The alternative use of the tetrazole photoclick reaction was thus explored: tetrazoles were incorporated into Bodipy and Acedan dyes, providing novel photo‐crosslinkers with one‐ and two‐photon fluorescence Turn‐ON properties that may be developed into protein‐detecting biosensors. Further introduction of these photo‐activatable, fluorogenic moieties into staurosporine resulted in the corresponding probes capable of photoinduced, no‐wash imaging of endogenous kinase activities in live mammalian cells.

Abstract:Click chemistry has been rapidly developed in recent years. Among various established click reaction systems, the spontaneous amino-yne click reaction stands out. In addition to the general advantages associated with click reactions, amino-yne click reaction possesses extra merits, such as spontaneity of reactions, ubiquity of amines and cleavability of products with specific stimuli. Therefore, the amino-yne click reaction has become a useful tool and provided convenience for scientists in various fields since its first report in 2017. In this review, we capture the recent progress in the applications of amino-yne click reaction, which are briefly classified and reviewed in the fields of surface immobilization, drug delivery carrier design, hydrogel materials preparation and synthesis of polymers with unique structures.

Abstract:In the last two decades, click chemistry has become a modular synthetic procedure to assemble new molecular structures. It is a powerful methodology that relies on the construction of carbon-heteroatom bonds of a variety of reactants. In modern synthetic chemistry, it has been applied in a large number of applications, from pharmaceutical to material science. The copper-catalyzed 1,2,3-triazole preparation, reacting organic azides with alkynes, has become the star of click chemistry due to its reliability and biocompatibility. As a consequence, this reaction has found many applications in diverse areas such as bioconjugation, material science, and drug discovery. In order to understand the effect of copper catalysts and ligands in click chemistry, it is important to understand the structural and mechanistic aspects involved. In this review, several physicochemical aspects of click chemistry are discussed. First, the mechanisms and intermediates involved in the preparation of 1,2,3-triazoles. Second, the different types of copper catalysts are used to perform the reaction regioselectively. The last section shows the structure and characteristics of effective ligands utilized to improve click chemistry under different experimental conditions.

Abstract:Introduction: Click chemistry has been exploited widely in the past to expedite lead discovery and optimization. Indeed, Copper-catalyzed azide-alkyne cycloaddition (CuAAC) click chemistry is a bioorthogonal reaction of widespread utility throughout medicinal chemistry and chemical biology.Areas covered: The authors review recent applications of CuAAC click chemistry to drug discovery based on the literature published since 2013. Furthermore, the authors provide the reader with their expert perspectives on the area including their outlook on future developments.Expert opinion: Click chemistry reactions are an important part of the medicinal chemistry toolbox and offer substantial advantages to medicinal chemists in terms of overcoming the limitations of useful chemical synthesis, increasing throughput, and improving the quality of compound libraries. To explore new chemical spaces for drug-like molecules containing a high degree of structural diversity, it may be useful to merge the diversity-oriented synthesis and 'privileged' substructure-based strategy with bioorthogonal reactions using sophisticated automation and flow systems to improve productivity. Large compound libraries obtained in this way should be of great value for the discovery of bioactive compounds and therapeutic agents.

Abstract:Click chemistry is a powerful chemical reaction with excellent bioorthogonality features: biocompatible, rapid and highly specific in biological environments. For glycobiology, bioorthogonal click chemistry has created a new method for glycan non-invasive imaging in living systems, selective metabolic engineering, and offered an elite chemical handle for biological manipulation and glycomics studies. Especially the [3 + 2] dipolar cycloadditions of azides with strained alkynes and the Staudinger ligation of azides and triarylphosphines have been widely used among the extant click reactions. This review focuses on the azide-based bioorthogonal click chemistry, describing the characteristics and development of these reactions, introducing some recent applications in glycobiology research, especially in glycan metabolic engineering, including glycan non-invasive imaging, glycomics studies and viral surface manipulation for drug discovery as well as other applications like activity-based protein profiling and carbohydrate microarrays.

Abstract:Carbohydrates, traditionally known for their energy-providing role, have gained significant attention in drug discovery due to their diverse bioactivities and stereodiversity. However, pure carbohydrate molecules often exhibit limited bioactivity and suboptimal chemical and physical characteristics. To address these challenges, functional groups with bioactive scaffolds have been incorporated into carbohydrate to enhance their bioactivity and improve their overall properties. Among the various synthetic methods available, click chemistry has emerged as a powerful tool for the synthesis of carbohydrate-containing bioactive scaffolds, known as glycohybrids. Click chemistry offers several advantages, including high chemo- and regioselectivity, mild reaction conditions, easy purification and compatibility with multiple functional groups. In the present review, we have emphasized the recent advances and most pertinent research on the development of 1,2,3-triazole-containing glycohybrids using click reaction, their biological evaluations and the structure-activity relationship during 2017-2023. Abstract:This review highlights a modular, rapid and regioselective 1,2,3‐triazole forming 'Click Chemistry' via [3+2] reaction of diverse range of azido‐alkyne scaffolds under intramolecular fashion to furnish hgh‐to‐excellent yield of respective triazole‐appended molecular architectures including fused‐heterocycles, well‐defined peptidomemics, and macrocycles. The intramolecular click chemistry exemplified different variant of cyclyzation tool such as metal‐catalyzed (CuAAC/RuAAC) cyclization, organo‐catalyzed cyclization, topochemical, thermally induced, and cyclization as part of multicomponent reaction as the key step of cycloaddition led to respective triazole systems of notable features in various emerging domains of investigation ranging from chemical biology to catalysis and medicinal chemistry to material science. Click Chemistry, a modular, rapid, and one of the most reliable tool for the regioselective 1,2,3‐triazole forming [3+2] reaction of organic azide and terimal alkyne is widely explored in various emerging domains of research ranging from chemical biology to catalysis and medicinal chemistry to material science. This regioselective reaction from a diverse range of azido‐alkyne scaffolds has been well performed in both intermolecular as well as intramolecular fashions. In comparison to the intermolecular metal (Cu/Ru/Ni) variant of 'Click Chemistry', the intramolecular click tool is little addressed. The intramolecular click chemistry is exemplified as a mordern tool of cyclization which involves metal‐catalyzed (CuAAC/RuAAC) cyclization, organo‐catalyzed cyclization, and thermal‐induced topochemical reaction. Abstract:The present review provides an overview of CuAAC chemical reactions with various chemical conditions and introduces triazole‐based antifungal compounds which assist researchers in designing and synthesizing new antifungal triazole hybrids. Click chemistry is widely used for the efficient synthesis of 1,4‐disubstituted‐1,2,3‐triazole, a well‐known scaffold with widespread biological activity in the pharmaceutical sciences. In recent years, this magic ring has attracted the attention of scientists for its potential in designing and synthesizing new antifungal agents. Despite scientific and medical advances, fungal infections still account for more than 1.5 million deaths globally per year, especially in people with compromised immune function. This increasing trend is definitely related to a raise in the incidence of fungal infections and prevalence of antifungal drug resistance. In this condition, an urgent need for new alternative antifungals is undeniable. By focusing on the main aspects of reaction conditions in click chemistry, this review was conducted to classify antifungal 1,4‐disubstituted‐1,2,3‐triazole hybrids based on their chemical structures and introduce the most effective triazole antifungal derivatives. It was notable that in all reactions studied, Cu(I) catalysts generated in situ by the reduction in Cu(II) salts or used copper(I) salts directly, as well as mixed solvents of t‐BuOH/H2O and DMF/H2O had most application in the synthesis of triazole ring. The most effective antifungal activity was also observed in fluconazole analogs containing 1,2,3‐triazole moiety and benzo‐fused five/six‐membered heterocyclic conjugates with a 1,2,3‐triazole ring, even with better activity than fluconazole. The findings of structure–activity relationship and molecular docking of antifungal derivatives synthesized with copper‐catalyzed azide–alkyne cycloaddition (CuAAC) could offer medicinal chemistry scientists valuable data on designing and synthesizing novel triazole antifungals with more potent biological activities in their future research.

Abstract:Of the thousands of known chemical reactions, a handful of reactions, called "click" reactions, stand out with features such as good chemoselectivity, good solvent compatibilities, modularity, minimum synthetic demands, bioorthogonality and high yields. Among them, the Cu(i)-catalyzed 1,3-dipolar cycloaddition reaction between azides and terminal alkynes has emerged as a powerful tool in chemical biology and proteomics. This perspective surveys the significant contributions of click chemistry in catalomics (a sub-area in chemical proteomics), with special emphasis on activity-based protein profiling (ABPP), posttranslational modifications (PTMs) and enzyme inhibitor developments.

Abstract:Click chemistry focuses on the development of highly selective reactions using simple precursors for the exquisite synthesis of molecules. Undisputedly, the Cu-I-catalyzed azide-alkyne cycloaddition (CuAAC) is one of the most valuable examples of click chemistry, but it suffers from some limitations as it requires additional reducing agents and ligands as well as cytotoxic copper. Here, we demonstrate a novel strategy for the azide-alkyne cycloaddition reaction that involves a photoredox electron-transfer radical mechanism instead of the traditional metal-catalyzed coordination process. This newly developed photocatalyzed azide-alkyne cycloaddition reaction can be performed under mild conditions at room temperature in the presence of air and visible light and shows good functional group tolerance, excellent atom economy, high yields of up to 99 %, and absolute regioselectivity, affording a variety of 1,4-disubstituted 1,2,3-triazole derivatives, including bioactive molecules and pharmaceuticals. The use of a recyclable photocatalyst, solar energy, and water as solvent makes this photocatalytic system sustainable and environmentally friendly. Moreover, the azide-alkyne cycloaddition reaction could be photocatalyzed in the presence of a metal-free catalyst with excellent regioselectivity, which represents an important development for click chemistry and should find versatile applications in organic synthesis, chemical biology, and materials science.

Abstract:In recent years, "click" chemistry, especially the Cu(I)-catalyzed azide/alkyne 1,3-dipolar cycloaddition "click" reaction, has emerged as an attractive and promising synthesis approach due to its prominent virtues such as high specificity, quantitative yields, compatibility with various solvents (including water), and mild reaction conditions. Since there are already many reviews concerning click chemistry itself or its applications in some fields, the present review focuses only on the click chemistry on the nano-surfaces that mainly include hard (e.g., Au, gamma-Fe2O3, Fe3O4, Si/SiO2, CNTs, CdSe, TiO2 nanoparticles) and soft nanosurfaces (e.g., polymeric nanoparticles and biomacromolecules). The "click" surface modification strategies of various nano-objects are summarized comprehensively, providing a valuable repository of knowledge for the interested researchers to further dive into click chemistry, surface chemistry, and biochemistry.

Abstract:Cu(I)-catalyzed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) - commonly known as the "click reaction" - serves as the most effective and highly reliable tool for facile construction of simple to complex designs at the molecular level. It relates to the formation of carbon heteroatomic systems by joining or clicking small molecular pieces together with the help of various organic reactions such as cycloaddition, conjugate addition, ring-opening, etc. Such dynamic strategy results in the generation of triazole and its derivatives from azides and alkynes with three nitrogen atoms in the five-membered aromatic azole ring that often forms gel-assembled structures having gelating properties. These scaffolds have led to prominent applications in designing advanced soft materials, 3D printing, ion sensing, drug delivery, photonics, separation, and purification. In this review, we mainly emphasize the different mechanistic aspects of triazole formation, which includes the synthesis of sugar-based and non-sugar-based triazoles, and their gel applications reported in the literature for the past ten years, as well as the upcoming scope in different branches of applied sciences.

Abstract:Topological macromolecules including branched macromolecules such as starlike,dendritic and cyclic and muticyclic macromolecules have many unique properties different from linear counterparts,and thus are new polymeric materials with potential applications in such areas as nanotechnology,biomedicines and pharmaceutics.However,their tedious synthesis is the inherent barrier preventing them from real applications.Click reactions,which are fast,selective under mild reaction conditions and create only inoffensive by-products,have greatly advanced the synthesis methodologies of topological polymers.In this review,we briefly discuss the applications of click reactions including copper catalyzed azide-alkyne cycloaddition(CuAAC),thiol-yne/ene,amine-acrylate,Diels-Alder cycloaddition reactions in synthesis of topological polymers.The future directions in this area are also discussed.
Unexpected Cyclization Product Discovery from the Photoinduced Bioconjugation Chemistry between Tetrazole and Amine

Abstract:Bioconjugation chemistry has emerged as a powerful tool for the modification of diverse biomolecules under mild conditions. Tetrazole, initially proposed as a bioorthogonal photoclick handle for 1,3-dipolar cyclization with alkenes, was later demonstrated to possess broader photoreactivity with carboxylic acids, serving as a versatile bioconjugation and photoaffinity labeling probe. In this study, we unexpectedly discovered and validated the photoreactivity between tetrazole and primary amine to afford a new 1,2,4-triazole cyclization product. Given the significance of functionalized N-heterocycles in medicinal chemistry, we successfully harnessed the serendipitously discovered reaction to synthesize both pharmacologically relevant DNA-encoded chemical libraries (DELs) and small molecule compounds bearing 1,2,4-triazole scaffolds. Furthermore, the mild reaction conditions and stable 1,2,4-triazole linkage found broad application in photoinduced bioconjugation scenarios, spanning from intramolecular peptide macrocyclization and templated DNA reaction cross-linking to intermolecular photoaffinity labeling of proteins. Triazole cross-linking products on lysine side chains were identified in tetrazole-labeled proteins, refining the comprehensive understanding of the photo-cross-linking profiles of tetrazole-based probes. Altogether, this tetrazole-amine bioconjugation expands the current bioconjugation toolbox and creates new possibilities at the interface of medicinal chemistry and chemical biology.

Abstract:Click chemistry is perhaps the most powerful synthetic toolbox that can efficiently access the molecular diversity and unique functions of complex natural products up to now. It enables the ready synthesis of diverse sets of natural product derivatives either for the optimization of their drawbacks or for the construction of natural product-like drug screening libraries. This paper showcases the state-of-the-art development of click chemistry in natural product modification and summarizes the pharmacological activities of the active derivatives as well as the mechanism of action. The aim of this paper is to gain a deep understanding of the fruitful achievements and to provide perspectives, trends, and directions regarding further research in natural product medicinal chemistry.

Abstract:Glycosidase-mediated glycosylation of alkynyl alcohols and azide-containing alcohols was followed by a click reaction, affording various types of triazole glycosides. Abstract:Visible-light-mediated methods were heavily studied as a useful tool for cysteine-selective bio-conjugation; however, many current methods suffer from bio-incompatible reaction conditions and slow kinetics. To address these challenges, herein, we report a transition metal-free thiol-sulfoxonium ylide photo-click reaction that enables bioconjugation under bio-compatible conditions. The reaction is highly cysteine-selective and generally finished within minutes with naturally occurring riboflavin derivatives as organic photocatalysts. The catalysts and substrates are readily accessible and bench stable and have satisfactory water solubility. As a proof-of-concept study, the reaction was smoothly applied in chemo-proteomic analysis, which provides efficient tools to explore the druggable content of the human proteome.
