Ultrasound-mediated Microbubble Destruction Enhances Cardiotrophin-1 Gene Expression in Bone Marrow Mesenchymal Stem Cells

Abstract:Objective To investigate the transfection method of cardiotrophin-1(CT-1) to bone marrow mesenchymal stem cells(MSCs) in vitro by ultrasound-mediated albumin microbubble destruction.Methods The complementary gene encoding rat CT-1 was inserted into expressing vector pIRES2-EGFP after being cloned from cultured ventricular myocardium of neonate SD rats.The target gene fragment,mixed gently with albumin microbubble,was transfected into MSCs mediated by ultrasound.Western blot,microscope and flow cytometry were applied to detect the expression of CT-1 and EGFP,respectively.Results The complementary gene encoding rat CT-1 was obtained,and the expressing vector pIRES2-CT-1 was constructed.In vitro,transfection twice with ultrasound intensity of 0.75 W/cm2 for 30 seconds could transfect CT-1 gene into MSCs efficiently.After being transfected,MSCs could express and secrete CT-1 and EGFP proteins.Conclusion The method of ultrasound-mediated albumin microbubble destruction can enhance CT-1 gene transfection and expression efficiently in vitro,which may provide a new way to treat acute myocardial infarction by combining gene with stem cell transplantation.
