Abstract:Oysters are the largest shellfish group cultured in China. The production of oysters reached 3.75 ¥ 10 6 metric tons in 2004, accounting for 32.3% of the total marine molluscan yield. 1 All oysters that are commercialized in China belong to the genus Crassostrea, with C. gigas, C. plicatula, C. ariakensis and C. hongkongensis being the most common species. C. gigas, C. plicatula and C. ariakensis are commonly distributed along the entire coast of China. C. hongkongensis is found from the Fujian to Guangxi Provinces in south China, with populations centering in Guangdong Province. 2 Because of the morphological plasticity, it is difficult to clearly discriminate these oyster species on the basis of their morphology, which has limited both culture efficiency and conservation of natural resources. Therefore, straightforward and reliable identification tools for commercially cultured oysters in China are required. In the last few years, mitochondrial genes with fast sequence evolution have proved to be a powerful tool in the discrimination of many species. 3–5 In oyster species, both restriction analysis of mitochondrial DNA (mtDNA) (polymerase chain reaction-restriction fragment length polymorphism [PCR-RFLP]) and sequencing or specific multiplex PCR of the mitochondrial DNA fragments have been used in the identification of species. 6–8 However, these methods are usually timeconsuming, labor intensive and expensive, being virtually impractical for multiple sample analysis. Denaturing gradient gel electrophoresis (DGGE) is an electrophoresis method based on differences in the melting behavior of double-stranded DNA fragments. It can separate DNA molecules with minor differences in nucleotide sequence. 9 DGGE has great potential to identify closely related species, and has been used in the identification and diagnosis of microorganisms 10 and a few fishes. 11 In this study, DGGE was adopted as a rapid, sensitive and easy-to-use molecular characterization method to discriminate five commercially important Crassostrea oysters. In total, 150 oysters collected from their typical habitats were studied. C. gigas samples (n = 36) were obtained from the coastal waters around Hiroshima, Japan, and farms in Shandong and Liaoning Provinces; C. plicatula (n = 32) were collected from Fuqing and Putian of Fujian Province, which is a typical habitat of this species in China. C. ariakensis (n = 32) were sampled from the Sea of Ariake, Japan, and Dongying, Shandong Province, China; C. hongkongensis (n = 30) were collected from Beihai, Guangxi Province; C. sikamea (n = 20) which originated from Japan were collected from Newport, Oregon, USA.
