The influence of chemotherapy and radiotherapy before operation on the colorectal tumor angiogenesis and its clinical significance

Abstract:Objective To research the influence of chemotherapy and radiotherapy before operation on the microvessel density(MVD) and vascular endothelial growth factor(VEGF) gene expression of colorectal cancer.Methods Seventy-eight patients who suffered from lower colorectal cancer were divided into 2 groups randomly.Patients in the therapy group received radiotherapy for 40 Grays as well as FOLFOX4 chemotherapy for twice,then operation therapy 4-6 weeks after the end of radiotherapy.Patients in the control group received operation directly after admission.All the specimen obtained from operation underwent the routine pathologic study as well as gene expression analysis of VEGF and MVD in tumor tissues.A semiquantitative method was used to score the gene expression of VEGF,and the Weidner Method was used to count the MVD.Results Scores of gene expression of VEGF in the therapy group were 1.05 0.35,while it were 1.91 0.78 in the control group(P<0.01).Counts of MVD in the therapy group were 12.6±3.45,and 23.8±7.67 in the control group(P<0.00).The rate of tumor shrinking,resection and anus sparing of the therapy group were all significantly higher than those of the control group.Conclusion Chemotherapy and radiotherapy before operation can repress the gene expression of VEGF and MVD in tumor tissues,and had down-staging effect before operation.
