There are a variety of sex-type in cucumber including monoecious,gynoecious,subgynoecious,hermaphroditic, andromonoecious and androecious.The F,M and a genes which control the female flowers ratio,unisexual flower and influence the male flowers,respectively,have been studied intensively.The cucumber sex determination hy- pothesis,molecular basis and research status for the major genes are reviewed and also the research prospects for the relative genes involved in this paper.

Abstract:Cucumber is a model in the study of unisexual flower development.Recently,although the study of sexual differentiation in cucumber had made some progress,the regulation mechanism of its sexual differentiation has not yet been fully clarified.The expressional regulation network of sexual differentiation in cucumber was set up completely at the molecular level by combinating the floral organ developmental genes,sex determination gene,endogenous hormones,environmental factors and sexual differentiation hypothesis in the paper.At the same time,the apoptosis mechanism of unisexual flower organ,which was about the interaction between hormones and sex determination gene was illustrated in the process of cucumber sexual differentiation,also the function of miRNA in the regulation of cucumber's sexual differentiation has been disscussed.

Abstract:A full-length cDNA encoding spermidine synthase CsSPDS of 317 amino acids was isolated from cucumber. CsSPDS showed high similarity to SPDSs of other species, with the greatest similarity to SPDS of Coffea arabica (identity, 87%). Northern blot analysis revealed that CsSPDS was expressed predominantly in rapid growing tissues, such as roots, young leaves and flower buds. The response of CsSPDS to the sex reversal of three-leaf stage androecious cucumber seedlings by Ethrel treatment was also examined. Our results showed that the CsSPDS was up-regulated in shoot apex during sex reversal process, which correlated with increases of spermidine synthase activity and free spermidine (Spd) content, and a decrease of putrescine (Put) content in shoot apices.

Abstract:The family Cucurbitaceae includes many economically important crops, such as cucumber (Cucumis sativus), melon (Cucumis melo), watermelon (Citrullus lanatus), and zucchini (Cucurbita pepo), which share homologous gene pathways that control similar phenotypes. Sex determination is a research hotspot associated with yield and quality, and the genes involved are highly orthologous and conserved in cucurbits. In the field, six normal sex types have been categorized according to the distribution of female, male, or bisexual flowers in a given plant. To date, five orthologous genes involved in sex determination have been cloned, and their various combinations and expression patterns can explain all the identified sex types. In addition to genetic mechanisms, ethylene controls sex expression in this family. Two ethylene signaling components have been identified recently, which will help us to explore the ethylene signaling-mediated interactions among sex-related genes. This review discusses recent advances relating to the mechanism of sex determination in cucurbits and the prospects for research in this area.

Abstract:Cucumber has been widely studied as a model for floral sex determination. In this investigation, we performed genome-wide transcriptional profiling of apical tissue of a gynoecious mutant (Csg-G) and the monoecious wild-type (Csg-M) of cucumber in an attempt to isolate genes involved in sex determination, using the Solexa technology. The profiling analysis revealed numerous changes in gene expression attributable to the mutation, which resulted in the down-regulation of 600 genes and the up-regulation of 143 genes. The Solexa data were confirmed by reverse transcription polymerase chain reaction (RT-PCR) and real-time quantitative RT-PCR (qRT-PCR). Gene ontology (GO) analysis revealed that the differentially expressed genes were mainly involved in biogenesis, transport and organization of cellular component, macromolecular and cellular biosynthesis, localization, establishment of localization, translation and other processes. Furthermore, the expression of some of these genes depended upon the tissue and the developmental stage of the flowers of gynoecious mutant. The results of this study suggest two important concepts, which govern sex determination in cucumber. First, the differential expression of genes involved in plant hormone signaling pathways, such as ACS, Asr1, CsIAA2, CS-AUX1 and TLP, indicate that phytohormones and their crosstalk might play a critical role in the sex determination. Second, the regulation of some transcription factors, including EREBP-9, may also be involved in this developmental process.

Abstract:This paper reviews the study of sex chromosomes, genes involved in sex determination and regulation of sex determination in several plants in recent years .The mechanism of sex determination in plants is discussed preliminarily. We find different plants have different mechanism in sex determination:the isolation and analysis of sex-determining genes in sex chromosome are in progress; some genes that affect the sexual expression of flowers have been cloned by mutation or subtraction library; other factors including DNA methylation, GA, ethylene and Ace play important roles in sex determination. This provides valuable methods and ideas for advanced study in the future.

Abstract:The genetic analyses of sex expression types for cucumber inbred-lines and their hybrid combinations were done to confirm the genetic pattern of the F and M genes and test genotypes of them in this study, The results of chemical treatments showed that gynoecious (FFMMAA), monoecious (ffMMAA), hermaphrodite (FFmmAA) and andromonoecious (ffmmAA) plants had identical response with ethephon and AgNO3, ethephon could induce female flowers and AgNO3 could cause male flowers to appear. These confirmed that the F and M would be closely related to ethylene pathway.

Abstract:Sex is a universal phenomenon in the world of eukaryotes. Attempts have been made to understand regulatory mechanisms for plant sex determination by investigating unisexual flowers. The cucumber plant is one of the model systems for studying how sex determination is regulated by phytohormones. A systematic investigation of the development of unisexual cucumber flowers is summarized here, and it is suggested that the mechanism of the unisexual flower can help us to understand how the process leading to one type of gametogenesis is prevented. Based on these findings, we concluded that the unisexual cucumber flowers is not an issue of sex differentiation, but instead a mechanism for avoiding self-pollination. Sex differentiation is essentially the divergent point(s) leading to heterogametogenesis. On the basis of analyses of sex differentiation in unicellular organisms and animals as well as the core process of plant life cycle, a concept of "sexual reproduction cycle" is proposed for understanding the essential role of sex and a "progressive model" for future investigations of sex differentiation in plants.

Abstract:It is well established that the plant hormone ethylene plays a key role in cucumber sex determination. Since the unisexual control gene M was cloned and shown to encode an ethylene synthase, instead of an ethylene receptor, the 'one-hormone hypothesis', which was used to explain the cucumber sex phenotype, has been challenged. Here, the physiological function of CsACS2 (the gene encoded by the M locus) was studied using the transgenic tobacco system. The results indicated that overexpression of CsACS2 increased ethylene production in the tobacco plant, and the native cucumber promoter had no activity in transgenic tobacco (PM). However, when PM plants were treated with exogenous ethylene, CsACS2 expression could be detected. In cucumber, ethylene treatment could also induce transcription of CsACS2, while inhibition of ethylene action reduced the expression level. These findings suggest a positive feedback regulation mechanism for CsACS2, and a modified 'one-hormone hypothesis' for sex determination in cucumber is proposed.

Abstract:Cucumber (Cucumis sativus L.) is becoming a model to study the sex determination mechanism in plant. The monoeciousdetermine M gene had been cloned as a key enzyme (CsACS2) which takes part in the plant hormone ethylene biosynthesis. In this study, we use the amino acid alignment, cis element analysis in promoter sequence, phylogenetic analysis and conserved domain prediction to study the CsACS2, and the results showed that: a. cucumber CsACS2 may be ortholog gene with melon CmACS7; b. two ethylene-response elements (ERE motif) and one floral organ development relative element (LEAFY motif) are found in the promoter of CsACS2 ; c. CsACS2 has a high homology (72.8%) with AtACS7 in Arabidopsis; d. a putative conserved motif (S-YF-GW) has been found in this ACC synthase. These results are useful to reveal the characteristics and potential biological role of the monoecious-determine gene CsACS2 in cucumber.

Abstract:Abstract Cucumber ( Cucumis sativus L.) is a typical monoecism vegetable with individual male and female flowers, which has been used as a plant model for sex determination. It is well known that light is one of the most important environmental stimuli, which control the timing of the transition from vegetative growth to reproductive development. However, whether light controls sex determination remains elusive. To unravel this problem, we performed high-throughput RNA-Seq analyses, which compared the transcriptomes of shoot apices between R2B1(Red light:Blue light = 2:1)-treated and R4B1(Red light:Blue light = 4:1)-treated cucumber seedlings. Results showed that the higher proportion of blue light in the R2B1 treatment significantly induced the formation of female flowers and accelerated female flowering time in this whole study. The genes related to flowering time, such as flowering locus T ( FT ) and SUPPRESSOR OF OVEREXPRESSION OF CO1 ( SOC1 ), were up-regulated after R2B1 treatment. Furthermore, the transcriptome analysis showed that up-regulation and down-regulation of specific DEGs (the differentially expressed genes) were primarily the result of plant hormone signal transduction after treatments. The specific DEGs related with auxin formed the highest percentage of DEGs in the plant hormone signal transduction. In addition, the expression levels of transcription factors also changed after R2B1 treatment. Thus, sex differentiation affected by light quality might be induced by plant hormone signal transduction and transcription factors. These results provide a theoretical basis for further investigation of the regulatory mechanism of female flower formation under different light qualities in cucumber seedlings.

Abstract:KEY MESSAGE:Molecular breeding of Cucumis sativus L. is based on traditional breeding techniques and modern biological breeding in China. There are opportunities for further breeding improvement by molecular design breeding and the automation of phenotyping technology using untapped sources of genetic diversity. Cucumber (Cucumis sativus L.) is an important vegetable cultivated worldwide. It bears fruits of light fragrance, and crisp texture with high nutrition. China is the largest producer and consumer of cucumber, accounting for 70% of the world's total production. With increasing consumption demand, the production of Cucurbitaceae crops has been increasing yearly. Thus, new cultivars that can produce high-quality cucumber with high yield and easy cultivation are in need. Conventional genetic breeding has played an essential role in cucumber cultivar innovation over the past decades. However, its progress is slow due to the long breeding period, and difficulty in selecting stable genetic characters or genotypes, prompting researchers to apply molecular biotechnologies in cucumber breeding. Here, we first summarize the achievements of conventional cucumber breeding such as crossing and mutagenesis, and then focus on the current status of molecular breeding of cucumber in China, including the progress and achievements on cucumber genomics, molecular mechanism underlying important agronomic traits, and also on the creation of high-quality multi-resistant germplasm resources, new variety breeding and ecological breeding. Future development trends and prospects of cucumber molecular breeding in China are also discussed.

Abstract:Cucumber (Cucumis sativus L.) is an important economic vegetable crop worldwide that is susceptible to various common pathogens, including powdery mildew (PM), downy mildew (DM), and Fusarium wilt (FM). In cucumber breeding programs, identifying disease resistance and related molecular markers is generally a top priority. PM, DM, and FW are the major diseases of cucumber in China that cause severe yield losses and the genetic-based cucumber resistance against these diseases has been developed over the last decade. Still, the molecular mechanisms of cucumber disease resistance remain unclear. In this review, we summarize recent findings on the inheritance, molecular markers, and quantitative trait locus mapping of cucumber PM, DM, and FM resistance. In addition, several candidate genes, such as PM, DM, and FM resistance genes, with or without functional verification are reviewed. The data help to reveal the molecular mechanisms of cucumber disease resistance and provide exciting new opportunities for further resistance breeding.

Abstract:Summary Fruit development can be generally classified into a set of biologically sequential stages including fruit initiation, growth, and ripening. Cucumber, a globally important vegetable crop, displays two important features during fruit development: parthenocarpy at fruit initiation and prematurity at harvest for consumption. Therefore, fruit growth plays essential role for cucumber yield and quality formation, and has become the research hot spot in cucumber fruit development. Here, we describe recent advances in molecular mechanisms underlying fruit growth in cucumber, include key players and regulatory networks controlling fruit length variation, fruit neck elongation, and locule development. We also provide insights into future directions for scientific research and breeding strategies in cucumber.

Abstract:Sex differentiation of flower buds is an important developmental process that directly affects fruit yield of cucumber (Cucumis sativus L.). Plant hormones, such as gibberellins (GAs) and ethylene can promote development of male and female flowers, respectively, however, the regulatory mechanisms of GA-induced male flower formation and potential involvement of ethylene in this process still remain unknown. In this study, to unravel the genes and gene networks involved in GA-regulated cucumber sexual development, we performed high throughout RNA-Seq analyses that compared the transcriptomes of shoot tips between GA3 treated and untreated gynoecious cucumber plants. Results showed that GA3 application markedly induced male flowers but decreased ethylene production in shoot tips. Furthermore, the transcript levels of M (CsACS2) gene, ethylene receptor CsETR1 and some ethylene-responsive transcription factors were dramatically changed after GA3 treatment, suggesting a potential involvement of ethylene in GA-regulated sex expression of cucumber. Interestingly, GA3 down-regulated transcript of a C-class floral homeotic gene, CAG2, indicating that GA may also influence cucumber sex determination through an ethylene-independent process. These results suggest a novel model for hormone-mediated sex differentiation and provide a theoretical basis for further dissection of the regulatory mechanism of male flower formation in cucumber. Statement: We reveal that GA can regulate sex expression of cucumber via an ethylene-dependent manner, and the M (CsACS2), CsETR1, and ERFs are probably involved in this process. Moreover, CAG2, a C-class floral homeotic gene, may also participate in GA-modulated cucumber sex determination, but this pathway is ethylene-independent.

Abstract:Sex determination is a crucially important developmental event that is pervasive throughout nature and enhances the adaptation of species. Among plants, cucumber (Cucumis sativus L.) can generate both unisexual and bisexual flowers, and the sex type is mainly controlled by several 1-aminocyclopropane-1-carboxylic acid synthases (CsACSs). However, the regulatory mechanism of these synthases remains elusive. Here, we used gene expression analysis, protein-DNA interaction assays, and transgenic plants to study the function of a gynoecium-specific gene, ETHYLENE RESPONSE FACTOR31 (CsERF31), in female flower differentiation. We found that in a predetermined female flower, ethylene signaling activates CsERF31 by CsEIN3, and then CsERF31 stimulates CsACS2, which triggers a positive feedback loop to ensure female rather than bisexual flower development. A similar interplay is functionally conserved in melon (Cucumis melo L.). Knockdown of CsERF31 by RNAi causes defective bisexual flowers to replace female flowers. Ectopic expression of CsERF31 suppresses stamen development and promotes pistil development in male flowers, demonstrating that CsERF31 functions as a sex switch. Taken together, our data confirm that CsERF31 represents the molecular link between female-male determination and female-bisexual determination, and provide mechanistic insight into how ethylene promotes female flowers, rather than bisexual flowers, in cucumber sex determination.
