Abstract:Background: As a membrane G protein coupled receptors (GPCRs) family, gastrin/cholecystokinin-2 receptor (CCK2R) plays a key role in the initiation and development of gastric cancer. Objectives: Targeting CCK2R by immunotherapeutics such as single-chain variable fragments (scFvs) may provide an effective treatment modality against gastric cancer. Thus, the main objective of this study was to isolate scFvs specific to CCK2R. Methods: To isolate scFvs specific to the CCK2R, we capitalized on a semi-synthetic diverse phage antibody library (PAL) and a solution-phase biopanning process. The library was panned against a biotinylated peptide of the second extracellular loop (ECL2) of CCK2R. After four rounds of biopanning, the selected soluble scFv clones were screened by enzyme-linked immunosorbent assay (ELISA) and examined for specific binding to the peptide. The selected scFvs were purified using immobilized metal affinity chromatography (IMAC). The binding affinity and specificity of the scFvs were examined by the surface plasmon resonance (SPR), immunoblotting and flow cytometry assays and molecular docking using ZDOCK v3.0.2. Results: Ten different scFvs were isolated, which displayed binding affinity ranging from 0.68 to 8.0 (nM). Immunoblotting and molecular docking analysis revealed that eight scFvs were able to detect the denatured form of CCK2R protein. Of the isolated scFvs, two scFvs showed high-binding affinity to the human gastric adenocarcinoma AGS cells. Conclusions: Based on our findings, a couple of the selected scFvs showed markedly high-binding affinity to immobilized CCK2R peptide and CCK2R-overexpressing AGS cells. Therefore, these scFvs are proposed to serve as targeting and/or treatment agents in the diagnosis and immunotherapy of CCK2R-positive tumors. Graphical abstract ᅟ.

Abstract:Gallbladder cancer (GBC) is a very aggressive malignant neoplasm of the biliary tract with a poor prognosis. There are no specific therapies for the treatment of GBC or early diagnosis tools; for this reason, the development of strategies and technologies that facilitate or allow an early diagnosis of GBC continues to be decisive. Phage display is a robust technique used for the production of monoclonal antibodies (mAbs) involving (1) the generation of gene libraries, (2) the screening and selection of isoforms related to an immobilized antigen, and (3) the in vitro maturation of the affinity of the antibody for the antigen. This research aimed to construct a human immune library from PBMCs of GBC patients and the isolation of scFv-phage clones with specificity against the larger extracellular loop belonging to claudin 18.2, which is an important biomarker overexpressed in GBC as well as gastric cancer. The immune-library-denominated GALLBLA1 was constructed from seven GBC patients and has a diversity of 6.12 × 1010pfu mL-1. After three rounds of panning, we were able to identify clones with specificity against claudin 18.2. GALLBLA1 can contribute to the selection, isolation, and recombinant production of new human mAbs candidates for the treatment of gastrointestinal cancers.
[Production of phage-displayed single chain variable fragments of monoclonal antibody MGb1].

Abstract:OBJECTIVE:To lay a foundation for obtaining a tumor-targeting vehicle for in vivo study on diagnosis and treatment of gastric carcinoma by generating single chain variable fragments (ScFv) of monoclonal antibody MGb1 directed against the cancer. METHODS:mRNA was isolated from MGb1-producing mouse hybridoma cell line, and the variable regions of heavy and light chain cDNAs were amplified separately and assembled into ScFv DNAs with a specially constructed linker DNA by PCR. The ScFv DNAs were ligated into the phagemid vector pCANTAB5E and the ligated sample was transformed into competent E. coli TG1. The transformed cells were infected with M13KO7 helper phage to yield recombinant phage, which display ScFv fragments as a fusion with gene 3 protein on the tips of the phage M13. After two rounds of panning with gastric carcinoma cell line KATO III highly expressing MGb1-binding antigen, the phage clones displayed ScFv fragments of the antibody were selected by enzyme-linked immunosorbent assay (ELISA) from the enriched phages. The affinity of the positive phage clones was detected by competition ELISA. RESULTS:The VH, VL and ScFv DNAs were about 340 bp, 320 bp and 750 bp respectively. 17 phage clones displayed ScFv of MGb1 were selected from 40 enriched phage clones. 4 out of the 17 phage clones could strongly compete with the original hybridoma antibody MGb1 for binding to the antigen expressed on KATOIII cells. CONCLUSION:The phage-displayed ScFv fragments of monoclonal antibody MGb1 are successfully produced by phage antibody technology, which may be useful to widen the range of application of the antibody.

Abstract:Objective To construct a human ovarian carcinoma cDNA phage expression library for screening tumor antigens.Methods The human ovarian sarcomatiod carcinoma cell line BUPH∶OVSC-1 was cultured and total RNA was isolated.In the presence of switching mechanism at 5' end of RNA transcript oligonucleotide,the single-strand and double-strand of cDNA were synthesized through reverse transcription and long distance PCR.After removed the fragments smaller than 500 bp,the remaining cDNAs were combined with the dephosphorylated λTriplEx2 phage vector.The ligated cDNAs were packaged into phage particles in vitro with Gigapack III gold packaging extract.A small portion of the packaged phage was used to infected E.coli XL1-Blue and the primary library was titrated and the percentage of recombinant clones was determined by blue white test.Sfi I restriction endonucleases was used to identify the size of inserted cDNA.The primary library was amplified for better preserving.Results Agarose gel electrophoresis of PCR product showed several characteristic bands corresponds to advantage transcript in the background of 400 bp ~7 kb smear,which according to mammals cell feature.The primary library consisted of 5.0 ×106 pfu/ml and recombinant rate was 99.0%.The length range of the inserts was between 500 bp ~ 4 kb with the average of 1.79 kb.The amplified library titration was 2.7×109 pfu/ml.Conclusions A high quality human ovarian carcinoma cDNA phage expression library derived from ovarian sarcomatiod carcinoma cell line is constructed,which may be used to screen the ovarian tumor-associated antigens.

Abstract:Accurate diagnostic techniques and effective therapeutic methods are required to treat H. pylori. The application of chicken single-chain variable fragment (scFv) antibodies may diagnose and treat H. pylori. This study used the phage display technique to construct a chicken-derived immune scFv antibody library against H. pylori. Total RNA was extracted from the spleens of five immunized chickens and reverse transcribed into cDNA. A fragment of scFv was produced by overlap extension PCR and cloned into a pHEN2 phagemid vector. After the package with the M13KO7 helper phage, the recombinant HpaA protein was used as a target antigen to validate the screening ability of our antibody library by bio-panning. The dilution counting results showed that the size of the primary antibody library was estimated to be 1 × 109 cfu/mL. PCR analysis of 47 clones from the library revealed that about 100% of the clones were positive with scFv fragments, and there were no identical sequences, indicating the good diversity of the antibody library. After three rounds of bio-panning, high-affinity antibodies against recombinant HpaA protein were successfully obtained. The selected antibody specifically recognized HpaA protein in nine different H. pylori strains, confirming the screening ability of our library. The chicken immune scFv antibody library against H. pylori was successfully constructed, and the antibody library's screening ability was validated by selecting specific scFv antibodies against recombinant HpaA and clinical strains. It provided a simple and rapid method to obtain antibodies against H. pylori for diagnosis or treatment.
[Mimic Epitope Recognized by Monoclonal Antibody MG7 Against Gastric Cancer Through Screening Phage Displayed Random Peptide Library].

Abstract:OBJECTIVE:To get mimic peptide epitopes that could be recognized by a monoclonal antibody against gastric cancer named MG7 from random peptide library displayed by phage, and to provide useful information for further study of the interaction between antigen and antibody.METHODS:Through affinity enrichment and immunoscreening of two phage-displayed non-apeptide libraries constructed in pVIII with MG7 MAb separately, several positive phages were obtained. Fluorescence labeling and dot blot were carried out for further identification of their binding activities. Then, some of the positive phages were sequenced and their corresponding peptide sequence was deduced according to their DNA sequence. Finally, HLA binding prediction software was applied for HLA binding analysis.RESULTS:Based on several rounds of screening and binding activity detection, we got twelve and thirty positive phage clones respectively from two libraries. Through DNA sequencing, peptide deducing and sequence aligning analysis of the positive phages, some preserved epitope information such as PLX(0 - 2)S, SAVR, XRMX and YARN were obtained. The prediction using HLA binding analysis software showed that most of the sequenced peptide had the potentiality to bind with HLA molecules.CONCLUSION:PLX(0 - 2)S, SAVR, XRMX and YARN may be some of the motifs which could be recognized by monoclonal antibody MG7.
[Construction, Expression and Activity of a Single Chain Antibody to Gastric Cancer Cells].

Abstract:OBJECTIVE:To construct single chain antibody from McAb 3H11, which has been successfully used in clinical radioimmunoimage, for its widel application in biological treatment of human cancer.METHOD:Reversetranscription and polymerize chain reaction (RT-PCR) were used to clone the V region genes of McAb 3H11 (VH, VL). The cloned VH and VL were sequenced and recombined through a (Gly4Ser) 3 linker. The constructed single chain antibody gene was joined with glutathione-S-transferase gene (gst) and its fusion proteins (GS3H11) were expressed in E. coli. The activity of GS3H11 was characterized with competition experiments after its denature and renature treatment.RESULTS:The recombinant GS3H11 was highly produced in inclusion bodies and the activity was recovered after being renatured. The competition experiments showed that GS3H11 could competitively inhibit the McAb 3H11 binding to gastric tumor cells MGC803. The inhibition was complete when the molecular ratio of GS3H11 to McAb 3H11 reached to 30:1. However, the GS3H11 did not interfere in binding of McAb PD4 and 3G9 to MGC803.CONCLUSION:The specific binding activity of McAb 3H11 is retained in single chain antibody which is potentially useful for therapeutic application.

Abstract:BACKGROUND: Peptide probes can be applied for biomarker targeting to improve the diagnostic accuracy. Cluster of differentiation 44 (CD44) is up-regulated in gastric cancer (GC). Among all the variants of CD44, CD44v6 is reported the most promising biomarker for GC. The purpose of this study was generating and identification a peptide ligand specific to CD44v6.METHODS: A 12-mer phage peptide library was screened on CD44v overexpressed HEK-293 cells with an improved subtractive method. Five candidate sequences emerged. Candidate phages were selected using enzyme-linked immunosorbent assay and competitive inhibition assays. Then the sequence (designated ELT) was chosen for further study. Its binding affinity and specificity were verified on recombinant protein, GC cells, GC tissues and xenograft models based on BALB/c-nu/nu mice using dissociation constant calculation, immunofluorescence, immunohistochemistry and in vivo imaging separately.RESULTS: The dissociation constant of ELT with recombinant protein was 611.2 nM. ELT stained CD44v overexpressed HEK-293 but not the cell expressing wild-type CD44s. On GC cell lines, ELT co-stained with anti-CD44v6 antibody. ELT binding on tumor tissues significantly increased compared with that of paracancer tissues, also showed a linear positive correlation with CD44v6 expression. ELT specifically accumulated in tumor and eliminated in short time in vivo.CONCLUSIONS: ELT can target GC in vitro and in vivo via CD44v6, indicating its potential to serve as a probe for GC targeting diagnosis and therapy.

[Screening of Breast Cancer Antigen from a Phage Display Random Peptide Library].

Abstract:AIM:to isolate novel breast cancer antigen for developing cancer marker and vaccine, a phage display peptide library was screened with serum antibody from breast cancer patients.METHODS:a phage display peptide library was screened with serum IgG from six breast cancer patients, and then identified the positive clones by ELISA. According to the sequences of the positive clones, the synthesized oligonucleotides of one small peptide (P15) was cloned into PGEX-4T-1. Finally the fusion protein (GST-P15), which was purified from E.coli. BL21, was checked its binding activity with two kinds of serum antibodies (20 breast cancer patients and 20 normal people) by ELISA.RESULTS:38 positive clones which specifically bound with serum IgG from breast cancer patients were screened. Some of these clones had the same DNA sequence. A small peptide (P15) and purified its fusion protein (GST-P15) was obtained successfully. ELISA assay showed GST-P15 had higher affinity with the serum antibody from breast cancer patients than with the normal serum and there was a significant difference (P<0.01).CONCLUSION:the peptide P15 can be recognized specifically by serum antibody from the breast cancer patients and they will be helpful for isolate new cancer antigen from breast cancer.
Construction and Preliminary Screening of Anti Endotoxin Phage Antibody

Abstract:Objective: To prepare a murine anti endotoxin single chain phage antibody. Methods: The total RNA was extracted from the murine splenic cells and the mRNA was reverse transcribed into cDNA. Then the designed primers were used to amplify the variable region genes of the heavy and light chain (V H,V L) with polymerase chain reaction. The linker was used to assemble the V H and V L into ScFv. The NotⅠ and SfiⅠ restriction enzymes were used to digest the ScFv in order to ligate into the pCANTAB5E phagemid vector digested with the same restriction enzymes. The ligated vector was introduced into competent E.coli TG1 cells to construct a single chain phage antibody library. After rescued with M13KO7 helper phage, phagemid containing bacterial colonies displayed ScFv fragments in the supernatant and could be selected with endotoxin. Then enriched positive clonies were reinfected into TG1 cells. Finally, 190 colonies were randomly selected to detect the anti endotoxin antibody with ELISA. Results: The titer of anti endotoxin in murine sera was 1∶12 800. The concentration of total RNA was 12.3813 μg/ml. V H was 340 bp while V L 320 bp, ScFv 800 bp. 1.9×10 7 colonies were grown after transformed into TG1. 3×10 4 colonies were gotten after panning once. Two positive colonies were confirmed with ELISA among 190 colonies. Conclusion: A 1.9×10 7 murine anti endotoxin single chain phage antibody library was successfully constructed. Two anti endotoxin antibodies were successfully screened from the library. [

Abstract:A non-immunized human single chain variable fragment (scFv) library containing 2.5 × 10 7 individual clones was constructed from antibody variable region genes of 200 non-immunized donors. ScFv gene repertories were generated by randomly combining rearranged variable regions of heavy chain (VH) and natural occurring light chain (VL) using overlapping extension PCR (OE-PCR). Five recombinant protein antigens from different species were successfully used to select specific binders. Phage ELISA showed that the recombinant phage particle could specifically bind to non-structural protein 1 of Avian influenza virus. This method can therefore efficiently generate a phage antibody library.
Screening and Identification of Vascular-Endothelial-cell-specific Binding Peptide in Gastric Cancer

Abstract:Antiangiogenesis therapy has become a hot field in cancer research. Blood vessels of tumor carry specific markers that are usually related to angiogenesis. Study of these heterogeneous molecules in different tumor vessels may be beneficial for promoting antiangiogenic therapy. In this study, we established an in vitro co-culture model of human umbilical vein endothelial cells (HUVECs) and gastric adenocarcinoma cell line SGC7901, screened the peptides binding specifically to the HUVECs co-cultured with gastric cancer cells (Co-HUVECs) using phage display peptides library, and studied the affinity of these peptides to gastric cancer vascular endothelial cells. After four rounds of panning, there was an obvious enrichment for the phages specifically binding to the Co-HUVECs, and the output/input ratio of Co-HUVECs increased about 590-fold (from 0.95×10−7 to 561.25×10−7). Five phage clones (M6, M3, M9, IN12, IN11), which could strongly bind to Co-HUVECs instead of wild-type HUVECs, were characterized by ELISA. In vitro cellular binding assay showed that phage IN11 preferably bound to Co-HUVECs rather than control HUVECs, and the number of the phage IN11 recovered from Co-HUVECs was 5.7- and 16.9-folds, respectively, as much as those from control HUVECs and GES cells. Immunocytochemical and immunohistochemical staining confirmed that phage IN11 could specifically bind to Co-HUVECs as well as vascular endothelial cells in gastric cancer tissue sections. Competitive and inhibitory assay revealed the synthetic peptide GEBP11 (CTKNSYLMC) displayed on phage IN11 could competitively inhibit binding of the phage IN11 to Co-HUVECs. Immunofluorescence microscopy showed that the fluorescence-labeled peptide GEBP11 was located at the membrane and perinuclear cytoplasm of Co-HUVECs. Meanwhile, GEBP11 was found to be able to target the gastric cancer vascular endothelial cells. Therefore, GEBP11 may be a potential candidate for targeted drug delivery in antivascular therapy and diagnosis of gastric cancer.
[Construction and Expression of Prokaryotic Vector of Single Chain Antibody Derived from a New Clone of Anti-Cd14 Antibody ZCH-7-2F9].

Abstract:OBJECTIVE:To construct a prokaryotic vector of ZCH-7-2F9 single chain antibody (ScFv2F9) and to obtain the ScFv2F9 protein with biological activity for further studies.METHODS:Primers were synthesized according to the gene sequence of ScFv2F9, four tandem glycin and one serine (G4S) 3linker and multiple cloning site(MCS) of pIVEX2.3-MCS vector, which included NdeI and SmaI enzyme cleaving sites. ScFv2F9 gene was amplified through splicing by overlap extension (SOE) using the high fidelity Taq polymerase. Then the gene was cloned to pGEM-T easy and pIVEX2.3-MCS vectors. Positive recombinants (pIVEX2.3-MCS/ScFv2F9) were identified through enzyme cleaving and sequenced before expression. The recombinant plasmids were transfected into E.coli BL21star(DE3)plysS for expression. After purification with Ni+resin and renaturation in vitro, the relative molecular mass (Mr) and the binding activity of the interesting protein were determined by SDS-PAGE and flow cytometry (FCM), respectively.RESULT:The cloned ScFv2F9 gene was identified to be functional by sequencing and expressing. The interesting protein was detected in inclusion body with a Mr of 31 000. The blocking test showed that the positive cell percentage, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 11.73%, 11.96% and 31.57%, respectively after once blockage by scFv2F9 protein, and 26.44 %, 21.75 % and 42.11 % after blockage twice.CONCLUSION:The ScFv2F9 against human CD14 antigen has been successfully expressed in prokaryotic cells with partial biological activity, which lays the foundation for further studies on its immunotoxin and other kinds of engineered antibodies.

Abstract:Objectives: Breast cancer is a popular fatal malignant tumor for women with high of rates incidence and mortality. Development of the new approaches for breast cancer targeted diagnosis and chemotherapy is emergently needed by the current clinical practice, the important first step is finding a breast cancer specifically binding molecule or fragment as early clinical indicators. Results: By a phage-displayed peptide library, a 12-mer peptide, CSB1 was screened out using MCF-7 cells as the target. The consequently results under immunofluorescence and laser scanning confocal microscope (LSCM) indicated that CSB1 bound MCF-7 cells and breast cancer tissues specifically and sensitively with high affinity. Bioinformatics analysis suggested that the peptide CSB1 targets the 5-Lipoxygenase-Activating Protein (FLAP), which has been implicated in breast cancer progression and prognosis. Conclusions: The peptide, CSB1 is of the potential as a candidate to be used for developing the new approaches of molecular imaging detection and targeting chemotherapy of breast cancer in the future.

Abstract:AIM: To gain tumor endothelium associated antigen genes from human liver cancer vascular endothelial cells (HLCVECs) cDNA expression library, so as to find some new possible targets for the diagnosis and therapy of liver tumor.METHODS: HLCVECs were isolated and purified from a fresh hepatocellular carcinoma tissue sample, and were cultured and proliferated in vitro. A cDNA expression library was constructed with the mRNA extracted from HLCVECs. Anti-sera were prepared from immunized BALB/c mice through subcutaneous injection with high dose of fixed HLCVECs, and were then tested for their specificity against HLCVECs and angiogenic effects in vitro, such as inhibiting proliferation and inducing apoptosis of tumor endothelial cells, using immunocytochemistry, immunofluorescence, cell cycle analysis and MTT assays, etc. The identified xenogeneic sera from immunized mice were employed to screen the library of HLCVECs by modified serological analyses of recombinant cDNA expression libraries (SEREX). The positive clones were sequenced and analyzed by bio-informatics.RESULTS: The primary cDNA library consisted of 2x10(6) recombinants. Thirty-six positive clones were obtained from 6x10(5) independent clones by immunoscreening. Bio-informatics analysis of cDNA sequences indicated that 36 positive clones represented 18 different genes. Among them, 3 were new genes previously unreported, 2 of which were hypothetical genes. The other 15 were already known ones. Series analysis of gene expression (SAGE) database showed that ERP70, GRP58, GAPDH, SSB, S100A6, BMP-6, DVS27, HSP70 and NAC alpha in these genes were associated with endothelium and angiogenesis, but their effects on HLCVECs were still unclear. GAPDH, S100A6, BMP-6 and hsp70 were identified by SEREX in other tumor cDNA expression libraries.CONCLUSION: By screening of HLCVECs cDNA expression library using sera from immunized mice with HLCVECs, the functional genes associated with tumor endothelium or angiogenesis were identified. The modified SEREX, xenogeneic functional serum screening, was demonstrated to be effective for isolation and identification of antigen genes of tumor endothelium, and also for other tumor cell antigen genes. These antigen genes obtained in this study could be a valuable resource for basic and clinical studies of tumor angiogenesis, thus facilitating the development of anti-angiogenesis targeting therapy of tumors.

Abstract:Tumor is a serious threat to human health. At present, surgical resection, chemoradiotherapy, targeted therapy and immunotherapy are the main therapeutic strategies. Monoclonal antibody has gradually become an indispensable drug type in the clinical treatment of cancer due to its high efficiency and low toxicity. Phage antibody library technology (PALT) is a novel monoclonal antibody preparation technique. The recombinant immunoglobulin variable region of heavy chain (VH)/variable region of light chain (VL) gene is integrated into the phage vector, and the antibody is expressed on the phage surface in the form of fusion protein to obtain a diverse antibody library. Through the process of adsorption-elution-amplification, the antibody library can be screened to obtain the antibody molecule with specific binding antigen as well as its gene sequence. PALT has the advantages of short antibody production cycle, strong plasticity of antibody structure, large antibody yield, high diversity and direct production of humanized antibodies. It has been used in screening tumor markers and preparation of antibody drugs for breast cancer, gastric cancer, lung cancer and liver cancer. This article reviews the recent progress and the application of PALT in tumor therapy.

Profiling Tumor-Associated Autoantibodies For The Detection Of Colon Cancer

Abstract:Purpose: The purpose of the present study was to screen the autoantibody signature of colon cancers to develop serum markers for colon cancer detection.Experimental Design: A phage cDNA expression library of colon cancer was built. The library was sequentially screened by a pool of 10 colon cancer sera, goat antihuman IgG, and a pool of two healthy sera to identify phage-expressed antigens recognized by tumor-associated antibodies. The clones picked out by these screening were subjected to a training set with 24 colon cancer sera and 24 healthy sera. The antigen combination, which got the most satisfactory classification, was tested by an independent set of 24 colon cancer sera with equal number of sera from normal donors. The carcinoembryonic antigen (CEA) level of these sera was detected for the additional classification analysis with or without the antigen combination.Results: A cDNA expression library consisting of 2 x 106 primary clones was prepared. After three turns of screening, 24 antigens recognized by tumor-associated antibodies were picked out for serum marker identification. The training set showed that a six-marker combination got the most satisfactory classification in a logistic regression model; leave-one-out validation achieved 91.7% sensitivity and 91.7% specificity. In a testing set with this marker panel, we correctly predicted 85% of the samples. Although according to CEA level alone, we correctly predicted 75% of the samples with 42% of cancer patients misclassified. When CEA was combined with the six markers, the sensitivity and specificity increased to 91.7% and 95.8%, respectively. The six antigen sequences in the phage display system are relatively short peptides. Only two of them showed homology to known protein sequences.Conclusions: Autoantibodies against phage-expressed antigens derived from colon cancer tissues could be used as serum markers for the detection of colon cancer.

Abstract:The murine phage antibody library was amplified and then panned by human chorionic gonadotropin (hCG) for four rounds.The last round enriched phage clones were used to reinfect E coli TG1 and 15 separate colonies were assayed by ELISA.The recombinant phages which were rescued from colony 12# showed the highest affinity to immobilized hCG.The Scfv gene was inserted into the reconstructed pET\|21a(+) and expressed in E coli BL21(DE3).The dot blot hybridization verified that the ScFv was secreted to the medium.

Abstract:Tonsillitis is the inflammation of the tonsils due to infection, many patients ultimately have to undergo tonsillectomy. In order to improve the accuracy of diagnosis and even create a new treatment for tonsillitis, we constructed a prokaryotic expression single-chain antibody fragment library against Streptococcus pneumoniae with immunoglobulin heavy chain variable region (VH), κ light chain (Vκ), and λ light chain (Vλ) genes by using human tonsil tissue. Plasmid DNA sequencing showed that single-chain antibodies were complete and constructed correctly. The binding activity of recombinant clones was detected by enzyme-linked immunosorbent assay (ELISA), results showed that the binding activity and specificity of anti-S. pneumoniae single-chain fragment variable (scfv) is proved to be successful. The single-chain antibody may be an attractive strategy for tonsillitis etiologic diagnosis and therapy.

Abstract:Kita-kyushu lung cancer antigen 1 (KK-LC-1) is a kind of cancer-testis antigen with anti-tumor potential for clinical application. As a class of small-molecule antigen conjugates, tumor-targeting peptides have broad application prospects in gastric cancer diagnosis, imaging, and biological treatment.Here, we screened specific cyclic nonapeptides from a phage display library. The targeting peptide with the best affinity was selected and further verified in the ex vivo tissue sections. Finally, the enrichment of targeting peptides in tumor tissues was observed in vivo, and the dynamic biodistribution process was also observed with micro-PET/CT imaging.Studies showed the specific cyclic nonapeptide had a high binding capacity for KK-LC-1 protein. It has a strong affinity and specificity for KK-LC-1-expressing positive tumor cells. Targeting peptides were significantly enriched at tumor sites in vivo, with very low normal tissue background.These findings demonstrated that the KK-LC-1 targeting peptide has high clinical potential.
