Construction of recombinant adenoviruses containing small interfering RNA targeting rat protein tyrosine phosphatase 1B

Abstract:Background: Protein tyrosine phosphatase 1B (PTP1B) regulates insulin signal transduction in down-regulation. A key link of restricting PTP1B function is lack of an effective tool. Objective: To construct recombinant adenovirus containing small interfering RNA (siRNA) targeting rat PTP1B mRNA (Ad-siRNA-PTP1B). Design, time and setting: The experiment was performed at the Central Laboratory, Changhai Hospital Affiliated to Second Military Medical University of Chinese PLA from May to December 2007. Materials: 293 cells were obtained from Shanghai Institute of Cells, Chinese Academy of Sciences. Eukaryotic expression plasmid psiRNA-PTP1B was synthetized at the Shanghai Jikai. AdEasy-1 bacterium, shuttle vector pAdTrack and bacillus coli DH5 α were kept in this laboratory. Recombinant adenovirus polymerase chain reaction (PCR) identified primer was designed and synthetized at the Shanghai Sangon Biological Engineering Technology & Services Co., Ltd. Methods: Plasmid vector containing rat PTP1B siRNA was obtained from the company. The siRNA cassette, containing the U6 promoter and the hairpin siRNA sequence, was excised and ligated into pAdTrack. The plasmid of pAdTrack-siRNA-PTP1B was linearized with Pme I, and the fragment was reclaimed and transformed into E. coli BJ5183 cells containing adenovirus backbone plasmid pAdEasy-1, then pAdsiRNA-PTP1B was generated by homologous recombination. Plasmid pAdsiRNA-PTP1B was identified by Pac I digestion, then transfected into 293 cells to package the adenovirus, followed by identification of the recombinant adenovirus by means of observation of green fluorescence protein expression and PCR. Additional vector amplification was also performed in the 293 cell line. High titer viral vectors were purified by centrifugation and CsCI banding. Main outcome measures: Green fluorescent Protein (GFP) expression was observed by fluorescent microscope. Recombinant adenoviral shuttle vector and recombinant adenovirus construction. Results: Recombinant adenoviral vector pAd-siRNA-PTP1B was constructed successfully, which was confirmed by Pac I digestion. GFP was expressed in 293 cells infected with recombinant adenovirus Ad-siRNA-PTP1B. After being packaged in 293 cells, and purified by CsCI banding, the recombinant adenovirus Ad-siRNA-PTP1B, whose liter was as high as 3.6 × 10 9 efu/ml, were produced. Conclusion: AdEasy-1 system is an efficient way to construct recombinant adenovirus containing siRNA targeting rat PTP1B mRNA.
