Effects of Fluvastatin on Monocrotaline-Induced Pulmonary Vascular Remodeling and Right Ventricular Hypertrophy in Rats

Abstract:Objective To investigate the therapeutic effects of fluvastatin on monocrotaline-induced pulmonary vascular remodeling and right ventricular hypertrophy.Methods 66 rats were randomly distributed into 3 groups.One group(M+F,n=24) received a single subcutaneous injection of MCT(60 mg/kg) for 2 weeks and then were given fluvastatin(1 mg/kg) once daily by gavage from 3rd week to 6th week.Another group(MCT,n=24) received a single subcutaneous injection of MCT(60 mg/kg) for 2 weeks and then were given an equivalent volume of saline gavage from the 3rd week to 6th week.The third group(Saline,n=18) received only equivalent volume of 0.9% saline injection or gavage for 42 d.Morphometric changes,pulmonary arterial pressure(PAP),percent wall thickness(PWT),right ventricular hypertrophy index(RVHI) were detected at various times during the 6-week observation.Results Pulmonary vascular remodeling and right ventricular hypertrophy were resulted 2 weeks after MCT administration.PWT,mPAP and RVHI of rats in MCT group worsened further after withdrawal of MCT;however,mPAP(23.3±3.2) mmHg,PWT(42.3±2.7)% and RVHI(0.32±0.02) was significantly lower than that of MCT group.Furthermore,no difference was noted between M+F group and Saline group in RVHI value.Conclusion Fluvastatin is effective in attenuating MCT-induced pulmonary vascular remodeling and ameliorating right ventricular hypertrophy in rats.
