Abstract:OBJECTIVE:To investigate the association between high mobility group box1 (HMGB1) and cervical squamous cell carcinoma (CSCC), and explore the role of HMGB1/RAGE pathway in the metastasis of CSCC. METHODS:Levels of HMGB1 mRNA and RAGE mRNA in CSCC and normal cervical tissues were detected by real time quantitative polymerase chain reaction (qRT-PCR), and the level of HMGB1 protein was determined by immunohistochemistry and Western blotting. RESULTS:The mRNA and protein expression of HMGB1 was significantly higher in CSCC than that in normal cervical tissue (P < 0.05), correlated with stage, invasion and metastasis (P < 0.05), but not with tumor size and differentiation (P > 0.05). The levels of RAGE mRNA and HMGB1 mRNA were both significantly higher (r = 0.663, P < 0.05) in metastatic CSCC in comparison with those in the non-metastatic cases. CONCLUSION:HMGB1 is involved in the invasion and metastasis of CSCC, and HMGB1/RAGE pathway plays an important role in the metastasis of CSCC.

Abstract:Objective:To investigate the expression and clinico-pathological findings of high mobility group box-1(HMGB-1)mRNA and receptor for advanced glycation end products(RAGE)mRNA in cervical squamous epithelial carcinoma,to explore their roles during the carcinoma process,invasion and metastasis.Methods:Real time quantitative polymerase chain reaction(qRT-PCR)were employed to detect the expression of HMGB-1 mRNA and RAGE in CSEC and nomal cervical tissues.Results:The expression of HMGB-1 mRNA in the CSEC and their paraneoplastic tissues was higher than that in normal cervix tissues.And the overexpression of HMGB-1 mRNA in the CESC tissues was significantly correlated with the stage,the presence of metastasis,but not correlated with the diameter and grade of tumor.And expression of RAGE was no significantly differences between the CSEC,paraneoplastic and normal cervical tissues.Statistical analysis showed that RAGE mRNA had a relation with the metastasis.But no relation in stage,diameter and grade of CSEC.Conclusion:HMGB-1 could be a prognostic predictor of malignant degree in the evaluation of CSEC patients.The pathway of HMGB-1/RAGE is the main mechanism correlated with the metastasis of CESC and they may become new targets for treating CSEC lyphatic metastasis.

Abstract:Objective:To investigate the expression of the receptor of advanced glycosylation end-products(RAGE) in human cervical squamous cell carcinoma(CSCC) and to explore the relationship of this expression to several factors such as differentiation degree,size and stage of CSCC,and invasion,as well as metastasis of CSCC.Methods:Immunohistochemical staining of tissue microarrays was conducted to detect the expression of RAGE in the following tissue samples:30 carcinomas in situ,90 invasive CSCCs without metastasis,30 invasive CSCCs with metastasis,and 30 cases with normal cervical squamous epithelia.Western blot analysis was used to detect the expression of RAGE in CSCC.Results:The positive expression rates of RAGE were 48.0%in CSCC(72/150) and 13.3%in normal squamous epithelial tissues of the cervix(20/150),indicating significant differences between the two conditions.The overexpression of RAGE was correlated with tumor progression,stage,invasion,and metastasis of CSCC(P<0.01),but it had no relation with the degree of differentiation and tumor size(P>0.05).Conclusion:Overexpression of RAGE in CSCC,especially in cervical squamous epithelial tissues with tumor metastasis,can be used as one of the parameters for determining tumor invasion,metastasis, and prognosis.

Abstract:Cervical cancer is the third leading cause of cancer death among women in less-developed regions. Because of the poor survivorship of patients with advanced disease, finding new biomarkers for prognostic prediction is of great importance. In the current study, mRNA datasets (GSE9750 and GSE63514) were retrieved from Gene Expression Omnibus and was used to identify differentially expressed genes. The underlying molecular mechanisms associated with high-mobility group box 1 protein (HMGB1) were investigated using bioinformatics analysis. Immunohistochemical analysis of HMGB1 was performed on 239 cases of cervical cancer samples to investigate its possible correlation with clinicopathological characteristics and outcomes. A preliminary validation has been made to explore the possible correlation factors with HMGB1 that promote migration of cervical cancer cells. Bioinformatics analysis showed that adherens junction was significant for both P-value and enrichment scores, which was consistent with the clinical study. The underlying molecular mechanisms might be the interaction among HMGB1, RAC1, and CDC42. HMGB1 expression was significantly associated with tumor size, parametrial infiltration, the depth of cervical stromal invasion, and FIGO stage (P=0.003, 0.019, 0.013, and 0.003, respectively). FIGO stage, lymph mode metastasis, and HMGB1 expression were independent predictors of a poorer prognosis of patients with cervical cancer. Knockdown of HMGB1 inhibits migration of Siha and C33A cells in vitro Western blot and quantitative real-time PCR (qRT-PCR) showed that the expression of RAC1 and CDC42 was positively correlated with HMGB1. HMGB1 is a useful prognostic indicator and a potential biomarker of cervical cancer. RAC1 and CDC42 may be involved in the progression of cervical cancer migration induced by HMGB1.

Abstract:Objective: To investigate the expressions of High Mobility Group Box 1(HMGB1) and matrix metalloproteinase-9(MMP-9) in human cervical squamous epithelial carcinoma(CSEC),and to explore the relationship among HMGB1,MMP-9 and clinical pathological characteristics of CSEC.Methods: Immunohistochemisty methods were adopted to detect the expressions of HMGB1 and MMP-9 in cervical squamous epithelial infiltrating carcinoma(CSEIC),carcinoma in situ(CIS) and normal control groups.Results: The positive expression rate of HMGB1 in CSEIC,CIS and control group was 80.0%(32/40),53.3%(8/15) and 15.8%(3/19),respectively,with a significant difference among them(P<0.01).The expression of HMGB1 had no relation with age,differentiation degree,lymphatic metastasis or clinical classification(all P>0.05).The positive expression rate of MMP-9 in CSEIC,CIS and control group was 65.0%(26/40),53.3%(8/15) and 21.5%(4/19),respectively(P<0.01).There is a positive correlation between the expression of HMGB1 and MMP-9 in CSEC,and their co-positive rate was 47.3%(26/55,rs=0.305,P<0.05).Conclusion: HMGB1 plays a role in the development of CSEC.The expressions of HMGB1 and MMP-9 in CSEC have a positive correlation with each other,which suggests that HMGB1 might be related with the invasive and metastasis of cervical carcinoma.

Abstract:Receptor for advanced glycation end products (RAGE), along with its ligand high mobility group box 1 (HMGB1), is believed to play an important role in prostate cancer. The aim of this retrospective study was to investigate the expression of RAGE and HMGB1 and their clinical impact on prostate cancer progression and prognosis. The expression of RAGE and HMGB1 were assessed by immunohistochemistry in cancer lesions from 85 confirmed prostate cancer cases. We determined the potential association between the expression level of these two proteins and the clinicopathological features and overall patient survival. The RAGE and HMGB1 were expressed in 78.8% (67/85) and 68.2% (58/85) cases of prostate cancers, respectively, and in the majority (54/85) of cases, these two proteins were co-expressed. There was a strong correlation between RAGE and HMGB1 expressions (P<0.001). The expression of RAGE, HMGB1 and their co-expression were all associated with advanced tumor clinical stage (P<0.05 for all). RAGE expression was also associated with the prostate specific antigen (PSA) level (P=0.014). However, neither the individual expression of those genes nor their co-expression was significantly related with age and Gleason score. The co-expression of RAGE and HMGB1 was associated with poor overall survival in patients with stage III and IV prostate cancer (P=0.047). These results suggest that the expression of RAGE and HMGB1 is associated with the progression and poor prognosis of prostate cancer. RAGE and HMGB1 could be new prognostic biomarkers for prostate cancer as well as molecular target for novel forms of therapies.

Abstract:Abstract Objective: To clarify the expression of HMGB1 and its relationship with clinical features of gastric cancer.Methods: Multiple databases were systematically searched for relevant studies from 1997 to 2021. We screened case - control studies stating that the association between HMGB1 protein expression and clinical characteristics in gastric cancer.Results: In total, eight independent studies with 872 patients were included. Meta – analysis indicated higher odds of HMGB1 being present in gastric tumor tissues compared to para - tumor controls ［OR ＝ 4.15, 95％ CI (3.09, 5.58), P ＜ 0.01］. High HMGB1 expression was also associated with poor differentiation rate ［OR ＝ 0.73, 95％ CI (0.54, 0.99), P ＝ 0.04］, TNM stages Ⅲ - Ⅳ ［OR ＝0. 28, 95％ CI (0.18, 0.44), P ＜ 0.01］, lymphatic metastasis ［OR ＝ 2. 08, 95％ CI (1.52, 2.84), P ＜0.01］ and diffuse type of Lauren classification [OR ＝ 0.57, 95％ CI (0.38, 0.84), P ＜0. 01］. However, there was no significant correlation between HMGB1 expression between gender and age.Conclusion: Our results illustrated that HMGB1 protein was strongly associated with adverse clinicopathological features of gastric cancer. HMGB1 may be a promising target in the development of gastric cancer.

Abstract:The aim of this study was to investigate the role of the cell surface expression levels of HLA class I and II molecules, the costimulatory molecules CD80/B7-1 and CD86/B7-2, and the adhesion molecules CD54 and CD58 during cervical carcinogenesis. The expression levels of MHC class I and II molecules, the costimulatory molecules CD80/B7-1 and CD86/B7-2 and the adhesion molecules CD54 and CD58 on CD14+ peripheral blood monocytes (PBMs) from 21 cases of cervical intraepithelial neoplasia (CIN) II-III, 51 squamous cervical carcinomas (SCCs) and 18 healthy controls were analyzed using flow cytometry analysis. We found increased expression levels of HLA-DR (p=0.000), HLA-DQ (p=0.000), CD86/B7-2 (p=0.002) and CD58 (p=0.000) on PBMs from patients with SCC and CIN II-III, compared with healthy control subjects, whereas no significant difference existed in the expression levels of HLA class I antigens, HLA-DP CD80/B7-1 and CD54. Upregulated expression levels of HLA-DR, HLA-DQ, CD86/B7-2 and CD58 were associated with disease progression, indicating that an increased expression of HLA-DR, HLA-DQ, CD86/B7-2 and CD58 on PBMs may be correlated with the evolution of cervical cancer.
Expression and Significance of NMP238 and HSP70 in Cervical Squamous Cell Cancer

Abstract:[Objective] To explore the expression and significance of Nuclear Matrix Protein (NMP)238 and heat shock protein (HSP)70 in cervical squamous cell cancer. [Methods] Immunohistochemical method for NMP238 and HSP70 were performed on 60 cases of cervical squamous cancer and 30 cases of normal cervical tissues. [Results] In cervical squamous cell cancer, the expressions of NMP238 and HSP70 were significantly higher than those of normal cervical tissues. However the expression of neither NMP238 nor HSP70 was significantly correlated with the pathological grade and clinical stages. The expression of NMP238 was significantly correlated with the expression of HSP70. [Conclusions] NMP238 and HSP70 play important roles in the genesis and development of cervical squamous cell cancer, and there may be some extent coordinated regulation among them. NMP238 and HSP70 can be an important index in predicting the prognosis of cervical squamous cell cancer.
The Clinical Significance of the CD44v6 Expression in Peripheral Blood Mononuclear Cell and Cancerous Tissue of Patients with Gastric Cancer

Abstract:Objective To investigate the clinical significance of cell surface differentiation antigen 44 (CD44) expression that shared spliced variant 6 (CD44v6)in peripheral blood mononuclear cell(PBMC)and cancerous tissue of patients with gastric carcinoma.Methods Flow cytometry was used to analyze the CD44v6 expression in the primary tumor tissue and these corresponding paracancer normal tissue after gastrectomy and in PBMC at pre-operation and post-operation one week later in 50 patients with gastric cancer and 20 normal controls.Results CD44v6 expression in PBMC was significantly higher in pre-operation of gastric cancer (9.72%±6.65%) than that in both normal controls(3.11%±1.15%) and post-operation one week later (4.44%±2.33%,P0.001 and P0.01 respectively). The significant increase (P0.01) of CD44v6 expression was found in cancerous tissue (3.67%±0.63%) when compared with corresponding paracancer normal tissue of gastric cancer(1.33%±0.51%). The level of CD44v6 expressions in PBMC and cancerous tissue in patients with gastric cancer of Ⅲ ± Ⅳ stage(12.56%±6.97% and 3.91%±0.87% respectively) was significantly higher than that of Ⅰ ±Ⅱ stage(6.89%±3.37% and 2.86%±0.65% respectively,both P0.05). CD44v6 expressions in PBMC and cancerous tissue in patients with lymph node metastasis (11.29%±5.36% and 4.41%±0.79% respectively)were significantly higher than that of patients without lymph node metastasis(7.32%±4.28% and 2.61%±0.78% respectively,both P0.05).Conclusions The level of CD44v6 expression in PBMC may be a marker of monitoring the changes of CD44v6 in gastric cancerous tissue and of important clinical value in the diagnosis,judgment of tumor development and metastasis and therapeutic evaluation for the patients with gastric cancer by flow cytometry.

Abstract:OBJECTIVE:To investigate the mRNA and protein expressions of high mobility group box-1 (HMGB1) in the tumor tissues and sera of patients with laryngeal squamous cell carcinoma (LSCC) and their clinical significance. METHODS:Semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot were used to detected the expressions of HMGB1 mRNA and protein in the tumors and adjacent normal epithelial tissues in 30 patients with LSCC. Serum HMGB1 protein levels in the patients with LSCC and in 10 healthy volunteers were detected with enzyme-linked immunosorbent adsorption experiment (ELISA). RESULTS:RT-PCR demonstrated that the mean relative mRNA expression levels of HMGB1 (HMGB1/GAPDH) in LSCC tissues and in adjacent normal epithelial tissues were 1.25 ± 0.12 and 0.32 ± 0.04, respectively (t = 40.27, P < 0.05). Western blot revealed that the mean relative protein expression levels of HMGB1 (HMGB1/β-actin) were 1.29 ± 0.10 and 0.34 ± 0.03 (t = 49.84, P < 0.05), respectively. Both mRNA and protein expression levels of HMGB1 were associated with T stage, clinical stage, lymph node metastasis status and smoking (all P < 0.05), but no significant correlation with age, alcohol consumption and primary tumor grade and location (all P > 0.05). Mean serum HMGB1 protein levels in patients with LSCC and healthy volunteers were (24.80 ± 14.08) ng/ml and (23.58 ± 14.69) ng/ml (t = 0.37, P > 0.05). CONCLUSIONS:Both mRNA and protein expressions of HMGB1 were obviously elevated in LSCC, which were associated closely with T stage, clinical stage and lymph node metastasis.

Abstract:High mobility group protein box1 (HMGB1) and its receptor-receptor for advanced glycation end products (RAGE) are pivotal factors in the development and progression of many types of tumor, but the role of HMGB1-RAGE axis in hepatocellular carcinoma (HCC) especially its effects on metastasis and recurrence remains obscure. Here, we report the role of HMGB1-RAGE axis in the biological behaviors of HCC cell lines and the underlying molecular mechanism. We show that the expressions of HMGB1, RAGE, and extracellular HMGB1 increase consistently according to cell metastasis potentials, while the concentration of soluble form of RAGE (sRAGE) is inversely related to metastasis potential of HCC cells. Furthermore, our data show that rhHMGB1 promotes cellular proliferation, migration, and invasion, and increases the level of nuclear factor kappa B (NF-κB), while administrations of HMGB1-siRNA, RAGE-siRNA, anti-HMGB1 neutralizing antibody, anti-RAGE neutralizing antibody, and sRAGE inhibit cellular proliferation, migration, and invasion. Moreover, we also demonstrate that the expression of NF-кB is inhibited by knockdown of HMGB1 or RAGE. Collectively, these data demonstrate that HMGB1 activates RAGE signaling pathways and induces NF-кB activation to promote cellular proliferation, invasion, and metastasis, in HCC cell lines. Taken together, HMGB1-RAGE axis may become a potential target in HCC therapy.
Effect of High-mobility Group Box-1 on Expression of Stem Cell Markers OCT4, Sox2 and Nanog in Cervical Carcinoma

Abstract:Objective To investigate the role of High-mobility group box-1 (HMGB1) protein in regulating the expression of cancer stem cell markers Nanog, OCT4 and Sox2. Methods We transferred recombinant plasmid and target-specific siRNA of HMGB1 into cervical cancer cells. The expression of cancer stem cell markers (OCT4, Sox2 and Nanog) were measured by Western blot and flow cytometry. Immunohistochemical method was applied to detect HMGB1, OCT4, Sox2 and Nanog expression in 66 cases of cervical tissues. Rank correlation analysis was used to analyze their correlation. Results The full-length coding sequence of HMGB1 was 648 bp and recombined to overexpress the plasmid GV230. The selection of the highest interference rate was carried out for subsequent experiments. The protein expression and rates of stem cell markers OCT4, Sox2 and Nanog were significantly higher in HMGB1 overexpression group than those in low HMGB1 expression group and empty vector group(P < 0.05). The protein expression and rates of stem cell markers OCT4, Sox2 and Nanog were significantly lower in low HMGB1 expression group than those in HMGB1 overexpression group and empty vector group(P < 0.05). HMGB1, OCT4, Sox2 and Nanog protein expression in cervical carcinoma tissues were higher than those in chronic cervicitis tissues. Positive correlations were also found between HMGB1 and OCT4, Sox2 and Nanog expression. Conclusion HMGB1 could regulate the expression of cancer stem cell markers Nanog, OCT4 and Sox2, therefore, it is probably a potential target for anti-tumor therapy.
Clinical Implication of the Relationship Between High Mobility Group Box-1 and Tumor Differentiation in Hepatocellular Carcinoma

Abstract:BACKGROUND/AIM: High mobility group box-1 (HMGB1) induces the release of proinflammatory cytokines and chemokines as a late-acting mediator of inflammation. Hepatocellular carcinoma (HCC) is a typical inflammation-related cancer. However, little is known about the relationship between HCC and HMGB1 and its receptor RAGE (receptor for advanced glycation end products). This study analyzes the clinicopathological relevance of HMGB1 expression level and the effect of HMGB1 expression on the characteristics of HCC.MATERIALS AND METHODS: Samples from 75 HCC patients including 13 with positive hepatitis B surface antigen and 36 with hepatitis C antibody were studied. The expression of HMGB1 in paired cancer and non-cancerous tissues from patients with HCC was assessed using reverse-transcription polymerase chain reaction (RT-PCR) and western blotting. Quantitative RT-PCR data were analyzed in association with the clinicopathological factors of patients with HCC.RESULTS: The expression of HMGB1 mRNA in HCC was high in well-differentiated tumors, but declined as tumors dedifferentiated to moderately and poorly differentiated HCC. The levels of HMGB1 mRNA showed a negative correlation with the presence of portal invasion (p=0.005) and the rise of serum PIVKA-II (p=0.034). There was no clear correlation between HMGB1 expression and proliferation activity of HCC using Ki-67 staining.CONCLUSION: In HCC, HMGB1 expression level correlated inversely with tumor differentiation. The RAGE-HMGB1 interaction may play a greater role in the early stages of HCC tumorigenesis than during cancer development.

Abstract:OBJECTIVE To investigate the expression and biological significance of HMGB1 and VEGF protein in tissue specimens of laryngeal squamous cell carcinoma (LSCC), and further study the correlation between HMGB1 and VEGF protein. METHOD The expression of HMGB1 and VEGF protein was evaluated by immunohistochemical staining in 69 cases of LSCC specimens and 15 cases of adjacent epithelial tissue samples, and futher correlated with clinicopathologic parameters. RESULT The positive rates of HMGB1 and VEGF in LSCC tissues were significantly higher than those in adjacent non-cancerous mucosa (P < 0.01), and the expression of these two marks was closely correlated with clinical stage (P < 0.05) and metastasis (P < 0.05) in LSCC. While the expression of HMGB1 and VEGF had no significant correlations with age, sex, histological differentiation and tumor site (P > 0. 05). There was a positive correlation between the expression of HMGB1 and VEGF (P < 0.05). The Kaplan-Meier survival analysis showed that patients with strong expression of HMGB1 or VEGF had poorer overall survival compared with that in patients with relative low HMGB1 or VEGF expression (P < 0.05). Multivariate COX regression analysis revealed that both lymph node metastasis and HMGB1 expression were independent prognostic factors for patients with LSCC. CONCLUSION This study demonstrated that HMGB1 and VEGF protein overexpression were closely associated with clinical stage, metastasis and poorer prognosis in patients with LSCC. Increased expression of these two proteins in LSCC suggested that HMGB1 and VEGF might play a critical role in the initiation and progression of LSCC.

Abstract:To evaluate the expression of HMGB1 protein in tissue specimens of laryngeal squamous cell carcinoma (LSCC) and adjacent normal mucosa, and explore the correlation of HMGB1 protein expression with clinicopathologic features and prognosis in LSCC.Ninty-three cases of LSCC and 5 cases of adjcent mucosal tissue samples were included in this study. Immunohistochemical staining was performed on paraffin-embedded tissue specimens to examine the HMGB1 protein expression. The data were futher correlated with the clinicopathological features and prognosis of the LSCC patients.The positive rates of HMGB1 expression in LSCC specimens was 87.1%, significantly higher than that in the adjcent normal mucosa samples (46.7%, P = 0.001), and its overexpresion was closely correlated with T stage (Chi2 = 10.878, P = 0.004), clinical stage (Chi2 = 21.115, P < 0.01), metastasis (Chi2 = 28.298, P < 0.01) and recurrence (Chi2 = 14. 923, P = 0.001) in patients with LSCC. Patients with HMGB1 overexpression had both poorer disease-free survival and poorer overall survival compared with that in patients with low HMGB1 expression (Chi2 = 13.815, Chi2 = 11.912; Both P < 0.01). Univariate and multivariate Cox regression analyses revealed that HMGBI expression is an independent prognostic factor for patients with LSCC.The results of this study demonstrate that HMGB1 protein expression is significantly increased in LSCC tissues, and HMGB1 protein overexpression is associated with a poorer prognosis in patients with LSCC. These results suggest that HMGB1 may play a critical role in the initiation and progression of LSCC, implicating HMGB1 may become a valuable marker for the prediction of prognosis in patients with LSCC.

Abstract:Objective To detect expression of human leukocyte antigen protein G(HLA-G)in cervical cancer and explore its significance in the development of cervical cancer.Methods The SP immunohistochemical method was used to detect the expression of HLA-G in 54 cervical cancer tissues and 16 normal cervix tissues and its relationship with clinical pathological characteristics of cervical cancer was analyzed.Results The positive expression rate of HLA-G was 66.7%(36/54) in cervical cancer tissues,while no positive expression(0/16)was found in normal cervix tissues.The positive expression rate of HLA-G was closely related with clinical stage and differentiation degree(P0.05),but was not related with age,histological classification,and lymph node metastasis(P0.05).Conclusion Expression of HLA-G in cervical tumors is significantly higher than that in normal cervix tissues,and HLA-G plays an important role in the development of cervical cancer.
