survey the frequencies and characteristics of polymorphism of seven genes coding drug-metabolizing enzymes in a Han Chinese population.METHODS:Genomic DNA was extracted from peripheral blood in 1382 unrelated Han Chinese volunteers. Genotypes of CYP450, NQO1, MPO, MTHFR, NAT2, TPMT*3B(G460A) and TPMT*3C(A719G) were analyzed by PCR-RFLP. GSTM1 and GSTT1 genotypes were detected by multiple PCR and TPMT*2(G238C) genotypes by allele-specific PCR.RESULTS:The allele frequencies of wild-type, heterozygosity and homozygosity were 99.8%, 0.2% and 0 in CYP3A4*1B genotypes, 8.4%, 34.3% and 57.3% in CYP3A5*3 genotypes, 28.7%, 49.7% and 21.6% in NQO1(C609T) genotypes, 75.0%, 23.2% and 1.8% in MPO(G463A) genotypes, 25.9%, 44.9% and 29.2% in MTHFR(C677T) genotypes, 67.3%, 30.4% and 2.3% in MTHFR(A1298C) genotypes, and 96.8% 3.2% and 0 in TPMT*3C(A719G) genotypes, respectively. The frequencies of present and null genotypes were 36.1% and 63.9% in GSTM1, 54.4% and 45.6% in GSTT1 respectively. The frequencies of NAT2*4/*4, *4/*5, *4/*6, *4/*7, *5/*5, *5/*6, *5/*7, *6/*6, *6/*7 and *7/*7 genotypes were 34.5%, 4.3%, 24.3%, 18.2%, 0.1%, 1.8%, 1.5%, 5.0%, 7.6% and 2.6% respectively. The frequencies of specific NAT2 alleles were 57.9%, 3.9%, 21.8% and 16.3% for NAT2*4, *5, *6 and *7 respectively. The frequencies of genophenotypes were 81.4% and 18.6% in NAT2 fast acetylator and low acetylator respectively. The allele frequency of wild type were 100% in both TPMT*2(G238C) genotypes and TPMT*3B(G460A). The variant alleles of TPMT*2(G238C) and TPMT*3B(G460A) were not found in 1382 Han Chinese subjects.CONCLUSION:Significant differences in the distributions and frequencies in genetic polymorphisms of drug-metabolizing enzymes are identified between Han Chinese population and Caucasians, as well between Han Chinese population and Melanoderma while only some heterogeneity has been observed between Han Chinese population and other Xanthoderms in Asia.

Abstract:OBJECTIVE:To investigate the frequencies of GSTM1, GSTT1 and GSTP1 polymorphisms in Dong, Yi and Yao ethnic groups from Guizhou.METHODS:In 321 volunteers who were population-based, GSTM1 and GSTT1 polymorphisms were analyzed by a multiplex-PCR procedure, whereas GSTP1 polymorphism was analyzed by PCR-RFLP method.RESULTS:Null genotype for GSTM1 and GSTT1 was 59.6%-71.2% and 39.4%-72.5%, respectively. The genotypic distribution of GSTP1 was 63.3%-75% for AA, 23.2%-35.8% for AG, 0-1.9% for GG, whereas the allelic frequencies were 81.2%-86.6% for the A allele, and 13.4%-18.8% for the G allele.CONCLUSION:There is a significant relationship between GSTT1 frequencies and ethnic populations.

The most frequent genotypes were CYP2B6*1/CYP2B6*1 (50.8%), CYP2B6*1/CYP2B6*6 (24.4%), and CYP2B6*1/CYP2B6*4 (7.3%) in Han subjects, whereas the most frequent genotypes in Uygur subjects were CYP2B6*1/CYP2B6*1 (36.3%), CYP2B6*1/CYP2B6*6 (25.3%), CYP2B6*1/CYP2B6*5 (5.5%) and CYP2B6*6/CYP2B6*6 (5.5%). The frequencies of 64C > T mutation in Han and Uygur Chinese were significantly lower than that in American Caucasian (P < 0.05). These results indicate that there were marked ethnic differences in the mutant frequencies of CYP2B6 between Chinese and other ethnic groups.
