Induced Mutation for Biocontrol Microorganisms Against Mango Stem End Rot and Screening of Pesticides for Use with The Microorganisms

Abstract:Sixteen mutant strains with higher inhibitory effects on the growth of Botryodiplodia theobromae,the casual pathogen of mango stem-end rot,were obtained by UV induced mutagenesis.The diameters of the inhibition circles were increased by 47.6%,42.9%,38.1%,36.8%,36.8% and 29.4% by Bacillus strains,4a-3-4,4a-3-1,4a-3-2,5a-2-1,and yeast strains,E3-3-1 and B2-2-2,respectively,compared with their respective original strains,showing a significant mutagenic effect.Fungicides were screened to find those compatible with the use of the mutant strains and were identified that carbendazim at the concentration of 2 μg/mL and thiophanate-methyl at 1 μg/mL were the best.Inhibitory results showed that most of the mutant strains had synergistic inhibitory effects when used with the two fungicides.Compared with the use of thiophanate-methyl alone,use of thiophanate-methyl plus strains of Bacillus 4a-3-3,5a-2-1 and yeast E3-3-1 increased the rates of protected fruits to 75.0%,74.5% and 73.5%,and reduced the decayed fruits by 8.0%,7.5% and 6.5%,respectively.Similarly,use of carbendazim plus Bacillus strains 4a-3-4,5a-2-1 increased rates of good fruits to 73.4% and 72.7%,which were about 2.3% and 1.6% higher than use of carbendazim alone.
