Methylenetetrahydrofolate Reductase C677t Gene Polymorphisms in Chinese Hypertensives

Abstract:Background and objective Mutations in the 5,10-methylenetetrahydrofolate reductase (MTHFR) gene could reduce the enzyme activity and lead to hyperhomocysteinemia, a condition that has been associated with several vascular conditions, in particular, coronary artery disease and deep vein thrombosis. The aim of this study was to assess the prevalence of the most common polymorphisms, C677T, which have not been well studied in hypertensives. Methods A total of 456 hypertensives from 6 cities distributed widely from north to south in China were studied. DNA samples were isolated from peripheral blood samples and genotyped using PCR-RFLP method. Homocysteine and folate concentrations were also examined. Results The frequency of CC, CT and TT genotypes in Chinese hypertensives was 25.1%, 49.8%, and 25.1%, respectively. Overall, the frequency of allele T was 50%, though it varied significantly across 6 Chinese cities. The frequency of TT homozygous was 14.6% among southern hypertensives and 29.7% among northern hypertensives. The difference was statistically significant (p<0.01). In both Northern and Southern subjects, TT homozygotes had significantly higher plasma Hcy levels and lower folate than those with CC or CT genotypes. Conclusion Compared with other populations reported so far, Chinese hypertensives, especially in Northern cities, had the highest frequency of allele T of MTHFR C677T polymorphism.
